Procedure for Polymer Extraction

1. Transfer 100mL of anhydrous Ethanol into a 250mL conical flask, keep the flask for stirring in a
magnetic stirrer with heated plate at 37°C.
2. Add dropwise 1.5mL of sample to the above conical flask with continuous stirring and keep the
flask for 3hrs to form a white coagulate.
3. Decant the ethanol layer completely to leave the white coagulate.
4. Add 100mL anhydrous Ethanol to the same conical flask and keep for stirring for another 30mins
with heated plate at 37°C.
5. Decant the ethanol layer completely to leave the white coagulate.
6. Further add 100mL of anhydrous ethanol to the same conical flask and keep the flask under
stirring at 37°C in a hot plate for overnight.
7. Decant the ethanol layer again and add 100mL anhydrous ethanol, to the same conical flask for
30min under magnetic stirring with heated plate at 37°C.
8. Decant the ethanol layer to get white coagulate.
9. Add 10mL acetonitrile to same conical flask containing only white coagulate.
10. Swirl the conical flask to obtain a clear solution, which contains POLYMER polymer.
11. Add 20mL anhydrous ethanol to above solution to form milky white precipitate.
12. Transfer the solution to a 50mL centrifuge tube.
13. Centrifuge the solution for 60mins at 10000 RPM at 5°C.
14. Decant the upper ethanol layer to obtain POLYMER precipitate polymer in the bottom of the
centrifuge tube.
15. Add 20mL of anhydrous ethanol to the above centrifuge tube containing POLYMER polymer,
swirl and decant the ethanol layer. Repeat the same procedure for one more time.
16. Add 5mL acetonitrile to the centrifuge tube containing POLYMER polymer and vortex for 2 mins
to dissolve the POLYMER polymer.
17. Transfer the clear solution to a 100mL RBF and add 5-7mL anhydrous ethanol to form a milky
white precipitate.
18. Attach the RBF to Rota evaporator and keep at 50mBar, 37°C and 250RPM for complete solvent
removal.
19. Collect the material and dry in a lyophilizer to obtain a solid mass.
