Probes Calibration Procedures

NOTE: This information is provided as an aid in Technical Support for trained support
personnel. Do not attempt to service the system without the appropriate technical training
and the proper tools.
Applicability All ADVIA Centaur® CP systems
Estimated Time 30 minutes
Special Notes This document provides instructions for calibrating the sample
probe and reagent probe to calibration points in the tip drawers,
the tip eject area, the sample compartment, ancillary reagent area,
primary reagent area, rinsing station, incubation ring, and system
reference point.
Action Perform these procedures before operating the system for the first
time. Repeat these procedures any time you replace or remove an
assembly for servicing.

Introduction
This document provides instructions for calibrating the sample probe and reagent probe
to calibration points in the following areas:
• tip drawers
• tip eject area
• sample compartment
• ancillary reagent area
• primary reagent area
• rinsing station
• incubation ring
• system reference point
Perform these procedures before operating the system for the first time, as part of the
system installation (refer to the support bulletin Installation Guide, part number
086D0006-xx). Repeat these procedures any time an assembly containing calibration
coordinates or a probe is replaced or removed for servicing.

Using the Probes Calibration Software


Accessing the Software
1. Turn the ADVIA Centaur CP system off.
2. Turn the ADVIA Centaur CP system on.
3. To access the Probes Calibration Software, double-select the ACCPCoo shortcut icon
on the desktop.
The Sign In window displays.
4. At the Sign In window, enter your password.
5. Select OK.
6. At the Message window, select OK.
The Probes Calibration window displays.

Using the Probes Calibration window


The Probes Calibration window is the main window from which most actions are
performed. From this window, you can access all the calibration areas. The Probes
Calibration window contains graphic depictions of the calibration areas showing the
calibration points. These points are shown on the window as red dots.

The Probes Calibration window also contains six buttons:

• Pick Tip – Select this button to have the probe pick a sample tip.
• Racks – This feature is not currently used.
• Init – Select this button to home the system modules.
• Sample Probe Reference – Select this button to calibrate the sample probe to the
system reference point.
• Reagent Probe Reference – Select this button to calibrate the reagent probe to the
system reference point.
• Done – Select this button to exit the Probes Calibration Software.
Using the Keyboard
To observe the probes in relation to the calibration points in the system interior, take the
keyboard in hand and move to the system as needed.
Moving the Probes with the Cursor Keys
Use the cursor keys on the keyboard to move the probe to the correct coordinates:
Key Movement
Left arrow key moves the probe to the left (x-axis)
Right arrow key moves the probe to the right (x-axis)
Down arrow key moves the probe to the front (y-axis)
Up arrow key moves the probe to the back (y-axis)
Page Up key moves the probe up (z-axis)
Page Down key moves the probe down (z-axis)
Shift key moves the probe down the z-axis in five steps to 1/2, 3/4, 6/8, …
of the previously set z-coordinate.
Plus (+) key increases the step size
Minus (-) key decreases the step size

Using the Shift Key for the Z-Axis

CAUTION
Ensure that you always use the smallest step increment for making a final calibration.
Using a larger step increment could result in damage to the probe.

CAUTION
Do not attempt to re-calibrate a probe if the probe crashes (goes offline) during the
calibration procedure. This could result in damage to the probe. If the probe crashes,
select Cancel to exit the window. Move the probes to a safe position. Start the program
and continue from the point at which the software crashed. If the program does not
respond, restart the computer.

When calibrating a probe to a z-axis coordinate, use the Shift key to move the probe close
to the surface in five half-increment steps (to 0.5, 0.25, 0.125, 0.0625, and 0.0312).

Then use the smallest increment step to move the probe down until it touches the surface
of the calibration pin.
This is the safest method of moving the probe.

Using the Calibration Window


At the Probes Calibration window, select a calibration area (or one of the reference
buttons) to view or calibrate the coordinates for that calibration area. The Calibration (or
Adjust Position) window displays for that calibration area.

The probe moves to the current x- and y- coordinates. The z-coordinate is not
automatically applied, to avoid damaging the probe.
Increment Bar

CAUTION
Ensure that you use the lowest step increment for making a final calibration. Using a
larger step increment could result in damage to the probe. When making your final
adjustment to a calibration, always use the smallest step increment.

Use the increment bar to adjust the distance the probe moves per step increment. To
increase the step increment, move the increment bar up. To decrease the step increment,
move the increment bar down. In the lowest position, the step increment is minimal.

CAUTION
Ensure that you use the lowest step increment for making a final calibration. Using a
larger step increment could result in damage to the probe. When making your final
adjustment to a calibration, always use the smallest step increment.

X-, Y-, Z- Coordinates


The probes move along three axes: x, y, and z. Each axis references the probe’s direction
of motion.

The x-, y-, and z- axis coordinates reference the position of the probe tip along the three
axes. The three axis coordinates represent the actual position of the probe tip.

The number of steps translates to the distance according to the following table:

x left-right 10 1 mm
y front-back 8 1 mm
z up-down 20 1 mm

Previous Z

CAUTION
Do not select the Previous Z button unless you are familiar with the previous calibrated
position. Using the Previous Z feature without knowing its actual previous position could
result in damage to the probe.

The Previous Z button allows you to move the probe down to the last calibrated z-axis
coordinate.

Aligning the Probes to the Calibration Points


NOTE: When aligning a probe to a coordinate pin, ensure that the probe is touching the
center of the pin.
1. Using the Shift key, move the sample probe down to a point approximately 0.5 cm
(0.2 in) above the calibration pin.
2. Using the keyboard controls, adjust the x and y coordinates until the probe is center-
aligned to the calibration pin.
3. Using the smallest increment steps, carefully lower the probe until it touches the
center of the calibration pin.

WARNING
Ensure that you are clear of subassemblies that can move before selecting OK. Remove
yourself from the machine. The probe moves rapidly and can cause injury.
4. To save the coordinates, select OK.
The probe moves to the next position.

Calibration Points
When calibrating the probes to the calibration points, follow the numeric order indicated
below. Always calibrate the sample probe to the system reference point (calibration point
8) before calibrating points 1 – 10. Always calibrate the reagent probe to the system
reference point (calibration point 8) before calibrating points 11 – 19.

Calibrating the Sample Probe


Calibrating the Probe to the System Reference Point
The coordinates for all the calibration areas are related to the system reference point
(calibration point 8). The system reference point is located on the front right edge of the
sample compartment shutter guide.
Always calibrate the probes to the system reference point before calibrating the probes to
the other coordinates. If you change the reference point calibration coordinate, all other
calibration point coordinates will change.
1. At the Probes Calibration window, select Init to home all system modules.
2. Select the Sample Probe Reference button.
The Calibration window displays. The sample probe moves to the last calibrated
system reference point x- and y-coordinates.

CAUTION
Do not attempt to re-calibrate a probe if the probe crashes (goes offline) during the
calibration procedure. This could result in damage to the probe. If the probe crashes,
select Cancel to exit the window.

WARNING
Ensure that you are clear of subassemblies that can move before selecting OK. Remove
yourself from the machine. The probe moves rapidly and can cause injury.

3. Calibrate the sample probe to the system reference point (calibration point 8):
a. Using the keyboard controls, align the probe to the calibration pin.

CAUTION
Do not select the OK button unless the calibration is complete for that coordinate.
Selecting OK prior to completing the procedure overwrites the previously calibrated
coordinates and could damage the probe. If you need to stop the calibration before
completing the procedure, stand clear of the probes, and select Cancel.
b. Select OK.
The probe moves to the home position.

Calibrating the Probe to the Tip Drawers Coordinates


After calibrating the sample probe to the system reference point, calibrate the probe to
calibration points 1, 2, 3, and 4 in the tip drawers. The tip drawers area is located on the
left side of system interior.

CAUTION
Ensure that the probe is calibrated to the system reference point before proceeding. If the
probe is not properly calibrated to the reference point, damage to the probe could result.

1. At the Probes Calibration window, select the tip drawers area.


2. At the Warning window, select OK.
The Adjust Position window displays. The sample probe moves to calibration
point 1.

3. Calibrate the sample probe to calibration point 1 (left-back position):


a. Using the keyboard controls, align the probe to the calibration pin.
b. Select OK.
The sample probe moves to the next position.
4. Calibrate the sample probe to calibration point 2 (left-front position):
a. Using the keyboard controls, align the probe to the calibration pin (left-front
position).
b. Select OK.
The sample probe moves to the next position.
5. Calibrate the sample probe to calibration point 3 (right-back position):
a. Using the keyboard controls, align the probe to the calibration pin.
b. Select OK.
The sample probe moves to the next position.
6. Calibrate the sample probe to calibration point 4 (right-front position):
a. Using the keyboard controls, align the probe to the calibration pin.
b. Select OK.
The sample probe moves to the home position.

Calibrating the Probe to the Tip Eject Area Coordinates


After calibrating the sample probe to the tip drawers calibration points, calibrate the
probe to calibration point 5 (all three positions) in the tip eject area. The tip eject area is
located between the sample compartment and the metal housing of the barcode scanner.

CAUTION
Ensure that the probe is calibrated to the system reference point before proceeding. If the
probe is not properly calibrated to the reference point, damage to the probe could result.

1. At the Probes Calibration window, select the tip eject area.
The Adjust Position window displays. The sample probe moves to calibration
point 5.
2. Calibrate the sample probe to calibration point 5 (back eject position):
a. Using the keyboard controls, move the sample probe back (y-axis) until the
center of the probe housing is aligned with the calibration point 6 on the sample
tray (figure 6).
b. Align the x-axis so that the eject sleeve is 1 mm from the left side of the tip eject
wall on the left (figure 11).
c. Using the keyboard controls, align the probe so that the lower black housing of
the probe adapter is approximately 0.2 mm above the metal surface of the
barcode scanner housing (z-axis).

d. Select OK.
The probe moves to the next position.

WARNING

yourself from the machine. The probe moves rapidly and can cause injury.

3. Calibrate the sample probe to calibration point 5 (front eject position):


a. Using the keyboard controls, move the sample probe (y-axis) until the front of
the sample probe housing is approximately 25 mm from the front of the opening
of the tip eject area.
b. Align the x-axis so that the eject sleeve is 1 mm from the left side of the tip eject
wall on the left (figure 11).

c. Using the keyboard controls, align the probe so that the lower black housing of
the probe adapter is approximately 0.2 mm above the metal surface of the
barcode scanner housing.
d. Select OK.
The software prompts you to provide the z start height for the de-acceleration
position.

CAUTION
Ensure that the tip adapter sleeve does not touch the housing when the sample probe
ejects a tip. If it does, the tip will not drop correctly. The distance between the tip adapter
and the metal housing must be greater than 1 cm.

WARNING
Ensure that you are clear of subassemblies that can move before selecting OK. Remove
yourself from the machine. The probe moves rapidly and can cause injury.

4. Calibrate the sample probe to calibration point 5 (de-acceleration position):


a. Using the keyboard controls, align the probe so that the lower black housing of
the probe adapter is approximately 10 mm (0.375 in) above the metal housing.

b. Select OK.

Calibrating the Probe to the Sample Compartment Coordinates


After calibrating the sample probe to the tip eject area calibration points, calibrate the
probe to calibration points 6, 7, 8, and 9 in the sample compartment. The sample
compartment is located between the tip eject area and the reagent compartment in the
system interior.

CAUTION
Ensure that the probe is calibrated to the system reference point before proceeding. If the
probe is not properly calibrated to the reference point, damage to the probe could result.

1. Ensure that a tip is present in the left-back position of the right-back tip tray (see
below).
This tip is used for determining the z-coordinate during this calibration procedure.

2. At the Probes Calibration window, select the sample compartment area.
3. At the Attention window, select OK.
The Adjust Position window displays. The sample probe moves to calibration
point 6.

4. Calibrate the sample probe to calibration point 6 (left-back position):


a. Using the keyboard controls, align the probe to the calibration pin.

b. Select OK.
The probe moves to the next position.
5. Calibrate the sample probe to calibration point 7 (left-front position):
a. Using the keyboard controls, align the probe to the calibration pin.
b. Select OK.
The probe moves to the next position.
6. Calibrate the sample probe to calibration point 8 (right-front position):
a. Using the keyboard controls, align the probe to the calibration pin.
b. Select OK.
The Tip Pickup window displays.

WARNING
Ensure that you are clear of subassemblies that can move before selecting OK. Remove
yourself from the machine. The probe moves rapidly and can cause injury.

7. Calibrate the sample probe to calibration point 9 (lane 7, position 10):


a. Manually open the sample compartment shutters.
b. If necessary, remove the sample rack from lane 7.
c. At the Tip Pickup window, select OK.
The probe moves to lane 7, position 10 in the sample compartment.
d. Ensure that the tip ejector is centered in the lane 7, position 10 hole of the sample
compartment shutters.
e. Using the keyboard controls, move the sample tip down until it just touches the
bottom surface of the sample compartment.

f. Select OK.
The sample probe drops the tip in the tip eject area and moves to the home
position. This confirms that the tip eject coordinate is correct.

Calibrating the Probe to the Sample Probe Incubation Ring


Coordinate
Calibrate the sample probe to calibration point 10 (left ring position) – one of two
calibration points on the incubation ring. The incubation ring is located behind the
reagent compartment in the system interior.

CAUTION
Ensure that the probe is calibrated to the system reference point before proceeding. If the
probe is not properly calibrated to the reference point, damage to the probe could result.
1. Remove the plastic incubation ring cover:
a. Using a Phillips screwdriver, loosen the captive screw at the front of the
incubation ring cover.
b. Using a Phillips screwdriver, loosen the captive screw at the front of the cuvette
loader assembly.
c. Remove the cuvette loader without disconnecting the ribbon cable and set it
aside.
d. Remove the plastic incubation ring cover.
e. Replace the cuvette loader.
2. At the Probes Calibration window, select Init to home the system subassemblies.

3. At the Probes Calibration window, select the sample probe incubation ring coordinate
area.
The Adjust Position window displays. The sample probe moves to calibration
point 10.

4. Calibrate the sample probe to calibration point 10 (sample probe incubation ring
position for three calibration pins):
a. Using the keyboard controls, align the probe to the calibration pin.`

b. Select OK.
The incubation ring moves the additional calibration pins beneath the probe.
c. Repeat this procedure for two additional pins on the incubation ring.
For these additional pins, the probe is calibrated to the same x- and y-coordinates
saved for the first point. Calibrate the probe to the z-coordinate only. After the
probe is calibrated to the third calibration pin, the probe moves to the home
position.
5. Replace the plastic incubation ring cover:
NOTE: If you will be calibrating the reagent probe to the reagent probe incubation
ring coordinate, skip these steps now, and perform them after calibrating the reagent
probe.
a. Remove the cuvette loader without disconnecting the ribbon cable and set it
aside.
b. Replace the plastic incubation ring cover.
c. Replace the cuvette loader.

Calibrating the Reagent Probe


NOTE: The distance from the tip of the probe to the bottom of the reagent probe housing
must be 110 mm (4.33 in). This is a critical dimension. Ensure that the final dimension is
precisely 110 mm (4.33 in). Perform this check before calibrating the reagent probe.

Calibrating the Probe to the System Reference Point


The coordinates for all the calibration areas are related to the system reference point.
Always calibrate the reagent probe to the system reference point before calibrating the
probe to the other coordinates. If you change the reference point calibration coordinate,
all other calibration point coordinates will change.

The reagent probe is calibrated to the same system reference point (calibration point 8) as
the sample probe.
1. At the Probes Calibration window, select Reagent Probe Reference.
The Calibration window displays. The reagent probe moves to the system reference
point (calibration point 8).

CAUTION
Do not select the OK button unless the calibration is complete for that coordinate. If you
need to stop the calibration before completing the procedure, select Cancel. Selecting OK
prior to completing the procedure will overwrite the previously calibrated coordinates,
and the probe could be damaged.

The probe moves to the home position.
Calibrating the Probe to the Ancillary Reagent Area Coordinates
Calibrate the reagent probe to calibration points 11, 12, and 13 in the ancillary reagent

CAUTION

1. Remove the top shutter of the reagent compartment.
If you remove the front panel to gain a better view of the calibration points, ensure
that you reconnect the front panel connector before calibrating the reagent probe to
the incubation ring position.
2. Open the reagent compartment door.
3. Load both ancillary reagent trays into the area without reagent packs.

The reagent probe moves to the area. The Adjust Position window displays.

Calibrating the Probe to the Reagent Probe Incubation Ring

CAUTION
Ensure that the probe is calibrated to the system reference point before proceeding. If the
probe is not properly calibrated to the reference point, damage to the probe could result.
1. If necessary, remove the plastic incubation ring cover:
a. Using a Phillips screwdriver, loosen the captive screw at the front of the
incubation ring cover.
b. Using a Phillips screwdriver, loosen the captive screw at the front of the cuvette
loader assembly.
c. Remove the cuvette loader without disconnecting the ribbon cable and set it
aside.
d. Remove the plastic incubation ring cover.
e. Replace the cuvette loader.
2. If necessary, select Init to home the system subassemblies.
3. At the Probes Calibration window, select the reagent probe incubation ring
coordinate area.
The reagent probe moves to the reagent probe incubation ring area.

4. Calibrate the reagent probe to calibration point 19 (reagent probe incubation ring
position):
a. Using the keyboard controls, align the probe to the calibration pin.

b. Select OK.
The incubation ring moves the additional calibration points beneath the probe.
c. Repeat this procedure for three additional points on the incubation ring.
For these additional points, the probe is calibrated to the same x and y
coordinates saved for the first point. The probe only needs to be calibrated to the
z coordinate. After the probe is calibrated to the fourth calibration point, the
probe moves to the home position.
5. Replace the plastic incubation ring cover:
a. Remove the cuvette loader without disconnecting the ribbon cable and set it
aside.
b. Replace the plastic incubation ring cover.
c. Replace the cuvette loader.
