Lane: 3.) Well: Part Il: Extended Thinking Questions In the case of this example, what is lane A used for? What section of this sample is negatively charged? How do you know? What does it mean if two bands are found in the same spot in different lanes such as B and C? What if they do not have bands in the same spot? Why might some bands look different than others in the same lane? In this example, they appear lighter colored, why? How might you calculate the percent similarities between two different lanes? What kind of real world applications could you use electrophoresis? Part Ill: Maternity Problems ‘Scenario: A woman has recently finished giving birth and has been waiting to see her baby for three hours, She is so excited to see her child and finally, the nurse comes through the door. She remembers what her baby looks like, and she does not think itis her child in the nurse's arm. Due to this confrontation, the hospital takes a small blood sample from the baby and the mother and runs PCR on the samples, where the DNA is then run through Electrophoresis in order to determine whether or not itis her child. The data is shown in the Example plate on the first page, The mothers sample is lane B, while the baby's sample is lane E. Is the baby from lane E her child? Why or why not? If it is not her child, which lane would have the DNA from her child? How could you tell? Is there any other lanes that could contain the DNA of her child (other than the one in the previous section? Why or why not? In this new scenario, a master thief has gotten away with the British Crown. It was in a glass container, which he had to shatter. Unfortunately he got a small cut while trying to reach his hand through the glass he shatters. A small drop of blood was found, and it was collected by the laboratory. It was replicated using PCR, where it was then ran through electrophoresis with DNA samples from multiple suspects. Using the example plate from the first page, itis your task to determine who the thief is. Who do you think the thief is? Explain your reasoning. If you do not think the DNA of the thief is present, put what the thieves DNA should look like in lane G. Explain why you think it is this way.
