Label the tubes

Blank Standard Test

Add 500 uL of Urea Add 500 uL of Urea Add 500 uL of Urea

Color Reagent Color Reagent Color Reagent

Add 10 uL of distilled Add 10 uL of standard Add 10 uL of patient

water reagent serum
 Add 500 uL of Urea Enzyme Reagent to all
tubes
Cover all tubes with parafilm

Mix gently and incubate for 5 minutes at 37 °C

Add 2 mL of Urea Base Reagent to all tubes

Cover all tubes with parafilm

Mix gently and incubate for 5 minutes at 37 °C

Zero spectrophotometer with reagent blank at 630 nm

Read and record absorbance at 630 nm
