ACTIVITY 7 – PCR Part 3: Real-time Assays and Troubleshooting March 21-27,2022

Objectives:
1. To complete an Assay Worksheets for SARS-CoV-2 (2019-nCoV) Real-time RT-PCR based on the
Instructions for Use (IFU) of Sansure Biotech kit, one of the most commonly used kits in Molecular
Biology and Diagnostics Laboratories in Cebu City.
2. To demonstrate knowledge on interpretation of Real-time RT-PCR assay results.
3. To discuss troubleshooting in Real-time RT-PCR assays.
4. To familiarize with the procedures similar in Laboratory SOPs or Work Instructions in Molecular
Biology and Diagnostics Laboratories.
5. To identify the laboratory rooms where major steps of Real-time RT-PCR assays are performed.

Note: Copy of the IFU and the Assay Worksheet for SARS-CoV-2 (2019-nCoV) rRT-PCR will be sent to your
Group Leader.

Instructions:

1. Watch and understand the laboratory lecture on Real-time Assays and Troubleshooting.

2. Utilize the remaining time of your class schedule today to perform the instructions below.

3. Fill out the required data in the five (5) tables of the Activity 7 - Assay Worksheet. If you need
more detailed information, refer to the IFU of Novel Coronavirus (2019-nCoV) Nucleic Acid
Diagnostic Kit (Sansure Biotech).

4. Submit the completed Assay Worksheet within the first hour of the next laboratory class.
