Laboratory Activity No. 5


Fungi

Fungi are eukaryotic organisms. Fungal cell structures are of medical significance: the
fungal cell wall that consist primarily of chitin; and the fungal cell membrane that contains
ergosterol and zymosterol.
There are 2 types of fungi: yeasts and molds. Yeast grow as single cells that reproduce by
asexual budding. Molds grow as long filaments (hyphae) and form mycelium. Some hyphae
form transverse walls (septate) whereas others do not (nonseptate) and are multinucleated called
coenocytic. Most are obligate aerobes whose natural habitat is the environment.

Objective:
1. To identify fungal structures from sample collected.

Materials
Scalpel
Glass slide
Cover slip
Alcohol lamp

Procedure
1. Use a sterile scalpel and scrape from the fungi from samples.
2. Cover with the clean cover slip and press gently.
3. Gently warm the slide over the alcohol lamp.
4. Examine under low power and high-power objectives with reduced lighting and
check for any fungal structures.
Illustrate, describe and label the structures you observed under the microscope.
