MUSHROOM TYROSINASE ASSAY:

Based on Dr Saja Hamed’s 96 well plate assay (Summer 2007)

Reagents:
 50 mM potassium phosphate buffer pH 6.8
 2 mM L-DOPA
 Mushroom Tyrosinase (Sigma Cat# T3824)
 Inhibitor made to stock concentrations for dilution to desired Final concentration

Rx mix for a 96 well plate

Per well mix for a final volume of 0.150ml:
 0.05 ml 2 mM L-DOPA
 0.05 ml 200 U/ml Mushroom Tyrsosinase
 0.05 ml Inhibitor stock

Place the assay plate at 37° C for 20 mins.

Read the plate at 490 nm using Bio-Rad plate manager program.
Export results into Excel for analysis.
