Microbiology Department, University of Medicine and Pharmacy, 38 Gh. Marinescu street, 540139, Tirgu Mures, Romania Objectives: The aim of this work was to synthesize a new Pt** complex Nayo[NaAs,WeoOrgoPtz"*}:xXH20 (AS4WaoPt) of Nap 7[NaAsqWqa140]+xH20 cryptand (As,Wgo) and to assess the antibacterial activity. Both compounds are polyoxotungstate (POW), inorganic componds with a broad range of pharmacological properties (antiviral, antibacterial, antitumoral activities), even that their molecular mechanism of action Is poorly understood. Methods: The As;Wio cryptand was synthsized following methods described in the literature (Na,WO., NaAsO,. The complex was obtained by direct addition of cation salt solution, KPtCl, to As;W4o sodium salt solution at pH=4 keeping the temperature between 70-80°C. The ratio of the reagents was calculated according to the reaction molar ratio(2:1). After purification the complex was identified and characterized using UV, IR, TGA, conductometry, X-ray methods. The biological activity was studied in vitro on Gram positive (Staphylococcus aureus ATCC 29213) and Gram negative (Escherichia coli ATCC 25922) bacterial strains by successive dilution method, with identification of minimum inhibitory and bactericidal concentrations. Results: The As,W.oPt complex structure is a Leyrie assembly which consist of 4 identical trilacunar Keggin anion structures ([AsW.033]”) joined together by 4 WOs octahedra in a ring-like form with high symmetry. Oxygen atoms delimit the structure, The pt** cation is central coordinated inside the structure, in active positions formed in the lacunar places with high electron density. The results obtained by successive dilution method are the following (MIC,MBC=mg/ml): As,WaoPt - for Staph.aureus MIC=1.33, MBC=1.33; for E.coli MIC=2.66, MBC=10.65; AscWio - for Staph.aureus MIC=2.00, MBC=2.00; for E.coli MIC=4.00, MBC=32.00, Conclusions: A new complex was synthesized, NaelNaAs,WeoQrgoPt>’*]-xH,0. Both compounds exhibit antibacterial activity. The complex has shown a bigger antimicrobial activity than the cryptand. Staph. aureus have proved to be more sensitive than E. coli.
