ELECTRON MICROSOCPE NOTES

 The Electron Microscope is able to magnify images in nano meter.


 The TEM is used to view very thin specimen.
 TEM is used to give the images of interior parts of the cell, structure of proteins and the
arrangement of protein molecules in cell membrane.
 SEM is depends on the secondary electrons from the surface of the specimen.
 SEM is otherwise called as sterio light microscope because of the great depth of focussing
ability.
 The organisim which are not view by TEM that can be achieved by SEM like particle counting
and size determination.
 Electrons can be scanned by SEM then it passes through the specimen.
 The TEM and SEM microscope exhibiting the shorter wavelength.
 The TEM is a efficient tool for high special resolution of structural and chemical
characterization of the organisims.
 TEM can be observe atoms at resolution close to 0.1 nm.
 This TEM is used to view the crystalline specimen.
 So, the TEM is powerful tool for obtaining information such as size distribution.
 The TEM has high resolution than SEM.
 TEM is capable of generating atomic level images on both bulk and nano materials.
 The TEM is used to magnify the specimen must be ultra thin sections less than 100 nm thick.
 TEM has three essential system,
 Electron gun and the Condenser system : Electron gun which produces the electrons
and the condenser system which focuses the electrons on the specimen.
 The Image Producing System : It consist of objective lens and movable specimen
stage.
 The Image Recording System : Which converts the electron image into some form
perceptible to the human eye.
 The image recording system usually consist Of fluroscent screen for viewing and focussing
the image and the digital camera is there for keeping permanent records.
 In addition to vaccum system it consist of thumbs, gauges, valves and power supply are
require.
