NEST PCR AND OLA PCR

NEST PCR
VAIRATION NEST PCR
Nested PCR is a modification of PCR that was designed to improve sensitivity
and specificity. Nested PCR involves the use of two primer sets and two
successive PCR reactions
PRINCIPLE OF NEST PCR
Nested PCR usually involves two
sequential amplification reactions,
each of which uses a different pair of
primers.The product of the first
amplification reaction is used as the
template for the second PCR, which is
primed by oligonucleotides that are
placed internal to the first primer pair.

ADVANTAGES OF NEST PCR
1-It is beneficial in studies such as phylogenetic analysis and genetic polymorphism.

2-The main advantage of the present method is that it gives 100% accuracy,
specificity and sensitivity. Disadvantage

DISADVANTAGES OF NEST PCR

1-The method is time-consuming.
2-Required more reagents such as an extra set of primer and one extra round of
agarose gel electrophoresis. This means the method is quite costly.

3-The chance of contamination is also higher.

OLA PCR
OLA PCR

(Oligonucleotide Ligation Assay) OLA PCR technology is based on the

polymerase chain reaction (PCR) technology, but this technology is used to
analyze PCR products once amplification is completed and after the
amplification is prevented from being replicated.

analyze diseases that may affect medical tests

Diagnosis of genetic diseases:

OLA PCR technology can be used to diagnose all diseases.

BENEFITS OF USING OLA PCR IN

SCIENTIFIC RESEARCH

1-Accurate diagnosis
2-Reducing repetitive analysis
3-Resistance production
ADVANTAGES OF OLA PCR DISADVANTAGES OF OLA PCR

Technical complexity:
