ABSTRACT

Background: According to the World Health Organization (WHO), in 2016,


severe periodontal disease causing tooth loss was ranked 11th in the world.
Periodontitis is usually caused by Gram-negative bacteria colonizing subgingival
plaque, one of them is P.gingivalis. Other than mechanical treatment, the use of
mouthwash and antibiotics are carried out by using natural ingredients such as P.
amboinicus leaf extract which has antibacterial effects.
Purpose: This study aims to determine the difference in the effect of the ethanol
extract and the ethyl acetate extract of P. amboinicus leaves on the growth of P.
gingivalis. Method: This study uses the disk-diffusion method with P.
amboinicus extract, fractions of hexane, ethyl acetate and ethanol as test materials
for P.gingivalis cultures. Then, aquadest as negative control, while chlorhexidine
and tetracycline as positive control.
Result: The result of this study shows that at 24 hours measurement, it can be
seen that the average diameter of the inhibition zone for the ethanol fraction with
a concentration of 25%, 50% and 75% (mg / ml) is 10.2 mm, 12.017 mm, and
13.017 mm. The average diameter of the inhibition zone for the ethyl acetate
fraction with a concentration of 25%, 50% and 75% (mg / ml) was 10.05 mm,
11.583 mm, and 12.133 mm. Conclusion: The ethanol fraction and ethyl acetate
of P. amboinicus leaves can inhibit the growth of P.gingivalis. The data obtained
showed that the inhibition zone produced by the ethanol extract was greater than
that of the ethyl acetate extract.
