4a)Agar plates are incubated with the surface of the agar facing downwards because it helps
stopping water vapour and to prevent contamination from occurring.
4c)You do not want to seal the Petri dish air tight as this will not allowing oxygen to enter as
bacteria is a unicellular organism and needs oxygen to survive.
5a)You shouldn’t incubate it above 25°C as if done, can lead it to multiply rapidly making it very
harmful to go near it and can get contaminated as well.
5b)Replace the culture bottle lid or sterilise the lid to prevent contamination and burn the tip of
the loop to kill any unwanted bacterium that could ruin the experiment. Also when transferring
the bacteria on to the agar, slide it across the agar instead of tapping it in.
