Tissue Culture (Cloning of Plants)

 Plants can be cloned by tissue culture.


 Explant is taken from the parent cell and cut into
small pieces
 The explant is sprayed with sodium hypochlorite
solution to prevent the growth of pathogen.
 The explant is sterilize and placed in a culture
medium containing nutrients such as glucose,
amino acids, mineral and growth hormone, auxin
for several weeks.
 The culture medium is maintained at an optimum
pH and temperature of 28 – 35 ‘c.
 The tissue cells divide by mitosis and form callus.
 The callus differentiate to somatic embryotic and
form shoots and roots.
 The callus is simulated by shoot-stimulating
hormone to form shoots and rooting is stimulated
with root-stimulating hormone.
 Once the roots is formed, plantlets is transferred
into the soil
