TOPIC: Effect of Ph on action of salivary
amylase
Form: Synopsis
Aim
To study the effect of ph. on action of salivary
amylase
Principle:
Optimal activity of most of the enzymes is
observed as ph. of 5 to 9. some of the
enzymes like active at ph. values outside its
range.
Materials required:
1: Glass test tubes, beakers,
dropers,funnel,
2: Chemicals like Nacl, Na2HPO4, KH2PO4,
iodine crystals, potassium iodide, buffer sol of
ph. 4 and 9

Equipments:
1: water bath, thermometer,cotton,rubber,
distilled water
Procedure:
1: Prepare three set of indicators in three test
tubes
2: In one take 5ml of 1% starch solution, 1ml
Nacl solution,1ml of buffer sol of 6.8 ph. take
it as A
3: take 5ml of starch solution,1ml of Nacl
solution 4 buffer solution. take it as B
4: take 5ml of starch solution, 1ml of iodine
solution,1 ml of ph. 9 buffer solution take as C
5: Drop 1ml of dilute saliva in each of them.
Place them at 37degree Celsius temperature
6: take a drop from test tubes and add to
indicator tubes of iodine solution. Note this
as zero-minute reading
7:at every 2minute repeat the steps till the
colour of iodine solution change
