Explant is taken from the parent cell and cut into small pieces The explant is sprayed with sodium hypochlorite solution to prevent the growth of pathogen. The explant is sterilize and placed in a culture medium containing nutrients such as glucose, amino acids, mineral and growth hormone, auxin for several weeks. The culture medium is maintained at an optimum pH and temperature of 28 – 35 ‘c. The tissue cells divide by mitosis and form callus. The callus differentiate to somatic embryotic and form shoots and roots. The callus is simulated by shoot-stimulating hormone to form shoots and rooting is stimulated with root-stimulating hormone.
