Hendro Pranoto, S.Pd., M.Si., Eko Prasetya, M.Sc.

1. You are given the task of analyzing the morphological structure of sperm taken from the
epididymis. Determine what method is most suitable for use in the analysis. Also describe
the work procedures that you have to do according to the method you have chosen.
2. Someone gives you fresh liver to make a permanent histological preparation of the liver
which will be used for the purpose of observing the effect of toxic substance X on the
structure and function of the liver. Determine the fixation method, the choice of the dye to
be used and the staining procedure to be used that permits the people to achieve their
research objectives.
3. You are given a bone specimen for permanent histological preparation. How can you
fulfill this request? What method should you use to make bone section using a
microtome? Describe your work scenario (step by step) from fixation to labeling.
4. Name at least 5 things must be considered when cutting the specimen block with a
microtome in order to obtain good sections. Describe what each of these considerations
means.
5. Describe briefly but sequentially correct how to take measurements with a microscope
(micrometry) using an ocular micrometer and a stage micrometer (objective micrometer).
6. Write a brief entrepreneurship proposal related to microtechnique. The proposal should at
least consist of: title, objectives, microtechnical linkages, rationale, economic prospects,
and sustainability.
