The screening of circulating miRNAs of interest will be performed by RT-qPCR. Briefly, total RNA will be
isolated from plasma and cardiac tissue at the 4 th and 8th week of experiment using RNAzol RT and will
be reverse transcribed in order to obtain cDNA using the MystiCq microRNA cDNA Synthesis Mix. Lastly,
quantification of the targeted miRNAs will be obtained using the MystiCq microRNA SYBR Green qPCR
ReadyMix and the MystiQ microRNA qPCR primers. All products required for miRNA quantification will
be purchased from Sigma Aldrich. Reactions will pe performed on a (nume instrument) using the
standard PCR conditions.
