Enhancing the anticancer effect of paclitaxel by using


polymeric nanoparticles decorated with colorectal cancer
targeting CPKSNNGVC-peptide

The lack of specificity in cancer treatment is a key drawback. Selective administration of


chemotherapeutic drugs to tumor areas has advanced rapidly in recent years as a diagnostic and
therapeutic tool in the fight against cancer. Using a tumor homing peptide (CPKSNNGVC, CPK
in short) as a nanoparticle surface-bound targeting ligand, we design a unique polymeric
nanoparticle delivery method that targets both MCT1 receptor-positive cancer cells and
angiogenic endothelial cells. Multipotent drug paclitaxel (PTX)-encapsulated poly (dl-lactide-co-
glycolide) (PLGA) nanoparticles (diameter of 215 4 nm; zeta potential of 12 3 mV; 16–17.5
percent w/w PTX) were manufactured utilizing an emulsion-solvent evaporation process. The
CPK peptide was then attached to the nanoparticle surface using maleimide-thiol chemistry
(CPK-PTX-NPS). In MCT1 receptor-overexpressing colorectal cancer (CRC) cells, the
extremely stable CPK-PTX-NPs demonstrated MCT1 receptor-mediated cellular uptake and
apoptosis-mediated cell death, whereas the other non-targeting nanoparticles failed to show
MCT1 receptor-mediated cellular uptake. In a chick embryo angiogenesis experiment, the
targeted CPK-PTX-NPs preferentially suppressed the development of new blood vessels from
pre-existing blood vessels (angiogenesis). Finally, our findings reveal that MCT1 receptor-
selective, PTX-encapsulated, and CPK peptide-decorated polymeric nanoparticles operate as
effective carriers for CRC-specific drug delivery, resulting in dramatically improved therapeutic
efficacy.
