IDENTIFICATION AND ANTIMICROBIAL RESISTANCE TO BACTERIA Escherichia
coli ISSOLATED FROM PLASTIC SHEET GUN INSEMINATION MADE IN BALI
COW THAT HAS REPEATED MARRIAGE IN CENTRAL LOMBOK KOPANG ABSTRACT The province of West Nusa Tenggara (NTB) has been recognized as a major area for cattle production under the One Million Cows Program(BSS) and Special Efforts for Compulsory Pregnancy Cows (UPSUS SIWAB).Artificial Insemination (IB) is one of the efforts made by the government to make the program a success. However, there are several factors that cause IB failure namely the presence of bacteria Escherichia coliin the reproductive tract of female cows and if it is not controlled in giving antibiotics it will cause problems with bacterial resistance to antibiotics. The purpose of this research is to isolate and determine resistanceEscherichia coliTetracycline, Gentamicin and Penicillin G antibiotics. This research was conducted in March 2021, sampling was carried out in Kopang, Central Lombok, while sample examination was carried out at the Public Health and Calibration Laboratory of West Nusa Tenggara Province. This study used six Balinese cows that experience repeated mating. The sample in the form of uterine fluid was found through the Artificial Insemination plastic sheet gun. A sterile plastic sheet is inserted into the vagina and then across the cervix to the body or horns of the uterus. The ends of the plastic sheets were cut and put into (BHI) Brain Heart Infusion and incubated for 24 hours. The sample was then implanted in Blood Agardan incubated for 24 hours at 37˚C in the incubator. Bacteria that grow will be stained with Gram stain and characterized by biochemical tests and analyzed based on Bergey's manual of determinative bacteriology (Holt et al, 1994). Furthermore, resistance testing was carried out using antibiotics, namely Tetracycline, Gentamicin, Penicillin G. The results showedwith the disc diffusion method is 0 mm or no inhibition zone is formed. keywords: Escherichia coli, Resistant, Tetracycline, Gentamicin, Penicillin G.
