Isolation of bacteria involved in the nitrogen cycle from different culture media

RESUMEN
ABSTRACT
Background. The nitrogen cycle is a biogeochemical cycle, which explains the biological and abiotic
transformations and processes by which nitrogen circulates in the planet, it is divided into 3 stages,
ammonification, nitrification and assimilation. Nitrogen-binding microorganisms play a key role in the
cycle and are found mainly in soil, these microorganisms are able to fix atmospheric nitrogen and
transform this gas into compounds such as nitrates (NO3 -) and ammonium (NH4 ), this process
carried out by microorganisms are necessary for the assimilation of nitrogen in plants, since they
acquire nitrogen directly from the atmosphere and fix it in compounds usable for plants . Goal. Check
the nitrogen binding capacity of bacteria present in plant roots and nodules using the isolation
technique and learn how microorganisms influence the nitrogen cycle. Methodology. For the isolation
of Rhizobium spp nitrogen assimilators, the root nodules of the plants were sown by triplicate in YMA
agar, for the isolation of Azotobacter spp, 20-25 ground granules were sown by triplicate in Ashby
agar, for the isolation of Azospirillum spp, the root taken from the grass was sown by puncture in the
medium JMB, everything was incubated at 35 C° for 48 hours, 30 C° for 2 weeks and 22 C° for 3
weeks, respectively. Finally, Gram stain and lactophenol stain were performed to identify bacteria,
fungi and yeasts on the plates that showed microbial growth. Results. From the isolation sowing in
YMA agar, there was no abundant isolation of bacterial colonies, for Ashby agar from the sowing of
the soil granules, a growth of bacteria and a fungus was obtained around the samples of granules, no
microbial growth was obtained from the planting of the root sample in the JMB medium and therefore
no blue umbrella formation was obtained. Finally, yeast, Gram-negative coconut bacteria and two
fungi with different morphology were observed for the stains. Conclusions. The results obtained for
the isolation of Azotobacter sp is not optimal or reliable due to the little Bactrian growth that present
and a methodological review is recommended to optimize the isolates of the microorganisms of
interest.
