Molecular Biology of Rejuvenation Processes

Abstract
The last decade has seen an tremendous development of the field of
rejuvenation of cells and tissues. Among the more exciting findings,
was the restoration of vision in retinal cells in old glaucomic mice via
OSK () adenovirus associated vectors and the and the systemic
multiomic rejuvenation of the complete organism using OSKM
cassettes (). It is notable that similar feats can be achieved with small
molecules and non protein factors. The present work analyzes the
relationships among the biological macromolecules as well as the
potential directions of future scientfic investments in order to facilitate
the translation of the lab finding to the a real world setting.

Introduction
In 2012 Yamanaka received a Nobel Prize for his group’s work () on
the use of Oct, Sox, Kef, Myc to induce the transformation of somatic
cells t ostem cell like pluripotent cells. In parallel other laboratories
worldwide developed processes that used related biomolecules to
rejuvenate cells without losing their differentiated state (....)
A particularly valuable work was made by Lu et al (), nothing less than
the restoration of vision in retinal cells in old glaucomic mice via OSK
() adenovirus associated vectors. The Reik group in Germany….the
systemic multiomic rejuvenation of the complete organism using
OSKM cassettes (). Using small molecules and non protein factors ().
In order to comprehend the system that we are observing and decide
what are the optimal directions of future research,it of the utmost
importance to understand the relationships among the biological
macromolecules.

DMT1 Inhibitors Kill Cancer Stem Cells by Blocking Lysosomal Iron


Translocation (Turcu et al. 2022)Cancer stem cells (CSC) constitute a
cell subpopulation in solid tumors that is responsible for resistance to
conventional chemotherapy, metastasis and cancer relapse. The
natural product Salinomycin can selectively target this cell niche by
directly interacting with lysosomal iron, taking advantage of
upregulated iron homeostasis in CSC. Here, inhibitors of the divalent
metal transporter 1 (DMT1) have been identified that selectively target
CSC by blocking lysosomal iron translocation. This leads to lysosomal
iron accumulation, production of reactive oxygen species and cell
death with features of ferroptosis. DMT1 inhibitors selectively target
CSC in primary cancer cells and circulating tumor cells, demonstrating
the physiological relevance of this strategy. Taken together, this opens
up opportunities to tackle unmet needs in anti-cancer therapy.

Conclusion
If we are to get to the point where the science of rejuvenation actually
benefits us, we should consider using DNMT1 inhibitors
accompanied by…PAS1. The first stops the aging by methylation of
DNA, the second inhibits cancerous and metastatic processes.
