MANUAL PIPETTE
Prepare all materials a. Place the test tubes on the test tube rack
b. Place enough amount of water in the beaker
c. Place the serological pipette and bulb at a safe area

Proper use of bulb for aspiration of liquid a. Bulb was pressed before placing above the pipette
b. Bulb was placed properly above the pipette
c. No bubbles were formed in the beaker while aspirating
d. Liquid did not go directly inside the bulb

Proper use of pipette a. Pipette is in vertical, upright position while aspirating

b. Tip of the pipette is immersed properly in the liquid
c. Water level in the pipette is in lower meniscus
d. Made sure that the liquid is at zero marking before dispensing

Proper dispensing of liquid a. Bulb was removed and replaced by the finger to cover the top of the
pipette
b. Pipette is in vertical, upright position while aspirating
c. Released the finger and slowly let the liquid drop in the tubes
d. No bubbles were formed while dispensing the liquid to the test tubes
e. No tapping of the pipette while shifting from one test tube to the
other

Prepare all materials a. Place the test tubes on the test tube rack
b. Place enough amount of water in the beaker
c. Correctly choose a pipette for the volume requested
d. Correctly choose the pipette tip to be used

Proper use of pipette a. Adjust the pipettor for aspirating the desired volume
b. Correctly load the tip into the pipette

Proper aspiration of liquid a. Pipette is in vertical, upright position while aspirating

b. Pressed the plunger in the first stop to aspirate liquid into to the
pipette tip
c. Release of plunger was not too fast nor too slow
d. No bubbles formed in the pipette tip

Proper dispensing of liquid a. Pipette is positioned at around 20–40-degree angle with tip touching
the walls of the tube
b. Pressed the plunger at the last stop to dispense and blow out all
liquid in the tip
c. Pressing of plunger was not too fast nor slow, properly dispensing
the liquid inside the tube
d. Made sure all the liquid was dispensed into the tube
e. No bubbles formed in the pipette tip
f. No spillage
