Analysis of the HLA B27 alleles

Major Histocompatibility Complexes (MHC) code for a multitude of histocompatibility


antigens (HLA). These antigens determine the immunological individuality of each human.
It is well known that certain diseases are strongly asscociated with specific HLA profiles - so
it is also in the case of Morbus Bechterew (ankylosing spondylitis) and HLA B27 alleles. In
Europe, about 10 – 15 % of all persons are HLA B*27 – positive.
Therefore, the detection of HLA B*27 alleles is performed as pre-screening for potential
M. Bechterew patients and 95 % of all patients are positive. Subsequent subtyping of positive
samples in this pre-screening allows a further differential diagnostic.
Especially individuals with unclear symptoms for the disease benefit

develop M. Bechterew and only 5% of all patients remain B*27-

The MutaGEL® HLA B27 test (KT0003) from Immundiagnostik
allows the detection of HLA B*27 allels in an easy and effective way
(ready to use PCR tubes). The pre-screening test-kit is sufficient for
24 determinations. A small sample of EDTA-blood is necessary for
a preliminary extraction of DNA (isolation reagents are not inclu-
ded) which is used as matrix in the kit. The subsequent processing
with molecular biological methods leads to specific amplification
products detectable by gel electrophoresis.
