1 INTRODUCTION

1.1 Intended Use


The AVE-772 Integrated Urinalysis Analyzer is combination of Urine Formed Elements
Analyzer and Urine Analyzer. The urine microscopy results can also be combined with urine
chemistry results in an integrated report.The system is designed specifically for professional
use, as an In Vitro Diagnostic (IVD) device used by qualified physicians and laboratory staff.
The system is intended for use in screening to assist diagnosis for urinary system diseases,
kidney diseases, circulatory system diseases or infectious diseases.

1.2 How to use the User Manual


The User Manual is intended to explain the instrument operations in detail, to be a
training guide for new users and to be a troubleshooting reference manual.
This manual is designed for anyone who will operate the analyzer and must have a
thorough knowledge of it. It also specifically references information concerning analyzer
components, operation, theory, utility and performance. The manual is also loaded onto the
instrument computer and can be accessed by clicking the Help menu after entering the Main
Screen.

1.3 Precautions and Warnings Symbols – Definitions


The User Manual includes information and warnings that must be observed by the user to
ensure a safe operation of the system.

1.3.1 Cautions

CAUTION: Identifies situations that could lead to an accident or equipment failure.

CAUTION: Identifies electrical caution! Unplug before handling.

CAUTION: Identifies laser light. A laser is used to read the barcodes only. Protect
eyes from the laser light.
1.3.2 Warnings

WARNING: Identifies potentially hazardous situations that could result in serious

injury to laboratory personnel.

1.7 Use of Third Party Computer Products


AVE does not recommend that the computers used as a functional part of the AVE-772 be
employed for performing any software or hardware based applications other than those
specifically furnished to operate and support the system, or those recommended and offered
by AVE specifically as accessories or enhancements for the system. No other third party
application software should be installed in these computers in addition to those provided or
recommended by AVE, without the expressed approval of AVE Technical Service, in order to
avoid potential performance and reliability problems which can result from incompatibility
factors, errors in use of such software, or software-based “viruses.”
Installation of such third party software, or non-approved electronic cards or other devices,

without advance approval by AVE may affect the terms of or void any warranty otherwise in
effect, covering AVE-772 supplied software and hardware on the computers and the overall
performance and reliability of the entire system.

2 SYSTEM DESCRIPTION

2.1 Operation Theory


The user puts the test tube containing urine sample in the provided rack and place the
rack on the automatic sampling module. The device will detect the rack and automatically
forward it to the sampling position. If the sample is barcoded, the analyzer will read the
barcode, blend the sample thoroughly and aspirate it via the sampling needle into the
counting chamber for testing. The microcamera controlled by the host device scans the
sample in the counting chamber through enlarging the formed elements by microscope and
takes images for analysis and processing. The chemistry part will complete the separation of
chemistry strip and sampling automatically, also the test strip will be conveyed to the
chemistry components to do automatic chemistry analysis. The chemistry part uses CCD
camera to to detect the color changes of urine test strip, calculate the related items’
concentration, and output the semi-quantitative results.
During microscopy, the instrument regulates the focus automatically for different view
field to capture the cell images clearly. Then it recognizes, classifies and counts the particles
in each image with auto-recognition software which works with neural network algorithm.
Each image can be classified into one of 9 categories: normal RBCs, abnormal RBCs, WBCs,
Casts, Epitheliums, Crystals, Fungi, Mucus and others according to advance trained
recognition template.
The instrument simulates manual microscopy using fully-automated machine visual
technology. It can automatically select special analysis processes for different samples
according to the preset parameters. First the instrument screens for negative samples with
low magnification scanning before samples in the counting chamber have completely
precipitated to significantly increase the analysis speed. For positive samples, the instrument
performs LP (low powerfield) targeting and HP (high powerfield) tracking for distinguishing of
cells in order to improve the processing speed and accuracy. These steps ensure the
detection of cells in the un-centrifuged specimens. All the artificial intelligence functions
including data access, analysis and integration are achieved through the software. The
complete Urinalysis report including the images may be printed via a laser printer.

2.3 System Components


The AVE-766 analyzer is a bench top instrument, which consists of Host, PC Module,
Peripherals (including display, mouse and keyboard) and other accessories(such as printer
etc.).
Host consists of the following four modules:

• Automatic Sampler
• Strip Feeder
• Strip Loader
• Chemistry Analyzing module

2.4 Software Functions


The analyzing software is a set of microscopic analysis and urine chemistry analysis in
one, it has the following functions:
•Automatic microscopy function: Automatically detects whether a sample exists,
automatically moves and extracts a sample, and captures images automatically.
•Chemistry analysis function: Automatically capture the image of test strip, identify the
image, then provide the chemistry results.
• Automatic recognition function: Classifies microscopic particles of each sample using
the world's leading machine visual technology.
• Manual review function: The software provides editing capabilities for reclassification of

the particles as decided by the user.


• Barcode scan matching and chemical negative screening function.
• Quality control function: Provides quality control test.
• Data inquiry and report printing function.
• Data backup and restoring function

2.4.1 Main Programme Screen


After booting, the user can enter the main programme screen as shown in below through
log-in. The function menu bar is located at the top of the screen, clicking on the different
drop-down menus enables entering each function of the operation interface. The second row
shows shortcut keys to the menu.

The common shortcut keys are located in the shortcut key area and are convenient for daily
use, including microscopic examination, verification, urine history verification, urine inquiries, user
management, data backup, quit the system buttons, as shown in Figure 2-6. Clicking on the
corresponding function key enable the corresponding operation or entering the appropriate
function operation interface.

2.4.2 Microscopy and Verification Screen


After entering the Main Screen, to enter the Microscopy and Verification (Microscopy)
screen by one of the following ways:
1: Click “Operation”-“Microscopy” to enter the Microscopy Screen.
2: Click the “Microscopy and Verification” icon in the Shortcut keys area to enter the
Microscopy Screen.

2.4.2.1 Urine Real-time Monitoring Screen


As shown in Fig.2-14, the microscopy screen consists mainly of the Online status
Area,the Instrument Adjustment Area, the Special Specimens Handling Area, the Microscopic
Field, the Specimen ID Area, the Patient Information Area, the View Switch Area and the
Channel Status Bar. Their respective functions are
Online Status Area: Display online status information of each part of the instrument as
well as convert its start and stop working state.
Instrument Adjustment Area:Common instrument functions can be adjusted here.
Special Specimens Handling Area:Some handlings on special specimens (QC, STAT,
reinspection, etc.) can be operated here.
Microscopic Field:Display microscopic images.
Patient Information Area:Display the detail information of the selected patient samples
as well as edit and modify it.
Specimen ID Area:This area includes both urine routine and special microscopy data
lists which display the data of tested specimen, including Specimen ID, barcode, microscopy
ID, chemistry ID and sample status. Double-clicking on the selected specimen ID opens its
microscopy screen.
View Switch Area:Mainly switch between real-time monitoring screen, urine microscopy
screen, and daily maintenance settings.
Their functions are:
A. Online Status Area:
“Start” button: A dynamic button whose initial status is “Begin microscopy”, will turn grey
after being clicked. The “Pause” button can also be clicked to achieve conversion between

the two states. Functions: The instrument will fulfill the current sample analyzing by clicking
“Start” . This operation is for normal microscopy. The instrument will stop working temporarily
on clicking “Pause” and turn to maintenance state. The user can conduct a relevant quick
maintenance and resume its work by clicking on “Continue”. It will be converted to the specific
channel by clicking the corresponding one and begin sampling, focusing and washing.
“Stop”: Stop testing normally. The system will stop working after finishing the sample,
which is currently tested in the cell by clicking on “Stop”. The next sample on the current rack
will not be tested.
“Forced”: Forced to stop testing. The system will stop testing both the sample in the cell
and the next one on the current rack by clicking on “Forced”.

B. Instrument Adjustment Area:


“↑ / ↓”:Be used for focus fine-tuning. “↑”in the first row means each click equals a focus of
the microscope upward 5 steps, while “↓” in the same row means each click downward 5
steps. “↑” in the second row means each click 10 steps upward and “↓” 10 steps downward.
“HP / LP”: Realize the microscope HP/LP conversion.
“Save”: If you need to save the focus you’ve adjusted, just click this button.
“Sample”:Sample through a single selected channel.
“Wash”: Washing of a single selected channel (Cleanser A).
“Spe-Wash”: Compulsorily washing a single selected channel (First the channel is soaked
with Cleanser B, then Cleanser A is used for cleaning).
“Wash+”: Wash a single selected channel with double volumn cleanser (Cleanser A).
“Manual”: Allow for manual capturing of microscope pictures.
“Pause”: Click this button will pause the instrument, while performing any regulation in
Instrument Adjustment Area.

C. Special Specimens Handling Area:


“Insert” button: Manually insert the type and ID of the next special specimen, mainly for a
special specimen into the consecutive ones.
“Insert at”: Enter the insertion position of a special specimen into the consecutive ones.
System defaults the test of the special sample in the position.

D. View Switch Area:


“Microscopy”: Switch to the real-time monitoring screen of testing.
“Verification”: Start report review procedure. The user can perform the manual verification
of the microscopy results here for the final examination report.
“Tools”: Click this button to pop-up a drop-down menu on daily maintenance and settings,
choose any of the options to enter the corresponding screen.

E. Patient Information Area:


“Print” Print the specified sample results report.
“Prview” Preview the specified sample results report.
“Batch” Print the specified batch of samples results report.
“Save” Save the recorded or modified information of the patients.

As shown in Fig.2-15 select “Verification” button, then the corresponding view is Urine

The verification screen consists mainly of the Urine Information List, the Patient
Information Field, the Current Cells Couting , the Chemistry Data, the Function Key, the
Frame and the Image Display.Their respective functions are:
“Urine Information List”: Display the information about samples being microscoped.
“Patient Information”: Display the information about the patient and verify or modify the
given information.
“Frame”: Show the captured frames of the testing sample and the selected image’s frame
number.
“Current Cell Counting”: Display the amount of items in current sample tested under HP/LP.
“Chemistry Data”: Display the real time received chemistry data.
“Functional Key”: Include some common functional buttons for verification, such as “Modify”,
“Previous”, “Next”, “Zoom”, “Print”, “Preview”, “Batch Print”, “Save” and “X”.
“Image display”: Display the images of the current sample captured under HP or LP.

A. Functional Buttons
“Modify”:Clear out the counting data and cell type of the current picture and change to
another.
“Previou”: Select the previous sample.

“LP/HP”: This button is a two-way button. LP/HP pictures can be selected through pressing it
alternately.

modifying the data.

B. Image display field


16 frames of the image captured are displayed here. The pictures can be divided into LP
and HP. If user select “LP” on the left of the “Current Cell Counting” field, then the pictures
captured under LP field appear. If select “HP” on the right, then the pictures under HP field
appear. Double click on any picture displayed in this field will enlarge the picture to a full-view.
Double click on the picture again will diminish it again.

C. Chemistry Data
Click the “▲”or “▼”button to preview the chemistry data received.

D. Current Cell Counting field


Detail information about the testing items are displayed in this field. The information given
are: “Result”, “Frame” and “Total”. Data in the “Frame” column are the data of the current
pictures captured; “Total” shows the total of the item tested under LP or HP.

E. Frame
The frame information of the current sample appears here.

3 INSTALLATION

3.1 Summary
Anyone who is not trained by AVE or authorized distributors of AVE may cause damage
to the analyzer when opening the box or installing the instrument. Therefore do not open the
box or install the instrument without an authorized technician being present.
On receiving the analyzer, please check the package carefully to see whether there is a
physical damage. If there is any, please notify your local distributor immediately.

3.2 Installation Requirements

3.2.1 Space
AVE-772 Integrated Urinalysis Analyzer (with its mainframe size of
825(L)x648(W)x550(H) mm) is a tabletop unit requiring free space on a countertop of
approximately 250x120 cm, with vertical clearance of at least 30 cm.

3.2.2 Power

CAUTION: UPS no less than 1500VA or voltage regulator are recommended for the
whole system operation if voltage is not stable or power outage is regular.

the same type and rating fuse.

WARNING: Equipment requires connection to protective earth for safety reasons.

WARNING: The instrument’s main supply inlet is being used as the mains
disconnect device.

3.2.3 Environmental requirements


The instrument is suggested to be installed in the environment as:
• Normal working temperature: 15°C – 30°C
• Relative humidity:30% - 70%;

• Do not get close to strong electromagnetic source as not to affect the normal
operation of equipment.
• Keep the environment as clean as possible. Keep it free from mechanical vibration,
noise and power interruptions.
• Avoid direct sunlight or exposure to heat or wind.

3.2.4 Installation
The AVE-772 Urinalysis System has to be installed by an authorized technician.

3.3 System Connection


The authorized technician will complete the following connections when installing the
analyzer.
The system connects with other peripherals as shown in Figure 3-1. Ensure connecting in
place and firmly.

3.4 Pipeline connection of the analyzer

Warning: User has the duty to comply with the relevant regulations set by the region
and country about the treatment and discard of expired reagents and wastes.

Warning: Reagents may cause some irritant to skin and eyes. User should always
comply with the safety operation of laboratory and wear personal protective equipment (such
as protective clothing, gloves, etc.) when coming contact with reagents.

Caution: Do not place reagent containers on the Analyzer


The system is connected with reagent bottles as shown in Figure 3-2. Ensure connecting
in place and firmly.

3.5 Power-cable connection


The main power-cable of AVE-772 connects the input power-outlet on the rear-side and the
power distribution board special for the system. Make sure that the power of AVE-772 and the
power distribution board is off before the connection is completed .Complete the cable connection
of the monitor and printer.

4 SETUP

The analyzer settings are already set by the factory and the screen you see after the
initial boot is set on system default. In order to meet the individual demands, some
parameters of the analyzer can be modified. Since the settings will directly affect the normal
use of the instrument, an access control has been set up for the user to ensure the safety of
the equipment.
There are two modes of use and appropriate rights: The "user" can perform clinical
urinalysis in daily use, but can not access "Management" main menu. In addition to the rights
owned by the "User", the "Administrator" can modify specific parameters on the
"Management" main menu.

CAUTION: The chapter is for “Administrator” only.

CAUTION:Changing of the factory parameter settings could affect the normal use of

the instrument , if it’s necessary, please read the user’s manual first or contact manufacture.

4.1 User Set

4.1.1 User login


You can enter the user set through the following two ways on the system software main
screen.
1. Click “Management”-User Set”.
2. Click the “User Set” icon in the Shortcut keys area.

4.1.2 User Set Operations


Operations as adding a new user, deleting a user and modifying a user can be performed
on the “User Set” screen.
Adding a new user:
a. Type the new user identification in the “User” field.
b. Type the new user password in the “Password” field.
c. Click drop-down menu in the “level” field to select the user level
d. Click “Add” button.
Delete a user:
a. Select the user to be deleted.
b. Click the Delete button.
Modify a user
a. Select the user to be modified.
b. Alter the user identification in the “User” field as needed.
c. Alter the user password in the “Password” field as needed.
d. Click drop-down menu in the “Level” field to alter the user level as needed.
e. Click “Save”.

4.1.3 Password Modification


1. Click “Management” in system main menu, select “User Set” in the drop-down
submenu as shown in Fig.4-1. The “User Set” application screen (as shown in Fig.4-3) open.
Following the instructions of “Modifying a user” in 4.1.2 the administrator can modify any

user’s password.
2. Click the daily operation main menu, select “Password” in the drop-down submenu as
shown in Fig.4-4. The “Password” modifying window will open. The password of the login
user can be modified in this window. This is available for all users.

4.2 Microscopy Setting


Click “Management” in system main menu, select “Microscopy Setting” in the
drop-down submenu, the three secondary submenus “Item Setting” , “Reference Values”
and “Comparison List” will open.

4.2.1 Item Setting


On this screen selecting “Microscopy” in the “Item Type” will allow to add, delete, alter
and sort the items.

CAUTION:It is not recommended to change the parameters in this interface, if any,

please contact technicians or manufacture.

4.2.1.1 Urine Item (Setting) Screen


Parameter: the type of formed element
Display Name: the item name as displayed in the microscopy verification list
Sen. Para. (Senior Parameter): the senior parameter of the item. The parameters are now
available RBC, WBC, Crystal, Cast, Epithelium, Yeast, Bacterium, Parasite, Sperm and
others. Senior parameter setting will default a new added item but not classify it to a senior
parameter with the same name otherwise lead to a logical error.
Unit: the unit of the item
Reference: the normal reference value. The reference values of RBC, WBC, Crystal, Cast,
Epithelium and Yeast shouldn’t be set here but being set in Urine Micro Reference interface.
See 4.2.2 for details.
Display type: Set the item name displayed in HP/LP Microscopy Verification List and right
click menu. Both means the item displays under both HP & LP, LP display means the item
displays only under LP and HP display means the item displays only under HP; Neither
means the item will not display under HP or LP.
Print: Whether to print this item or not.
Identify: Whether to identify the item automatically or not. It is a default setting, modification is

not recommended. For a new added item choose “No”.


QC: Whether to run a morphological QC. It is a default setting, modification is not
recommended. For a new added item choose “No”.
Identifier: the ID code of the item. It is a default setting, modification is not recommended .
The ID code of a new added item should not exist.
Remarks: Add notes for the item.
Delete: Delete the selected parameters.
Up/Down: Move up/down the selected parameters for sorting.
Quit: Close the modifying interface.
4.2.1.2 Add Urine Items
1: Choose a blank row, enter the appropriate parameter, display name, unit, reference (if any);
set display type, print or not; choose No for Identify and QC, set corresponding identification
as “-1”.
2: The new added item will be saved automatically after enter.
4.2.1.3 Delete Urine Items
1: Select the urine item in Urine Item interface. Caution: only items added as described
under 4.2.1.2 can be deleted. Systems items cannot be deleted.
2: Click the Delete button to confirm the operation.
4.2.1.4 Move Urine Items
1: Select the urine item in Urine Item interface.
2: Click the Up or Down button to change the location of the item in the list.

4.2.2 Reference Values


On this screen the user can set gender, maximum age and minimum age for the
reference value of RBC, WBC, Crystal, Yeast, Cast and Epithelium.

Reference Value of various age range and gender on this screen can be added, deleted
and altered.

Add operations:
1. Choose Female or Male in the Gender box.
2. Select a blank row, enter maximum age, minimum age and the corresponding reference
values of RBC, WBC, Crystal, Yeast, Cast and Epithelium.
3. Click Save button and click “OK” in the pop-up window to validate the adding.
Delete operations:
1. Choose Female or Male in the Gender box.
2. Select the row of reference values of age range to be deleted.
3. Click Delete then Save button, and click OK in the pop-up window to confirm.
Modify operations:
1. Choose Female or Male in the Gender box.
2. Select the parameter to be altered.
3. Enter a new corresponding parameter,
4. Click Save button and click OK in the pop-up window to confirm.
5. Click Quit to exit.

4.2.3 Comparison List


By setting the quantitative and qualitative data comparison of urine formed elements, the
Urine Comparison Table achieves the automatically comparison between sample’s strips and
sediment results, analyzing whether the two results are matched or not under clinical
diagnosis standard and whether reminding current user to verify the results or not. It is a
default setting, modification is not recommended. If any, please operate in strict accordance
with clinical requirements. As shown in Fig.4-6, select the secondary submenu Comparison
List to enter the Comparison List interface.

Each comparison parameter on the Urine Comparison Table setting screen can be added,
deleted and altered.

Save: Save newly added or modified parameter.

Select the “Chemistry Protocol” in “Management” menu to pop up the setting screen
shown in Fig.4-12. It is used for AVE-772 to set communication protocol with chemistry
analyzers.

4.3.2.1 Chemistry Messaging parameter setting


Instructions:
New:New added protocol name.
Protocol: Existing protocol name. In General, chemistry analyzers made in China can use
“common chemistry analyzer”interface program.
Items: This is used for the converting of external chemical data into the corresponding
software report data. For example, if the chemical result for urine occult blood is “Neg” and we
set the report data as“-”, the software can convert it into“-”automatically.
Add: Add a new protocol.
Delete: Delete the corresponding protocol.
Save(upper left): Save the protocol newly added, deleted or modified.
Save(lower left): Use the current selected protocol.
Quit: Quit the screen.
Test: Test the communication of the chemistry analyzer connected and transmission of the
results.
Clear: Clear the results.
4.3.2.2 Adding Existing Chemistry Protocol
1.Connect the chemistry analyzer to AVE-772 correctly.
2.Select the corresponding chemistry analyzer in “Protocol”, click “Save” (upper left).

3.Select the corresponding chemistry analyzer in “Save”, click “Save” (lower left) to
accomplish adding.
4.3.2.3 Adding New Chemistry Protocol
1.Connect the chemistry analyzer to AVE-772 correctly.
2.Enter a new chemistry protocol in “New”, click “Add”.
3.Select the corresponding chemistry analyzer in “Protocol”, click “Test”. Choose any data of
the chemistry analyzer and transfer it to the test screen, then fill out the “Start”, “End”, “ID”,
“Y”(year), “M”(month), “D”(date). Click “Save” (upper left).
4.Select the corresponding chemistry analyzer in “Save”, click “Save” (bottom left) to
accomplish adding.
4.3.2.4 Adding Comparison Data
1.Select the corresponding item in “Items”.
2. According to the actual data of the chemistry analyzer, click the blank in “Actual data” and
input the information. Click the blank in “Report data” and input the corresponding information.
Click “Save” (upper right) and click “OK” in the pop-up window to accomplish adding.
3. Select other items and fulfill all the parameter settings referring to step 2.Click Quit to exit.

4.4 Patient Info. Setting


As shown in below, Select Patient Info. Setting in Management menu to pop up the three
secondary submenus Input Patient Info., Print Patient Info. and Show Patient Info.

Inp-ID (Inpatient ID): If a patient must be hospitalized, they will be assigned a number, which we
call it inpatient ID.
Dept. (Department): The department that the patient belongs to.
Subm. D (submitted date): At which day when the sample is submitted to the laboratory.
Subm. T (submitted time): The time when the sample is submitted to the laboratory.
Samp. D (sample date): At which day the sample is examined by the operator.
Samp. T (sample time): The time when the sample is examined by the operator.
Subm. (submitted by): The clinician who submit this sample.
Conduct. (conductivity): The conductivity value of this sample.
Verified (verified by): The operator who verifies this sample.

4.4.3 Show Patient Info.


As shown in Fig.4-13, select Show Patient Info. Users can set whether the Patient
information item be displayed in the Patient Information Area of the Microscopy and Verification
screen or not, if ticked it will be displayed, otherwise it will not be displayed. Operations of
Microscopy Info Table are as same as above and the items will be shown at Specimen ID Area.

General Setting
Hospital: Enter the hospital name. It will be displayed on the lab report.
Address: Enter the hospital address.
Remark: Enter notes, if necessary.
Microscopy Setting
Format: Select A4 or A5 format.
RT Print: Real time Print. Automatic print will be proceed immediately after negative
specimen being tested, if Yes is selected.
Print Image: Print images, if Yes is selected.
Image Number: Set the image quantity in a lab report. The maximum quantity is 6 for A4 and
3 for A5.
Print Neg: Whether to print the image of negative specimen.
Report Field: Set the microscopy result as semiquantitative result, such as /LPF, /HPF.
Print Item: Print the microscopy result as quantity or quality. Select both to show the
quantitative and qualitative results at the same time.
Morphology: Whether to print the morphological graphs. Morphological graphs will only be
shown if more than 3 RBCs identified.
Morph. Lower Limit: Set up the cutoff value that printing morphological images. It is
recommended to set the lower limit at 10.
Chemistry Setting
Report Format: Set chemistry report format. Select Separate Report to print chemistry
results only.
Format: Select A4 or A5 format for chemistry report.

Select Tools in Microscopy main interface, users can perform Test Mode Setting, Users
settings, Edit ID, Unusual, Gallery, Positive List, MagnGlass On/Off etc.

Delay: Set the delay time after the chamber is filled in. Tick the options, enter the delay time
and click Save to confirm. Click Quit to exit, system will initialize the default settings.
Close CH: Tick to close each channel. After Quit , channel will remain close, if the channel
needs to be opened, please cancel the tick.
4.8.3 Gallery
Select Gallery in the drop down menu as shown in Fig.4-20, the Gallery includes various
kinds of formed elements pictures, such as RBC(HP/LP), WBC(HP/LP) , Cast(HP/LP),
Crystal(HP/LP), Epithelium(HP/LP) and Fungus(HP/LP), for users’ reference only. It is able to
ask our manufacture or technicians to add new pictures into the Gallery as shown in below.

4.8.4 Positive List
This function is mainly used for Microscopy and Chemistry results automatic statistics
and printing and convenience for review.
The Microscopy Positive list displays those samples where any data of RBC, WBC, Cast,
Crystal and epithelium is positive. The Chemistry Positive list displays those sample where
any result of BLD, LEU and PRO is positive.

4.8.5 MagnGlass On
This function is used for open/close a magnifying glass, so that the picture could be
enlarging while performing verification. Click on the MagnGlass on, you can verify the picture
on the left of verification screen there the magnified images appead.

4.8.6 Test Mode Setting
Select Test Mode Setting in the drop down menu as shown in Fig.4-20, users can enter
the Test Mode Setting interface as shown in below. Click Save to confirm and Quit to exit.

Micro+Chem:Perform microscopy and chemistry test for each sample.
Chem:Perform only chemistry test for each sample.

5 SPECIMEN PROCESSING

5.1 Power On
1. Turn on the UPS (if any).
2. Turn on the power distribution board (if any).
3. Turn on the power switches of printer, monitor, etc.
4. Turn on the main power switch(Green switch on slide, if it’s on, the yellow LED inside of the
Green switch will be lighted).
5. Turn on the auto-sampler’s power switch(Green switch on slide, if it’s on, the yellow LED
inside of the Green switch will be lighted).
6. Press the Power button of the PC Module to enter Windows OS.

5.2 Login

Double click the shortcut icon on desktop to display the login window shown in
Fig.5-1

Type user name in the Name field, or select user name in drop-down menu.
Type user password in the Password field.
Click the Log in button to log into the main interface as shown in Fig.5-4.
If it’s a first time use, please set up parameters under the guidance of an authorized
technician. See Chapter 4 for details.

CAUTION: Ensure that the SQL Server in task bar is started before entering the
AVE software. If not, double click the icon as shown in Fig.5-2 to pop up the SQL Server
Manager interface as shown in Fig.5-3. Click the start / continue button to launch.

User Switch
After users login, if it’s needed to switch to other user, please operate as following steps:
Select Re-login in the drop down menu of Operation in system main menu to pop up login
interface as shown in Fig.5-1.Select or enter a new user name and password to login again.

5.3 Daily Quality Control (Daily QC)


Daily QC should be run to ensure reliable results before sample analysis. See Chapter 6
Quality Control for details.

5.4 Specimen Preparation

CAUTION:Use only fresh urine specimens, as defined in NCCLS (National


Committee on Clinical Laboratory Standards) GP16-A3 protocol, Good Laboratory Practices
and the Laboratory’s Procedure Manuals.

CAUTION:Collect urine in clean and sterilized containers. If a specimen is not


tested within an hour after collection, cover the container tightly and store at 2-8°C. Make sure
the specimen back to room temperature before testing.

CAUTION: Mixing the specimen well before testing.

CAUTION :Do NOT add disinfector or cleanser into the specimen.

CAUTION:Keep specimens out of direct sunlight.

CAUTION:Do NOT centrifuge the urine specimens.


5.4.1 Temperature
Specimens should be tested at room temperature. If the temperature is beyond the room
temperature range, make sure that all refrigerated specimens back to room temperature
before testing.

5.4.2 Specimen Volume


Specimen volume for AVE-772 Integrated Urinalysis Analyzer should be minimum 4ml
per test, if users want test the specimen more than one time, please prepare enough volume.

Biological WARNINGS: Specimen, QC material and waste are potential


biological hazards. User contacting with related items in the laboratory should comply with the
safety requirements and wear personal protective equipments(such as laboratory protective
clothing, gloves, etc.)
5.4.3 Sample Tube
Φ16 mm x 100 mm (L), glass or plastic round bottom tube.

5.4.4 Preparing Sample Racks

CAUTION:Please use the racks special for AVE-772.

CAUTION:Make sure the barcode labels face to barcode scanner.

1. Apply barcode labels to sample tubes, make sure that the barcode (not the barcode label)
is ½ inch (about 12.7 mm) below the top edge of the tube(Φ16 x 100 mm).
2. Prepare at least 4ml well-mixed native specimens into sample tube.
3. Put the tubes into the sample rack correctly and in order.
4. Launch the sample rack on the Ready Area of the sampler.
5. Mark the tubes as shown in Figure 5-5.

Barcodes accepted by the scanner are: Code 128, Code 39, Codabar and Interleaved 2 of 5 (I
2 of 5).

CAUTION: Make sure the barcode labels are properly oriented in the rack. The
sample tubes must be placed straight and resting in the middle of the grommets located in the
base of the rack.

5.5 Urine test strip preparation


AVE-772 analyzer can only use the urine test strip produced by AVE company.
Please place the test strips into the instrument following below steps before sample test.
Step 1: Check whether the quality guarantee period in test strip is within the valid date. If
it is not, do not use it.
Step 2: Open the front panel of the instrument, then open the the test strip can, then
open the test strip tank, put the test strips into the test strip tank according to the direction as
shown in figure 5-6.
Step 3: Put the strip can’s cap into the strip tank door’s dryer hole, and close the tank
door. Then close the front panel of the instrument.

CAUTION: Each time to put a new can of test strips, make sure to replace the can’s
cap into the strip tank door’s dryer hole.

5.6 Running Samples


There are two ways to enter the Microscopy interface as shown in Fig.5-3.
1.Select Microscopy in the drop down menu of Operation on the system main menu as
shown in Fig.5-7.
2.Click Microscopy shortcut icon in the shortcut keys area as shown in Fig.5-8.

As soon as Microscopy interface is entered, the system will perform auto initialization and
auto focusing. It will take approximate 30 seconds to finalize initialization.

Select Test Mode Setting in the Tools menu, users can enter the Test Mode Setting
interface as shown in below. It’s recommended to change the setting before test starts or the
current samples have finished.

Click the Start button on the Microscopy interface to achieve ready status, as shown in
Fig.5-11. Launch sample racks to the Ready Area of the sampler, microscopy testing will
processed automatically.

CAUTION: Do NOT move the sample racks in Ready Area during testing.

CAUTION: No operations should be performed during the self-checking procedure.

WARNING: Don’t place your hand below the sampling position to avoid injuries by the

sampling needle.

For each testing result, the system will presents the status for this sample accordingly, the
user can define the further processing according to the status. In general, the status are:
 NE (Not Examined): The microscopy examination of this sample is not finalized yet.
 NV (Not Verified): The microscopy result of this sample is positive, or the strip result did
not coincide with microscopy result, further verification is recommended.
 V (Verified): The microscopy result of this sample is negative, or the strip result coincide
with microscopy result, it’s not necessary to perform further verification.
 R (Reported): This sample was already verified and printed out.
 NV! (Not Verified!): The microscopy result of this sample is positive, or the strip result did
not coincide with microscopy result, it’s NECESSARY to perform further verification.
 EV (Expert Verify): This sample was already verified by online expert. (only available for
remote connection mode)
 NS (No Sample): The specified volumn of this sample is not attained, thus it was not
examined. The user need to check the volumn and test this sample again.

5.7 Patient Data Entry


The software provides a patient data entry function. Users can type in the patient
information at any time even the analyzer is testing specimens. If the analyzer is already
connected to LIS, the way to type in patient information could be faster and easier, operation
steps are as following: Type in Inp.ID, if the Inp.ID is already existed in LIS system, system will
show all the information of the Inp.ID from LIS when users click other input box.

5.8 Dealing with Special Microscopy


AVE-772 provides emergency priority testing and repeated testing function.
5.8.1 STAT Test
STAT Test must perform at the emergency position.
Operation steps are as following:
1. Prepare the emergency sample.
2. Put the sample at emergency position and push it forward, wait until system pop up the
Insert STAT interface as shown in below.
3. Select Urine Micro in the Micro box, STAT in the Type box, Micro in the STAT box, type
in the sample ID in the Sample ID box. Sample ID and Sam-ID must be the same. Users
can only type in one sample ID at one time. In principle, the start number of emergency
samples must be larger than all the specimens numbers. The barcode of the emergency
sample must be typed in the Barcode box manually. Click the Insert button to add the
corresponding information into the Special Sample field.
4. Pull the emergency position back after the sampling is finished.

5.8.2 Recheck
If it’s needed, users can perform Redo operation to repeat the Microcopy testing. Operation
steps are as following:
1.Prepare the emergency sample.
2.Put the sample at emergency position and push it forward, wait until system pop up the
Insert STAT interface as shown in Fig.5-13.
3. Select Urine Microscopy in Micro box, Redo in Type box and Urine Redo Item in Redo
box as shown in Fig.5-13.The numbering rules is that start with 3 and last three number is as
same as Sample ID’s last three number, for example, if the Sample ID is 0013, then the Redo
ID is 3013.

4.Click Insert button and the Redo number will show in Urine Special field .
5. Pull the emergency position back after the sampling is finished, the testing will be finished
automatically.
6.Complete the sample verification according to verification process. Click the Save button
and click OK to confirm as shown in Fig.5-14. If click OK to save and overwrite the former
result in Urine Special field, click Cancel that will not overwrite the former one and both
results will be kept.

5.9 Verification operations

5.9.1 Basic operations


It is recommended to verify the result manually when the sample status are NV and
NV!.
Enter the Urine Verification interface and refer to the following:
LP Verification
1.Enter the Urine Verification interface, select a sample, click anywhere of the LP Current
Cell Counting to switch to LP pictures, double click to enlarge the picture.
2.Observe whether there are results in the Cast, Epithelium column among the Current Cell
Counting field or not. If not, Users need to observe the LP pictures to make sure there is no
cast or epithelium in any of the LP pictures.
Note:Particle in LP Current Cell Counting means all small targets located in LP, do NOT
change!
3. Click Next to check in order. Verify and mark what you see actually in the images by right
click and select the right type/name in the pop up menu. If the item type/name is changed, the
result numbers in the corresponding column will also be changed; If the item type/name is
canceled, the corresponding result numbers will also be decreased.

4. Click Next to view all LP pictures of the sample in full-view and repeat step 2 and 3 until
finished.
HP Verification
5. Click anywhere of the HP Current Cell Counting to switch to HP pictures, double click to

6.Observe whether there are numbers in the RBC, WBC, Crystal, Bacterium, Yeast, etc,
column among the Current Cell Counting field or not. If not, Users need to observe the HP
pictures to make sure there is no RBC, WBC, Crystal, Bacterium, Fungus, etc, in any of the
HP pictures.
7. Click Next to check in order. Verify and mark what you see actually in the images by right
click and select the right type/name in the pop up menu. If the item type/name is changed, the
result numbers in the corresponding column will also be changed; If the item type/name is
canceled, the corresponding result numbers will also be decreased.

8. Click Next to view all HP pictures of the sample in full-view and repeat step 6 and 7 until
finished.
Save Result
9. Click Save button under the Current Cell Counting field to confirm the verification result of
selected sample, and the sample status is changed to V.
5.9.2 Advanced operations
After mastering the basic operations, the user can use advanced operations for
verification.
Enter the Urine Verification interface and refer to the following:
LP Verification
1. Same as Basic operations step 1-2.
2. Click Modify, then click the "+ " symbol on the target needed to cancel , to cancel the
target. When the target is cancelled, the "+ " symbol will disappear, and the result of current
frame will reduce.
3. Keep Modify is selected, then click the target name in the Current Cell Counting field,

then click the "+ " symbol on the target needed to modify, the target name will change to the
selected name, and the "+ " symbol will also change. The result of current frame will increase.
Repeat this operation to modify other targets in the same frame.
4. Click Next to view all LP pictures of the sample in full-view and repeat step 2 and 3 until
finished.
HP Verification
5. Same as Basic operations step 5-6.
6. Click Modify, then click the "+ " symbol on the target needed to cancel , to cancel the
target. When the target is cancelled, the "+ " symbol will disappear, and the result of current
frame will reduce.
7. Keep Modify is selected, then click the target name in the Current Cell Counting field,
then click the "+ " symbol on the target needed to modify, the target name will change to the
selected name, and the "+ " symbol will also change. The result of current frame will increase.
Repeat this operation to modify other targets in the same frame.
8. Click Next to view all HP pictures of the sample in full-view and repeat step 6 and 7 until
finished.
Save Result
9. Click Save button under the Current Cell Counting field to confirm the verification result of
selected sample, and the sample status is changed to V.
5.9.3 Batch operations
The user can use batch operations to modify a specific targets in one frame or all frames.
Enter the Urine Verification interface and refer to the following:
LP Verification
1. Same as Basic operations step 1-2.
2. Click Modify, then choose Save Type in the Clear Mark drop-down menu, then the batch
mofiy window will appear. Choose Current/All in Picture to proceed current frame /all frames,
in Type choose the target name to be cleared, in Change to choose Cancel, then click Save.

in current frame /all frames will become 0. Repeat this operation to clear other targets.

1. Enter the Microscopy or Verification interface, click the input box after Batch.
2. If the samples numbers are continuous, enter start and end ID. For example, to print from
No.2 to No.16, type 2-16.
3. If the samples ID are discontinuous, enter separate ID number respective ID. For example,
to print No.2, No.8, No.9 and No.18, type “2, 8, 9, 18,” ( always ends with “,”).

5.11 Power Off


After all the specimens have been tested, click the “Quit” icon in the shortcut keys area to
open the quit popup window. Checking “Quit and maintain” allows the instrument to wash the
pipelines with maintenance liquid automatically. Checking “Quit and auto turn off” allows the
instrument to shut down automatically the computer system after exit. Both ways can be
chosen randomly.

6 QUALITY CONTROL

6.1 Quality Control


The Quality Control program is used to collect QC data and display QC reports and
statistics. These functions are used to obtain, verify and monitor proper system operation.

6.1.1 Control Material


To transport, store and use of the controls, please refer to the user’s manuals
respectively.

6.1.2 Control Frequency


Controls should be run at least once every 24 hours or as specified in the Laboratory’s
QC Procedure.
Focus should be performed at least once every 24 hours, preferably before running the
controls.

6.1.3 Handling QC Material


Hold the QC material bottle upside down repeatedly and shake it gently until the cells at
the bottom are completely dispersed and mixed well under macroscopic observation.
Then pour the QC material into a clean test tube (about 2ml per test), and use it for
testing. The material may be maintained in the fridge at 2-8°C with a tinfoil paper sealing the
bottle after using.

6.1.4 QC Parameters Input


The QC parameters should be input or set before running QC. Different parameters input
screen can be accessed according to different QC types. Clicking “QC” allows to access the
QC selection screen and select “Urine QC” and “Chemistry QC” respectively as shown in
Fig.6-1. Select any one of them to access the corresponding QC parameter input screen.

A. Accuracy QC
Select “Accuracy QC” to enter the “Accuracy QC” screen. Input “Lot ID”, “Level”, “Start
Date” and the following parameters:
 Lot ID (for example: 20110407, 6-10 digits )
 Mean;
 SD (Standard Deviation);
 CV (Coefficient Variation).
Note:After completing parameter input in a row, click the “ Save” button, and then move on to

the next row for input.


Click “Delete” to delete the information selected. Click “Print” to preview the charts to be
printed. Click “Quit” to quit the screen.

B. Sensitivity QC
Select “Sensitivity QC”( Fig.6-1) to enter the “Sensitivity QC” screen shown in Fig.6-3.
Just input “Lot ID” and click Save on this screen. Click Delete to delete the information
selected. Click “Quit to quit the screen.

C. Chemistry QC
Select Chemistry QC in figure.6-1 to enter the screen as shown in below. According to the
QC material instruction, input corresponding batch number and item standard value, then
click “Save”.

6.1.5 Running QC
The QC can be run after the parameters being entered. Just like sample testing, QC is
run on the “Microscopy” screen.
Operations:
1 Place the QC material in tube rack and place the rack at ready area.
2 Choose “Urine Micro” in the “Micro” box; “QC” in the “Type” box; and “Micro QC” in the
“QC Type” box. The default sample ID starts from 2001.

CAUTION: The Micro ID should be same as Sample ID.


3 Choose Accuracy/Sensitivity in the “Name” box, then choose LOT number in the “LOT”
box; For Accuracy QC, check the corresponding concentration in the “Concentration”
box; For Sensitivity QC, only LOT is needed.
4 Click the “Insert” button and the system will run the QC automatically.

5 After QC testing is finished, the results will be displayed in Urine Special area, and
there will show QC normal/ QC warning / QC out of control messages in the status bar.

6 If QC out of control message appears, see 6.2 Remedies on Out-of-control for


further processing.
Explanation about QC status
 QC normal: The results located within ± 1SD will give a 'QC normal' status.
 Warning: The results located between ±2SD with ±1SD will give a 'warning' status.
 OOC (Out of control): The results located out of ±2SD will give an 'Out of Control' status.
Further steps is needed (see 6.2. Remedies on Out-of-control)

6.2 Remedies on Out-of-control

Select “Accuracy QC” to enter the “Accuracy QC” screen shown in Fig.6-2. In the “Lot ID”
box, enter the ID or check the ID through the drop-down menu. After validation, corresponding
information will appear in the “Parameter” field on the screen, and QC Chart will also appear
in the Accuracy QC Chart.
The default statistic data is the last QC result of the day. You can also double click the QC
results of the day and choose one as the statistic data as needed.

6.3.2 Sensitivity Review


Select “Sensitivity QC” to enter the “Sensitivity QC” screen shown in Fig.6-8.
In the “Lot ID” box, enter the ID or check the ID through the drop-down menu. After
validation, corresponding information will appear in the “Parameter” field on the screen.
Double-click the corresponding data on the page to pop up QC data selection prompt box
where the “mean” and “ID” can be set. Click “Save” for save. Click “Quit” to leave the page.

The default statistic data is the last QC result of the day. You can also double click the QC
results of the day and choose one as the statistic data as needed.

6.3.3 Chemistry Review


Select Chemistry QC to enter the QC result list as shown in below.

In the ” batch ID” of the figure, input or click drop-down menu to choose the QC batch
number that you want to consult result. After you have confirm it, corresponding QC result
value will appear in QC setting value information, and in below QC curve graph will appear
corresponding QC statistical chart.
1.Choose different batch ID can look though corresponding batch ID’s QC standard value
and corresponding QC data, and displays of already exited QC data’s corresponding diagram.
2.Choose different item can see different item’ S QC result curve and print it.
3.If there are more than two pieces of QC data one day, choose one of them as that day’s
QC data as you can see in figure 6-11.

Notes: If the “item name” data displays red color, it indicates that the QC is out of control that
day. To say more specific, it indicates that corresponding date‘s QC data script is red, too.
Under the condition of many QC data appears some day, choose the last piece of data as a
benchmark if there isn’t display that a certain piece of data should be viewed as a benchmark.
Otherwise, treat the last piece of data as a benchmark. “deadline”: It means that the final QC
data of present QC batch ID.( It displays only when QC data has already exist).
“Print”: This botton can display the Chemistry QC statistics of present batch ID as it
shown in below:

7 ADJUSTMENT

7.1 Summary
The AVE-772 Analyzer’s adjustment include: Microscope Focus and CCD Image
Parameters adjustment, Pump Calibration and Blank Calibration etc. Enter the main screen
and choose “Management” to open the “Adjustment” submenu (shown in Fig.7-1).

7.2 Microscope and Image Adjustment


The Microscope is an indispensable instrument in the medical examination. Its
view image clarity directly influences the microscopic results.
Since the AVE-772 is based on an automatic microscope as the testing platform, the
quality of the results mainly relies on the adjustment of the microscope in the AVE-772
adjustment procedure. The adjustment procedure consists of the focus adjustment and the
CCD parameters adjustment.
Make sure to have the instrument turned on at least for 30 minutes before
performing the focus adjustment in order to have stabilized the lamp of the
microscope!
While mark position and focus are not satisfy with the standard gallery, focus and
mark position calibration are recommended.
While brightness and RGB are not satisfy with the standard gallery, image
adjustment is recommended.

7.2.1 Preparations for microscope focus adjustment


1. Prepare a suspension of RBCs (like a visual hematuria sample) with a standard tube, and
divide it into three tubes, then place the tubes in a rack. You may use the high level of the
Quality control for the microscope focus adjustment.
2. Locate the racks holding the tubes with the suspension of RBCs to the ready area of the
sampling module.
3. Select “CH1” then click “MechRS” button. There isn’t any fixed adjustment sequence for
the channels, however it is suggested to start from Channel 1, then Channel 2 and finally
Channel 3.
4. Click the “Samp All” button and the instrument will transfer the rack from the ready area to
the sampling area.
5. Wait for a about 2 minutes for the RBCs suspension to be fully aspirated into the Counting
Chamber, then the preparatory work is done and the next steps can be performed.

7.2.2 Adjust The Square Mark Position Of Microscope


The Microscope’s mark position, the position of the Counting Chamber square mark in the
microscopic view, determines the initial position of capturing images under different view fields
during microscopy. (The square mark is engraved at the bottom of each counting chamber).
Since the microscope has two view fields, a high power field (HP) and a low power field (LP),
the adjustment for the square mark position needs to be done for both.
The reference steps of each channel are in the following table:

Each Counting Chamber will provide these parameters, when the Counting Chamber is
replaced, click “Management” - “Adjustment”, then click Replace chamber, and in the popup
window, enter the new parameters, click Save , then click Quit.

After the new parameters are saved, the instrument will automatically adjust, when it is
done, check the square mark and focus of each channel; If the square mark and focus are not
in the best condition, please adjust these parameters refer to the following steps.
1. Choose CH1, and click LP Reset;
2. Check the , and enter a certain step in “Step”(for example 50), click “-” or
“+” to adjust the steps to 550;
3. Check the , and enter a certain step in “Step”(for example 50), click “-” or “+”
to adjust the steps to 400;
4. Check the , and enter 50 steps in “Step”, click “+” to move the chamber until
the square mark appears;
5. Check the and , and enter 5-10 steps in “Step”, click “-” or “+”
to move the chamber until the square mark at the upper left in the view field, then click Save
LP Mark;

6. Click HP Reset, change to HP, then check the and , and
enter 5-10 steps in “Step”, click “-” or “+” to move the chamber until the square mark at the
upper left in the view field, then click Save HP Mark;

7.2.3 Focus Adjustment


When the adjustment of mark position is done, focus adjustment is needed.
Procedure of Focus Adjustment
1. Check the Up-down value of CH1 and just remember this value. Then tap HP Reset
button to switch to HP lens.

2. In the HP lens interface, check whether the Up-down value is same with that of LP lens. If
the two values are not equal, please go to Step3. If the two values are equal, please just

go to Step4.

3. Make Up-down option tapped, and input proper value in the text box which located behind
Step:. Then tap + or – button to adjust the Up-down value same with that of LP lens. Tap
Save HP Mark button to save the change.

4. Switch to CH2 and CH3 respectively, and repeat step 1-3 to finalize the settings.
5. Reture to CH1 and tap LP Reset button to switch to LP lens, and click Auto Focus and
then the microscope will perform focus calibrating automatically.

6. A pop-up window will show a calibrated parameter after auto focusing. Click Save to save
the calibration.

In general, the three channels lie nearly in the same plane, which means that the
focus of the other two channels will be adjusted according to the alteration of
channel 1. Just to be sure, it’s recommended to check the focus of the two
channels, and if it’s necessary, the user need to adjust the focus manually by
following up the next steps.
7. Select “CH2” in the “Channel” field on the upper-left of the adjustment screen, then the
lens will switch to channel 2.
8. Check the Up-down value of CH2 and just remember the value. Then tap HP Reset
button to switch to HP lens.

9. In the HP lens interface, and input proper value in the text box. Then tap + or – button to
adjust the focus to a sharp condition. Remember the Up-down value and DO NOT SAVE
THIS VALUE for the moment. Tap LP Reset button to switch to LP lens interface.

10. In the LP lens interface, make Up-down option tapped, and input proper value in the text
box which located behind Step:. Then tap + or – button to adjust the Up-down value
same with that has the best focus of HP lens. Tap Save LP Mark button to save the
change.

11. Switch to CH3 and repeat step 8-10 to adjust the focus of CH3.

7.2.4 Image Adjustment


Click “Image Setting” to enter the “Image Adjust” window (Fig.7-13). The CCD
Background Exposure and White Balance Values can be adjusted on the screen as well as
the settings for the standard gallery.

The reference for the parameters are:
Gray scale:12;
Gain: 15-25;
Red/Green/Blue: The difference between Picture value and Reference value should less
than 10.
To adjust the image settings, refer to the following steps:
1. Check “HP” in the “Precise Adjust” field to enter the HP field.
2. Click “AE On” to wait for automatic exposure.
3. Input the fine-tune value and gain value in the “Bright/Gray scale” in the “Precise
Adjust” field according to the image and the reference value.
4. Click the “L+” or “L-” button in the “Brightness” field to make the exposure value of the
image consistent with the reference value (values of +/- 5 are considered as
consistent). Click “Save Exposure” for saving the settings.
5. Click “AWB On” to wait for automatic white balance adjustment. Click the “R+/R-”,
“G+/G-”, and “B+/B-” button respectively to allow the image value to be as close as
possible to the reference values (values of +/- 10 are considered as consistent). Click
“Save Gain”.
6. When the exposure values and white balance values under HP are adjusted, click
“Save All”.

7. Check “LP” in the “Precise Adjust” field to enter the LP filed; the operations are the
same as HP. Click “Save All” to save all the adjusted parameters.

7.3 Pump Calibration


The pump calibration should only be modified by authorized technicians!
Pump calibration is recommended at following conditions:
1. Bubbles occur in counting chamber or the amount of reagent is abnormal;
2. After replacement of Rinse/Waste Bath or tubes (including valve tube, pump tube
etc.);
3. Monthly maintenance.

7.3.1 Operations
The pump calibration can be performed at the lower right of the adjustment screen.

“Wash-pump Calib”: By selecting “WPC” (1#, 2# and 3# respectively), a dialog box will
open (shown in Fig.7-15). Click “OK” to continue, the new value will be saved automatically.
The reference value for Wash-pump Calib is 3.5-5s, repeat wash pump calibration, till the
value reaches the reference range.

“Sample-pump Calib”:By selecting “SPC” (1#, 2# and 3# respectively), a dialog box will
open (shown in Fig.7-16). Click “OK” to continue, the new value will be saved automatically.
The reference value for Sample-pump Calib is 4-5.5s, repeat sample pump calibration, till the
value reaches the reference range.

7.3.2 Caution
If the wash/sample calibration value is abnormal in the process of Pump Calibration,
please check whether the pipeline is clogged.

7.4 Blank Calibration


The operation is mainly used for determing dirty spots number in the Counting Chamber.
Operations can be done on the right side of the “Calibration” screen, in the “Blank Calib”
field.
1. Click “Blank” button. The instrument will run the Blank Calibration automatically.
2. A prompt will appear at the end of the blank calibration.

3. Click “OK” to save dirty spots number, Click “Cancel” not to save. Please note that
those will be susbtracted from each result randomly. Therefore we recommend to use the
blank calibration to determine, if the the channels are clean.

CAUTION:If the dirty spot exceeds 5, please perform special wash, then perform
Blank Calibration again; If the dirty spot still exceeds 5, it is recommended to replace the
counting chamber.

8.1 Urine Data

8.1.1 QC Inquiry
Click “Data”-“Urine Data”-“QC Inquiry” to enter the QC Inquiry Screen.
The user can inquire and backup previous QC records.

keys area.

1. Input patient’s various inquiry items in the “Inquiry Condition” field, click” Inquiry”
button at the lower right of the field. System will inquire automatically according to what you

2. Input one or more inquiry items as shown in Fig.8-3, click “ Inquiry” button. The
inquired results will appear in the “Inquiry Result” field.
b. By sample ID
Input the date as YYYY/MM/DD into the textbox of “Sample ID” to display the patient’s
information of the day in the “Inquiry Result” field. For example, input “200606060006” to
display the records of sample No.6 on 2006-06-06.
c. Inquire all
Select a time range, click the“ Inquiry” button. All the records of the patient will appear in
the “Inquiry Result” field.
Print records
Click Preview button to preview a selected sample’s report, in the perview window, click
Print to print the report.
8.1.3 Statistics
Enter the main screen, select “Data” - “Urine Data” -“Statistics” to enter the urine

statistics screen. Workload, urine result and chemistry result can be counted here. The type of
the statistics varies with the static option. Both test results of the day and those without
patient information cannot be counted.

Select the microscopy time, then you can perform the verification, mofify patient
information and print report.
Every time the operator makes verification, the instrument will automatically record the
result, and generates a history record; The operator can review all the informations of each
sample, including verified date, time and verifier in Audit Trail area.

8.2 Tube Records


Tube Records is mainly used to determine whether the actual sample tube number is
consistent with the automatic one identified by the system. Click “Data”-“Tube
Records”-“UrineTube Records” to review today’s finished sample tube and rack records.
Move the cursor on the tube icon, it will show the sample ID, barcode, rack number and
tube number for further verificaiton.

9.2 Daily Maintenance


This procedure should be performed at least once daily.
9.2.1 Cleaning Instrument Surfaces
Tools required: Disinfectant/cleanser, gauze, and protective gloves.
Procedure: Power off the instrument. Clean the instrument using gauze moistened in
disinfectant/cleanser. Wipe again using distilled water and then make it dry.
9.2.2 QC
Refer to 6 QUALITY CONTROL for details.
9.2.3 Cleaning Counting Chamber Surfaces
Tools required: 95% absolute alcohol, absorbent cotton swabs.
Procedure: Open the front cover, using a clean cotton swab moistened with 95% absolute
alcohol; wipe the Counting Chamber surfaces gently.

CAUTION: Do not wipe back and forth, wipe in one direction to avoid scratches on the
counting chamber surface!
9.2.4 Cleaning Waste Tank

WARNING: Wear protective gloves to prevent exposure to pathogens. Discard


contaminated materials according to applicable regulations.
Tools required: Disinfectant (such as solid dialdehyde, peroxide acetic acid, hydrogen
peroxide, chlorine dioxide, etc.), distill water, gauze, protective gloves.
Procedure: Take out the waste tank from the analyzer, and clean it with the disinfectant.
Then use distill water to clean it again and let it dry and put it back.

9.3 Weekly Maintenance


9.3.1 Cleaning the Sampler
To make sure the sampler running properly, timely cleaning the sampler is highly
recommended when there are liquid leaked on the sampler.

CAUTION: Immediately power off the sampler and clean it while a whole rack samples
are pouring onto the sampler.
Tools required: Disinfectant/detergent, gauze, and protective gloves.
Procedure:
1 Power off the instrument. Ensure that the maintenance is performed under power cut-off
condition.

2 Moisten gauze with disinfectant/cleanser and wipe the sampler to remove any deposits.
3 Check the flexibility of pawls. Moisten cotton swabs with disinfectant/cleanser to clean
pawls, remove any deposits.
4 Wipe again using distilled water.
5 Power on the instrument until the sampler is totally dry.

9.3.2 Checking microscope focus (Focus Adjustment)


Refer to 7.2.3 Focus Adjustment for details.
9.3.3 Cleaning the Strip Conveyor

WARNING: Wear protective gloves to prevent exposure to pathogens. Discard


contaminated materials according to applicable regulations.
Tools required: Disinfectant/detergent, cotton swabs, brush and protective gloves.
Procedure:
1 Power off the instrument. Ensure that the maintenance is performed under power cut-off
condition.
2 Remove the right panel of the instrument, and take out the Strip Conveyor’s Plate.
3 Use brush with disinfectant/cleanser and clean the plate to remove any deposits.
4 Clean it again using distilled water and let it dry.
5 Put the plate back and close the right panel of the instrument.

9.3.4 Cleaning the Pipette Slot

WARNING: Wear protective gloves to prevent exposure to pathogens. Discard


contaminated materials according to applicable regulations.
Tools required: Disinfectant/detergent, cotton swabs, brush and protective gloves.
Procedure:
1 Power off the instrument. Ensure that the maintenance is performed under power cut-off
condition.
2 Open the front panel of the instrument.
3 Use brush with disinfectant/cleanser and clean the Pipette Slot to remove any deposits.
4 Clean it again using distilled water.
5 Close the front panel of the instrument.

9.4 Monthly Maintenance


9.4.1 Checking pinch valves and moving pinch location in case of adhesion
Since the pinch pipelines are in the clamped state while the valves are not energized, monthly
perform rebound maintenance to prevent adhesion is recommended.
Procedure1: Take out the pinch from the valve and twist it gently by fingers to retrieve its’
elasticity.

Procedure2: Move the pinch pipelines upper or down 2mm per month to prevent adhesion or
broken after long time clamped.
Contact the service for replacement if you need a new pinch pipeline.
9.4.2 Cleaning the Waste Pool
In order to prevent pipeline block and invalidation of liquid detection, the waste pool is suggested
to perform cleaning every month. This includes cleaning the inner wall, cleanser nozzle upper and
waste nozzle at bottom.
Tools required: Deionized water, cotton swabs and protective gloves.
Procedure:
1 Make sure the analyzer is in shutdown state.
2 Open the front cover to access the waste pool. Manually lift the sampling needle out.
3 Using a tube brush moistened with deionized water, remove salt deposits present on the
waste pool. Clean the inner wall.
4 Using a cotton swab moistened with deionized water to swab cleanser nozzle upper and
waste nozzle at bottom.
5 Move the sampling needle back into the waste pool, close the front cover.
6 Power on the instrument and login the software. Click Wash All 1-2 times in Adjustment
interface to wash all the channels.

CAUTION: Do not insert the cotton swab inside the waster pool; cotton particles may
clog the tubing connectors.
9.4.3 Pump Calibration
Refer to 7.3 Pump Calibration for details.
9.4.4 Cleaning Sampling needle
Tools required: 95% absolute alcohol, cotton swabs and protective gloves
Procedure:
1 Make sure the analyzer is in shutdown state.
2 Open the front cover to access the waste pool. Manually lift the sampling needle out.
3 Using a cotton swab moistened with absolute alcohol to swab the interface of needle,
4 Move the sampling needle back into the waste pool, close the front cover.
9.4.5 Cleaning Photoelectric Sensors on auto-sampler
Cleaning photoelectric sensors on auto-sampler is recommended to perform every month.
Tools required: 95% absolute alcohol, cotton swabs and protective gloves
Procedure:
1 Power off the instrument, make sure the operation is in shutdown state.
2 Using a cotton swab moistened with absolute alcohol to swab the surface of photoelectric
sensors
3 Swabs dry them by using a dry cotton swab.

9.5 Semi-Annual Maintenance


9.5.1 Check Microscopic Image
Check the background of CCD images by comparing it with the standard images. If there is any
deviation, refer to 7.2.4 Image Adjustment for further adjustment.
9.5.2 Blank Calibration
If some spots impossible to clean out are found in the counting chamber, please refer to 7.4 Blank
Calibration for blank calibration.
9.5.3 Pump tube replacing
Since the pump tube is a kind of wearing parts, it should be checked regularly. Normally pump
tube should be replaced every 6 months. If any aging, deformation, inelasticity or broken is
happen, please replace it immediately.
The specification of pump tube is Φ3.2*Φ6.4*92mm, make sure using the same pump tube for
replacement.

WARNING: Wear the personal protective equipment such as protective clothing

and glove to protect you from the waste remaining in the pump tube!

9.6 As Needed Maintenance


9.6.1 Checking the Mark Position of Microscope
Refer to 7.2.2 Adjust The Square Mark Position Of Microscope for details.
9.6.2 Replacing the microscope lamp
Since the microscope lamp is a kind of wearing parts, its life may affect by environment (voltage,
temperature, humidity). It should be replaced according to the actual working situation.
If its lightness become poor or the background of images becomes dark, thus the recognition is
affected by this, please replace it immediately.
Specification of Microscope lamp: 2V,20W

CAUTION: Replace the new lamp of same specification!

CAUTION: Make sure the replacement is done in power off state! To prevent burning
by the lampshade, make sure it is already cooled before replacement!
Replacement procedure:
1 Power off the instrument; make sure the lampshade is already cooled. Open the front cover to
find the fix screw located on the base.

CAUTION: Socket oxidation may cause contact problem. If the lamp still does not

9.6.3 Replacing the Counting Chamber


Maintaining the Counting chamber
The counting chamber is a kind of wearing parts, its life may affect by the frequency of use and
maintenance condition. If there are spots inside it or it was blocked, following the next procedure.
1 Login the software and go to Adjustment interface, select the channel need to be cleaned,
click Wash+ to see whether it can be cleaned, if not, please refer to step2.
2 Remove the counting chamber. Connect a injector to one end via chamber pipeline, soak
another end into a container filled with cleanser (diluted hydrochloric acid, disinfectant,
detergent solution, for example). Repeatedly perform aspiration until the dirty are discharged.
3 If above steps do not work, please replace the counting chamber.
Replacing the Counting Chamber
When the counting chamber is broken or there are too many spots or scratch on it, this may affect
the recognition, please replace the counting chamber. Make sure the replacement is done in
power off state.
Replacement procedure:
1 Compare the serial number printed on the side of counting chamber with that on the insert
card to check if they are matched. If so, remove the hose connected with the two ends.
2 Open the front panel of instrument.

3 Manually switch the LP lens aside, lift the two tabletting up to pull out the bracket together with
counting chamber; take off the chamber pipeline connected at both sides, then take out the
bracket entirely.

5 Replace the new counting chamber. Connect the chamber pipelines onto both sides of three
channels respectively, the channels can be identified by its color ring.

CAUTION: The installation trends should follow this: the face with eminence is upper
surface and the other one is on the bottom; the side with serial number printed on should
be toward inside. The chamber pipeline should be plugged to 1/2 of iron tube (as shown
below).

6 Install the new counting chamber into bracket; press tightly the screws to fix it.

CAUTION: The counting chamber should be installed as much as possible into


upper right corner of bracket. The chamber tabletting should press to the chamber edge
from 1-2 mm.

7 Push the bracket back under the tabletting. Make sure the positioning balls just drop into the
fixed holes.
8 Close the front panel.
9 Input the parameters of new counting chamber.
After replacing the counting chamber, the parameters should be input in the software. In the main

interface, go to Management-Adjustment, click Replace Chamber button to pop-out the


Replace Chamber interface, in which input the parameters of new counting chamber. The user can
find the related parameters on the insert card. Click Save button to make the inputting take effect.
Click Quit button to finish the inputting.

10 Focus confirmation
After inputting the parameters, the microscope will respond the inputting to find its location
automatically. After that, it’s necessary to check focus and mark under LP & HP of three channels.
If the condition is not satisfied, refer to 7.2 Microscope and Image Adjustment for further calibration.
Normally the user just needs to perform fine turn to get better result.
9.6.4 Dredging Sampling needle
If the sampling needle is blocked, following the below steps to dredge the sampling needle.
1 Make sure the analyzer is in shutdown state.
2 Open the front cover to access the waste pool. Manually lift the sampling needle out.
3 Remove the pipelines from the top of needle, use the pricker thread through from top of the
needle and exit out from the end, clean the dirty stick on the pricker;
4 Repeat step3 several times until there is no dirty stick on.
5 Connect the pipeline back to the needle. Move the needle back to the waste pool and close
the front panel.
6 Restart the instrument and login the software, go to Adjustment interface and click Wash All
button several times to perform channels washing.

9.7 Data Backup/Restore


The instrument may run slower after a certain period of time due to the accumulation of
patient data. To make data backup periodically is recommended.
Go to Management-Backup to entry Backup interface as shown below.

9.7.1Data Backup
Auto Mode
To enable auto backup the examination results and system parameter to the specified path
monthly, you just need to select Monthly automatic.
Manual Mode
The examination results and system parameter can also be export manually. Follow the steps
below.
1 Click Browse button to choose the specified backup path.
2 Select tick box in Urine Data or Parameter area to backup data need to export.
3 Click Export button to export the selected item to the specified path.
4 Click Backup button at Backup Setting area to make a whole backup of the database.
Meanwhile a record will be recorded in Restore Setting list.

CAUTION: To prevent losing data, do not perform any other operation during the
process of backup.
9.7.2 Data Restore

CAUTION: Only properly trained and authorized service personnel can operate
restoring. Make sure that your operation are under the instruction of authorized service!
Operation procedure:
1 Click Browse button to choose the specified restore path.
2 Click File button to select files (bak. format) need to be restored in specified path.
3 Click Open button to start restoring your selected data.
4 Select tick box in Urine Data or Parameter area to restore data need to import.
5 Click Import button to import the selected item from the specified path.

9.8.1 Shutting Down the Instrument
When it is necessary to restart the instrument, shut down the instrument and the
computers in the following order:
1 Click Quit button on the Microscopy screen.
2 Select Quit and auto turn off, unselect Quit and maintain, then click Quit button.
3 After the device shutting down, turn off the power of sampler.
4 Turn off the switch at the left of instrument.
9.8.2 Restarting the Instrument

3 Press on the power switch on the front panel to start the system.

10 APPENDIX

10.1 Barcode Match


Note: Only use the barcode match function when working with urine analyzers!
The user must strictly comply with the rules of use in the testing process to avoid the
wrong ID and the condition that chemical and microscopic results do not match. The
instrument will match the results with barcode as top priority.
When the instrument fail to read barcode or the chemical and microscopic results’s
barcode do not match, use the barcode match function.

Operations:
1. Choose the Chemistry or Microscopy ID that needs to be modified.
2. Click “Tools”- “Barcode Match” in Microscopy interface.
3. Modify the barcode ID in “Microscopy ID Modify”/“Chemistry ID Modify” and click “Modify”,
the system automatically prompts the sample ID consistent with the barcode. Click “Yes”,
the system prompts the barcode match with the sample which has the same barcode ID.
Click “OK” to achieve matching.
4. The original error sample ID will be kept in the Verification List after barcode modification
is completed. If it is not needed, delete it in right-click popup window.
