ABSTRACT

An experiment was conducted to study sub acute toxicopathological


studies of Ipomoea carnea in Wistar rats for twenty eight days. Thirty six (male and
female) rats were divided in to three groups comprising of 12 rats (6male and 6
female). Group I rat served as control, Group II rats were given Ipomoea carnea
leaves powder @4% and group III rats were given 8% Ipomoea carnea leaves
powder in feed of male as well as female rats. The rats of treated group were dosed
daily through feed.

During experimental period rats of group II and III showed symptoms


of dullness and depression. There was decreasing trend of mean values of body
weight in male as well as female rats at scheduled intervals of study period.

At both interval of study period in male as well as female rats of group


II and III mean value of haemoglobin, TEC were reduced significantly except this
reduction in mean total erythrocyte count in female rat at 14th day found to be non-
significant decline. The mean TEC values at 28th day in female rats of group II and III
were reduced significantly.

The blood sample when subjected to total leucocyte count (TLC) and
differential leucocyte count (DLC) revealed significantly incline in mean values of
total leucocyte count, and non-significantly incline in neutrophil, eosinophil and
decline in lymphocyte. However, the mean values of monocytes showed no change
in normal count in male as well as female rats of treatment groups at both the
interval of study period.

There was significant increase in the liver biomarker enzymes like


alanine transaminase (ALT), aspartate transaminase (AST) in all treatment groups at
both the interval except at 14th and 28th day mean value of ALT and AST were
increased non-significantly. The mean values of blood urea nitrogen at both the
interval of study period in male and female rats were increased significantly. The
mean values of serum creatinine in male and female rats at 14th day were
significantly risen. At 28th day the mean values of serum creatinine in male and
female rats were differ non-significantly. There was significant decline in mean
values of serum total protein in male and female rats of group II and III at both the
interval of study period. The mean values of serum calcium in male as well as female
rats decline non-significantly at 14th day of study. There was significantly reduction in
mean values serum of calcium in male and female of group II and III at 28th day. The
mean serum phosphorous in male and female of group II and III at both the interval
of study were incline significantly than the control group values.

Gross pathological observation did not revealed any noticeable gross


change in liver, kidneys, lungs, testes, brain, intestines and spleen changes except
congestion in any experimental group. There was significant and non-significant
increase in mean absolute weights of liver in male and female rats of group II & III at
end study. The absolute weight of kidneys in male and female rats of group II and III
were significant and non-significant increase. There was significant decrease in
mean absolute weights of testes in male rats in group III while the weights of testes
in group II were reduced numerically than control group weights.

The histopathogical studies of kidneys of experimental rats in group III


male and female revealed moderate congestion, cystic degeneration and cystic
tubular necrosis with mono nuclear cell infiltration and multifocal necrobiotic
degenerative changes. In male and female rats of group III showed multifocal
vacuolar degenerative changes and cytoplasmic rarefaction in liver. The sections of
testes from rats of group III showed mild focal vacuolation in seminiferous tubules
and occasional rinkling of basement membrane of testes. Sections of brain from
male rats of group III revealed mono nuclear cell infiltration and mild vacuolation.
Spleen in male and female rats of group III showed mild depopulation of
lymphocytes and congestion respectively. Intestines in male and female rats of
group III revealed exfoliation of epithelial lining of intestine, hyperplasia of goblet
cells and acute enteric changes.

The histopathological changes were more pronounced in rats of


Group III than other treatment groups. The rats of control group did not show any
appreciable histopathological changes.
