Q1/ How is Hb measured in the lab. ?
Hb can be measured using arterial blood. Arterial
blood is oxygenated (i.e., flowing from the heart) and
has higher amounts of oxygen-bound Hb compared
with blood sources not flowing from the heart
Method :-
1. Acid haematin method by using ( sahli apparatus )
depends on comparison of HB with HCL by naked eye
(not recommended) because the color developed
slowly and begin to fade immediately after it reach its
peak
2. Alkaline haematin method:- by using (colorimeter
apparatus) using reagent 0.1N NaOH
also not recommended because som HB resist
denaturation with NaOH & participate in crystals
3- Oxyhaemoglobin method:- apparatus (
colorimeter) reagent 0.4% ammonia in H2O ,
blood sample is mixed with a weak ammonia
solution and then absorbance of this solution
is deliberated in a photometer using a
yellow-green filter or measured in a
spectrophotometer at 540 nanometer.

4-Cyanmethemoglobin method :- highly


recommended method for estimating HB
concentration considered as a reference
method , allow measurement of all type of Hb
except sulfhemoglobin (SHb)
Q2 / what is the mechanism of B12 and folic acid in
erythropoiesis ?

Erythroblasts require folate and vitamin B12 for


proliferation during their differentiation.
Deficiency of folate or vitamin B12 inhibits
purine and thymidylate synthesis, impairs DNA
synthesis, and causes erythroblast apoptosis,
resulting in anemia from ineffective
erythropoiesis.

Drugs:
Chloroquine (Aralen) Food:
Mefloquine (Lariam) fava beans
Pamaquine. red wine
Primaquine. all beans,
Quinidine. blueberries,
Quinine. soya products,
. tonic water and camphor
