ERNIE'S EXIT, Blood Typing Activity A small poo! of blood and a weapon were found near a garbage dumpster. After examining the area, the CSI on the scene discovered a body in the garbage dumpster and identified him as Earnest “One-eyed!” Earl, Earl had a wound to his chest that will be analyzed by the medical examiner The CSI tested blood samples from the blood pool and the weapon at the crime scene. It was determined that it was human blood, but he needs to know the blood type to help identify if it was from the victim or the person who murdered him. He has identified three suspects that either knew the victim or were seen in the area before the body was discovered. He would like to question them further while he waits for DNA test results, Lab Results Complete the chart using the class notes, ‘Clumping = + (Positive) No Clumping = - (Negative) A + - 8 - + AB + + ° - Z Rh Serum = Clumping = Rh Blood Follow your teacher's directions to complete the lab. Remember to be careful to prevent cross-contamination of the blood samples! Record the results (+ or -) from each group in the tray below. Suspect #2: “Slim” Jim Snot Suspect ff: Bobby “Baby” Brooks Blood Type:__ Blood Type: ___ Suspect #3: Barbie “Doll” Jones Blood Type:__ Rh Rh ee {Vena Vaan HOA) FO Victim Earnest “One-Eyed” Earl Crime Scene Sample Blood Type: Blood Type: A B Oo Weapon Sample Blood Type:. Procedure 1. You will be assigned to groups and sit at your table number. 2. All materials will be available at every table. Please do not cross contaminate any of the materials. In order to get accurate results, we can not allow for this. 3. Make sure you have the following at your station before starting: a. Anti-A Serum b. Anti-B Serum Rh Serum d, 2 well trays (1 Per subgroup) e. Eye Droppers To start off, fill three wells half way with the blood samples from the suspect. After filing three wells, take Anti-A Serum and place § drops into one of the wells. Take a toothpick and stir for 5 seconds. Repeat stops 5 and 6 using Anti-B Serum for the second well and Rh Serum for the third, DO NOT USE THE SAME TOOTHPICK TO STIR YOUR SAMPLES? 8, Once you have mixed and stirred all samples, record your results on page 1. If the sample began to clump, place a + in the box. If no clumping occurred in the well, place a-. 9. Ifa sample clumps with the Anti-A serum, the suspect or object contains the A allele and has Type A blood. If it clumps with the Anti-B serum, the suspect or object contains the B allele and has the Type B blood. If both Anti-A and Anti-B serum clump, the suspect or object has Type AB blood. If neither clump, the suspect or object contains Type O blood. 10. If the Rh Serum clumps, the suspect or object has positive blood. For example, A+ blood or Bt blood. If it does not clump, itis negative blood. For example, A- or B- blood. 11. You will be rotating through 6 total stations, There may be more than one suspect in this case, so no conclusions can be drawn until all evidence is collected. Noe Concluding Questions 1, Based on your results, which suspect could be linked to the crime? Explain how the results from the blood typing proves your accusation. 2. What is the difference between incomplete and complete dominance? Codominance- when both alleles are expressed at the same time Example: ABO system of blood types This is an example that uses a 3 allele system, A persons blood type is based on the absence or presence of certain proteins in his or her red blood cells. Ais codominant with B - neither covers the other both are expressed independently, Ais dominant over O - Only A is expressed if A and O are present. B is dominant over O - Only B is expressed if B and O are present. Possible Genotypes ‘TyeB | Type ab” AA AO BB BO AB 43. Ifyou have type A blood, what are your possible genotypes? 4, Ifyou have type B blood, what could your genotypes be? 5. If you have type AB blood, what must your genotype be? 6. If you have type 0 blood, what must your genotype be? 7. Could two individuals with type A blood ever produce any offspring with type 0 blood? Explain your answer completely and USE A PUNNETT SQUARE TO PROVE YOUR REASONING. 8. Could two individuals with type blood ever produce any offspring with type A blood? Explain your answer completely and USE A PUNNETT SQUARE TO PROVE YOUR REASONING. 9. Could a type O individual and a type AB individual ever produce any offspring with type A blood? Explain your answer completely and USE A PUNNETT SQUARE TO PROVE YOUR REASONING. 10. Awealthy elderly couple die together in an accident, Soon a man shows up to claim their fortune, saying that he is their only son who ran away from home as a boy, Other relatives dispute his claim. Hospital records show that the deceased couple were type AB and O respectively. The claimant to the fortune was type O. Could he be their son? Explain your answer completely and USE A PUNNETT SQUARE TO PROVE YOUR REASONING. 11. Suppose two newborn babies were accidentally mixed up in the hospital. In an effort to determine to which parents ‘each baby belonged, the blood types of the babies and parents were identified. From the following test results, identity which baby belongs to which parents and their genotypes. Individual: Blood Type Genotype Parents Name Baby #1 oO Baby #2 A Mrs. Brown, B Mr. Brown, AB Mrs. Smith B Mr, Smith a
