Introduction

I have done entrepreneurship development course in SBNM College of Agricultural


Biotechnology, Achloli, Mahad. The college was established in 2014 in affiliated to Dr.
B.S.K.K.V. Dapoli, Ratnagiri. The collage has modern and spacious laboratories. The
departments were fulfilling with expertise in Plant tissue culture, Molecular biology,
Microbiology etc. Plant tissue culture; provides disease free and healthy plantlets in large
quantity at a time which have same physiological appearance for farmers and other interested
parties. It aims to make entrepreneurs from students.

OBJECTIVES

Objectives of Plant Tissue Culture:-


• To propagate large quantity of good quality of plant material from elite mother plant.
• Aim to produce quality plant variants for farmers and other interested parties.
• To create more employment opportunities.
• To adopt the latest technology aimed at producing better quality products at lower
costs.

Location:
• SBNM College of Agricultural Biotechnology, Achloli, Mahad, Raigad.
• Tissue culture laboratory located in moderate climatic conditions.
• Having uninterrupted supply of water and power.
• All operations carried out under controlled conditions of temperature.

Land and Building:


• Approximate 2.5 hector adequate for College of Agricultural Biotechnology.
• Building is located away from source of contamination.
• The site accessible to service road, water supply, electric line.

The following facilities in the building:


 Storage room for chemicals.
We can store chemicals, glassware and plastic ware in the storage room. All the material
required for plant tissue culture are stored in storeroom.

Washing room:
This is very important for a tissue culture laboratory. It should be provided with a large
sink, running hot and cold tap water, brushes of various sizes, detergent and a bucket of single
distilled water for a final rinse of the washed glass goods. A number of plastic buckets are re-
quired for soaking the glass goods to be washed. Another separate bucket with lid is also
required for disposing off the used or infected media before cleaning. Only this bucket should be
kept outside the room or cleaning area and should be cleaned twice-in a week.

Media preparation room:
The area dedicated to media preparation should comprise of sufficient storage space for
the chemicals, culture glassware/vessels etc. Additionally, proper bench space for small
equipment for example: hot plates/stirrers, pH meter, balances, water baths, and media
dispensing equipment should be available. Other necessary equipments which are needed in
media preparation and decontamination include two autoclaves including one for sterilizing the
media before initiation of the experiment and second for sterilization of waste and leftover
materials/cultures/infected media after the experiment, refrigerators and freezers to store stock
solutions, and a convection oven for heating the solidified stored medium.

Sterilization room:
In this room Autoclave is used for sterilization of glassware and other material. The
method of choice for sterilization in most labs is autoclaving; using pressurized steam to heat the
material to be sterilized. Sterilization of plant media and autoclavable plastic wares, can
normally be achieved in 15 minutes by autoclaving at 121.6° C and15 psi pressure. This method
is also useful for sterilization of glassware, cotton, forceps, scalpels etc

Inoculation room:
This room should be without any window or ventilator in order to make it dust-free. The
room should be provided with double doors. The doors should have a automatic door closer. In-
side floor should be fitted with a rubber mat to facilitate cleaning. For entering into the room,
shoes should be left outside.

Culture room:
The culture room means the room for keeping or incubating the culture under controlled
temperature, light and humidity. The culture room is also fitted with double doors in order to
make it dust free and to maintain a constant room temperature. One should enter the culture
room keeping the shoes outside the door. To maintain the temperature around 25 ±2 ° C in-sides
the culture room, air coolers are used.

Instruments:
Major equipment and instruments in this plant tissue culture laboratory are as follows:

 Laminar Air Flow- Laminar air flow provides a work area with aseptic/sterile
conditions for the tissue culture. It has continuous displacement of air that pressure
through HEPA filter that removes the particulate from the air.
 Hot Air Oven– It is use extremely high air temperature over several hours to destroy
microorganisms and bacterial spores. A hot air oven is a type of dry heat sterilization. It
is used for sterilization of glassware’s, forceps etc.
 Autoclave (Horizontal and Vertical) –It is used to sterilize medium; glassware’s
and tools for the purpose of plant tissue culture. Sterilization of material is carried out by
increasing moist heat (121 0C) due to increased pressure inside the vessel (15 psi) for 20
minutes.

 Weighing Balance - To weigh various nutrients and chemicals of the preparation of
the media.
 pH meter- It is for the adjusting the pH of the medium.
 Hot plate magnetic starrier- It used to make a stir bar, immerse in a liquid, quickly
spine, or stirring or mixing a medium.
 Double Distillation Unit- This is distiller for producing distilled water for laboratory
research.
 Microwave oven – Nutrient medium can be sterilized and mixing/dissolving
chemicals using household type microwave oven.
 Refrigerator – It is essential for storing various thermo- labile chemicals like
vitamins, hormones, amino acids, coconut milk etc. stock solutions of salts are also kept
to prevent contamination.

Entrepreneurship in Plant Tissue Culture:


• The capacity and willingness to develop organize and manage a business venture like
plant tissue culture is always challenging in order to make a profit.
• Entrepreneurship is generally combined with land, labour, natural resources and
capital can produce profit.
• Careful planning is an important first step when considering and location of a
laboratory.
• A small lab should set up first until the proper techniques and markets are developed.

Problem:
• Tissue culture is an advanced technique to produce plants rapidly.
• The problem face by tissue culture laboratory if the experiment is not properly done,
maintaining complete aseptic conditions.
• Contamination in plant and browning of media is a problem that can face by a tissue
culture laboratory.
• Weather and climatic condition is a problem that can face by a tissue culture
laboratory.
• Problem is also face while transportation of plantlets.
• This will affect the time, money, and effort that you’ve put into culturing plants.

Solving:
• The problem should solved by maintaining aseptic condition that can avoid
contamination.
• The transportation should be done properly for avoiding the breakage of plant.

Networking and Communication:


• It is most valuable skill that you should have in every kind of business.
• Communication is done through various methods like Advertisement, Phone, and E-
mail etc.

Marketing:
• Understanding the market trend is an essential prerequisite in Tissue Culture area.
• The marketing is done in a village.
• The plants are selling in nurseries, to the direct to the farmers.
• You should have proper planning and proper team to do the best marketing of the
product.
• There are also many other options available for marketing.

SWOT Analysis:

Every organization or company their required analysis of SWOT system for successful
entrepreneur. SWOT analysis is a strategic planning and strategic management technique used to
help a person or organization to identify strengths, weaknesses, opportunities, and threats related
to business competition or project planning.

Strength
• Our plant tissue culture laboratory gives good offer , if buying in bulk
• Good Post service provided by our industry (as we have B.Tech biotech degree)
• We have skilled, knowledgeable and supportive staff
• We have all the modern equipment required for the product
• Supportive climatic condition.

Weakness
• Our experience in this field is low compared to other sellers.
• We have some financial issue and it requires more investment at initial stage
• If not a sell produced plantlets then these plantlets are burned.
• Care of this crop is expensive.
Opportunities
• Our product has opportunity in tissue culture industries
• We have opportunity to expand our business in rural population.
• We provide opportunity for aspirants to do work with us.
• Production cost less compared to conventional production method.

Threats
• Competitor may wipe our market positions we have low experience.
• Low market demand as people use traditional method.
• Climate change can affect plant during hardening.
• Have negative publicity.
• It is very difficult to convince the farmer to buy tissue culture plant.

Features:
