Endo Fullerene V I Ung Thư Và Covid 19

Endo Fullerene là một hoạt chất được hình thành từ các phân tử cacbon, chúng có cấu trúc
phân tử độc đáo mà không một loại chất nào có được. Từ năm 1985 khi 3 nhà khoa học đầu
tiên là Kroto, Curl và Smalley phát hiện ra mạng cacbon tinh thể thứ ba sau kim cương và
graphite. Đến năm 1990, phản ứng chức năng đầu tiên được báo cáo ngay sau khi các
fullerenes được tổng hợp với lượng cực nhỏ..
Từ đó đến nay đã có rất nhiều các nhà khoa học trên thế giới nghiên cứu để tìm cách điều chế,
tổng hợp chứng minh hoạt chất này có các tác dụng vô cùng to lớn đối cho sự tiến bộ của loài
người như trong y học, công nghiệp chế tạo và ứng dụng vào nhiều nền khoa học khác nhau.
Tuy nhiên các nghiên cứu này chủ yếu chỉ mới thành công trong phòng thí nghiệm, tạo ra vơi số
lượng cực nhỏ chưa đáp ứng được nhu cầu của cuộc sống. Nhận ra ý nghĩa và giá trị to lớn của
Endo Fullerene nên năm 1996 nhà khoa học Trịnh Đình Năng đã tiến hành nghiên cứu, trong
suốt 16 năm ông đã thực hiện hơn 3000 thí nghiệm. Cuối cùng, ông đã tìm ra công thức, từ đó
xây dựng hệ thống máy móc phục vụ sản xuất với quy mô công nghiệp.
Ở bài viết này chúng tôi chỉ tập trung phân tích về cấu tạo cũng như các tính chất của Endo
Fulleren Trịnh Năng để ứng dụng trong y học, đặc biệt là khả năng chống ung thư, phòng và
điều trị các bệnh về virus nhờ vào cấu trúc phân tử đa chiều. Hiện nay Endo Fullerene đã được
tổng hợp, dựa vào cấu trúc cacbon độc đáo sử dụng trong điều trị ung thư và điều trị Covid-19
1/ Fullerene là gì?
Fullerene là một dạng thụ hình của cacbon mà phân tử của nó bao gồm các nguyên tử cacbon
được nối với nhau bằng liên kết đơn và đôi để tạo thành một lưới khép kín hoặc khép kín một
phần, được ký hiệu là C n (trong đó n là số nguyên tử cacbon. Tuy nhiên, đối với một số giá trị
của n có thể có nhiều hơn một đồng phân) , với các vòng hợp nhất của năm đến bảy nguyên
tử. Phân tử có thể là hình cầu rỗng, hình elipsoid, dạng ống hoặc nhiều hình dạng và kích thước
khác nhau [1]
Liên kết Fulleren là một liên kết cộng hóa trị sp2 giữa các nguyên tử cacbon với các cấu trúc lục
giác và ngũ giác, có cấu hình e của cacbon như sau: 1s2 2s2 2p2
2/ Endo fullerene là gì?
- Là những fullerenes như trên nhưng có các nguyên tử , ion hoặc cụm bổ sung liên kết với các
nguyên tử cacbon hoặc được bao bọc bên trong các hình cầu của chúng. Những fulleren như
vậy được gọi là "endohedral". ("endon" - bên trong) và ("hedra" - mặt ngoài của một hình hình
học).
2.1 Cấu trúc của Endo fullerenes
Nguyên tắc: cần từ 36 nguyên tử cacbon và có ít nhất 75% nguyên tử cacbon mới hình thành
cấu trúc hình cầu, phần còn lại là kim loại hoặc phi kim loại, được phản ứng trong môi trường
không có oxy ở nhiệt độ từ 7000 đến 10000 độ C để tạo thành các metallofulleren nội tiếp( thể
hiện một cấu trúc nguyên tử trong một phân tử).[2][3]
- Trong công thức hóa học, các chất nội mặt được viết đầu tiên, tiếp đến là ký hiệu “@” và sau
đó là lồng carbon (ví dụ N @ C 60 biểu thị một phân tử fullerene nội tiếp, trong đó nguyên tử
nitơ được bao bọc bên trong lồng C 60, một ví dụ khác: K2(K@C59B), nghĩa là "một lồng
fullerene 60 nguyên tử với một nguyên tử được thay thế cho một cacbon, một kali duy nhất bị
mắc kẹt bên trong, và hai nguyên tử kali dính vào bên ngoài )
- Do sự khác biệt cơ bản trong các đặc tính cấu trúc và điện tử của chúng cũng như các cách
tiếp cận tổng hợp, các fulleren nội mặt có thể được chia thành hai lớp chính:
+ Metallofulleren (EMFs): là các fulleren bao bọc một, hai hoặc ba nguyên tử kim loại, cũng như
các cụm phức tạp bao gồm tối đa bốn nguyên tử kim loại và tối đa ba nguyên tử phi kim loại có
độ âm điện (N, O, C, S).
+ Fulleren nội tiếp phi kim loại (nMEFs): bao gồm các nguyên tử đơn lẻ (khí quý, nitơ, phốt pho)
cũng như đơn phân tử (H 2 , H 2 O, HF, v.v.)
- Trong trường hợp, hầu hết các nguyên tử là nghịch từ, nguyên tử kim loại và á kim cùng liên
kết ở ngoài cùng hình cầu, và có một nguyên tử nitơ hoặc một nguyên tử kim loại bên trong
hình cầu, gọi là Endo metallofullerene.
Cấu trúc hóa học của các endo fulleren không chỉ đơn giản là các C60, C70 mà còn bao gồm các
lồng lớn hơn nhiều, trong đó C60 và C70 chiếm ưu thế hơn cả và C60 có cấu trúc đối xứng là
bền vững nhất.
2.2 Tính chất vật lý của Endo fullerene
Thể hiện tính bán dẫn( hiệu ứng trường từ):
Khi kết hợp 2 lớp p-n với nhau sẽ dẫn tới việc trao đổi điện tích tại lớp tiếp xúc đó: các điện tử n
sẽ chuyển sang lớp p và ngược lại các lỗ trống lớp p sẽ chuyển sang lớp n do quá trình trung hòa
về điện, từ đó tạo ra một hiệu ứng từ trường.
- Kim loại liên kết trên khối cầu cacbon luôn có vùng dẫn và vùng hóa trị phủ lên nhau, do đó
luôn luôn có điện tử trên vùng dẫn vì thế mà chúng luôn luôn dẫn điện
- Ở 0 độ tuyệt đối ( vùng cấm) các điện tử nằm ở vùng hóa trị thì chất bán dẫn không dẫn điện
- Khi nhiệt độ càng tăng lên thì mật độ điện tử trên vùng dẫn càng tăng dần, lúc đó tính dẫn
điện của chất bán dẫn tăng dần theo nhiệt độ.
- Khi bị pha tạp, sẽ xuất hiện các mức pha tạp nằm trong vùng cấm khiến cho điện tử dễ bị thay
đổi chuyển lên vùng dẫn hoặc lỗ trống chuyển đến vùng hóa trị để tạo nên tính dẫn của vật liệu.
Do đó chỉ cần pha tạp với hàm lượng rất nhỏ cũng làm thay đổi lớn tính dẫn điện của chất bán
dẫn.
Dị thể :
Các dị thể xảy ra khi hai vật liệu bán dẫn pha tạp khác nhau được nối với nhau. Ví dụ, một cấu
hình có thể bao gồm p-pha tạp và n-pha tạp. Germanium . Điều này dẫn đến sự trao đổi điện tử
và lỗ trống giữa các vật liệu bán dẫn pha tạp khác nhau. Germanium pha tạp n sẽ có thừa
electron và Germanium pha tạp p sẽ có quá nhiều lỗ trống. Sự chuyển đổi xảy ra cho đến khi
đạt được trạng thái cân bằng bởi một quá trình gọi là tái hợp, khiến các electron di chuyển từ
loại n tiếp xúc với các lỗ di chuyển từ loại p.( Một sản phẩm của quá trình này là các ion tích
điện, dẫn đến hiệu ứng từ trường. )
Sự khác biệt về điện thế trên vật liệu bán dẫn sẽ khiến nó rời khỏi trạng thái cân bằng nhiệt và
tạo ra tình trạng không cân bằng. Điều này chuyển các electron và lỗ trống cho hệ thống, tương
tác thông qua một quá trình gọi là khếch tán xung quanh. Bất cứ khi nào cân bằng nhiệt bị xáo
trộn trong vật liệu bán dẫn, số lượng lỗ trống và điện tử sẽ thay đổi. Sự gián đoạn như vậy có
thể xảy ra do sự chênh lệch nhiệt độ hoặc photon và xâm nhập vào hệ thống và tạo ra các
electron vào lỗ trống. Quá trình tạo ra và tự hủy electron trong lỗ trống được gọi là thế hệ và tái
tổ hợp.
Ngoài ra tính bán dẫn này sẽ tăng dần trong các trường hợp có kích thích năng lượng khác như:

Thể hiện tính siêu dẫn:
Đây là hiện tượng lượng tử, nó có khả năng tạo ra lực hút phonon làm cho các electron dẫn
trong chất siêu dẫn biểu hiện pha siêu lỏng tạo ra từ cặp electron tương quan.[10]
Thể hiện tính nghịch từ:
- Chất nghịch từ là chất không có momen từ( tổng vecto từ quỹ đạo và từ spin của toàn bộ điện
từ bằng 0). Khi đặt vào từ trường ngoài trong các phân tử sẽ xuất hiện dòng điện phụ và tạo ra
từ trường phụ ngược chiều từ trường ngoài theo xu hướng cảm ứng điện từ( hiệu ứng vật lý
lượng tử).
Cho nên chất liệu nào cũng phát sinh nghịch từ khi từ trường tác dụng nhưng trong đa số chất
liệu hiệu ứng nghịch từ rất nhỏ và khó phá hiện. Chỉ trong chất siêu dẫn điện thì hiệu ứng này
mới mạnh.
Do đó độ thẩm điện môi của môi trường μ < 1, độ cảm từ χ < 0. Các chất nhóm này là các khí
hiếm như: I, He, Ne, Ar, Kr,...và các ion có các lớp electron giống khí hiếm. Nhiều kim loại như:
Bi, Zn, Ag, Cu, Pb, và 1 số́ chất khác như C, NaCl, SiO2, S, H2O.
3. 2 Sự khác biệt giữa Endo Metallofullerene Trịnh Năng và các Endo fullerene khác
1 - Là hoạt chất sinh học được cấu tạo chủ yếu từ Là các hoạt chất sinh học được
cacbon, chiếm 78,9%( chủ yếu là các C60, C70), những cấu tạochủ yếu bởi các phân tử
nguyên tử còn lại chỉ chiếm 21,09%, đó là các nguyên carbon (Cn), trong đó n >=36)
tử kim loại, á kim và khí hiếm pha tạp với các kim loại hoặc
cụm kim loại gọi là các dẫn xuất
- Cách điều chế ra hạt này tất cả đều là phản ứng hóa
fullerene
nhiệt để tạo ra lồng cacbon có các liên kết khối. Đây là
một chất trơ hoàn toàn, không một hóa chất nào có
thể phá vỡ liên kết khối này trừ ngâm trong toluen 60
giờ.
2 - Có kích thước từ 100nm đến 250nm đây là một kích - Kích thước phân tử thường
nhỏ (<100nm) nên khó bền, dễ

không đối xứng do có hình dạng

quang phổ, nó có thể hút được rất nhiều điện tích âm
hoặc dương ở xung quanh nó ( khả năng lưu trữ thông
tin lượng tử (qubit)) [8][9][4]
3 - Metallofullerene Trịnh Năng có lồng cacbon liên kết - Metallofullerenes khác thường
với 23 nguyên tử kim loại((23 loại: Al, Ca, Mg, Mn, Se, có từ 1- 4 nguyên tử kim loại
Be,Thiếc, Mo, Cd…. ) và nhiều á kim trong đó có 5 liên kết với lồng cacbon nên thể
nguyên tố thể hiện tính chất nghịch từ, do đó xảy ra hiện phản ứng oxy hóa khử kém

- Tính ổn định của N @ C60: sự hình thành N @ C60
không đơn giản về mặt năng lượng. Do sự tổng hợp
thành công nằm trong tính trơ hóa học của bên trong
lồng C60 (Nguyên tử nitơ có thể được sử dụng như
một đầu dò, để phát hiện những thay đổi nhỏ nhất
của cấu trúc điện tử trong môi trường của nó).
Mặc dù tất cả các cacbon trên lồng là sp 2, góc liên kết
của chúng gần hơn với góc liên kết trong sp 3, do đó
làm tăng sự liên kết do C60 thể hiện tính
trơ.[12][13][4]
5 Trong 5 nguyên tố có tính chất nghịch từ tại bảng - Rất hiếm các Endo fulleren có
Cerrificate của Analysis thì có nguyên tố được nguyên tố này do trong
Molypden(Mo) có mặt trong cấu trúc cabon này nên quá trình tổng hợp nhiệt độ
vật liệu này siêu cứng, gấp nhiều lần kim cương. chưa tới
6 Nguyên liệu tổng hợp các hoạt chất này hoàn toàn Nguyên liệu tổng hợp từ vô cơ
hữu cơ: sơ dừa, vỏ trấu, lõi ngô, vỏ tơ cafe… Do đó hoặc hữu cơ
hoàn toàn an toàn với cơ thể
7 Qúa trình tổng hợp được làm trên quy mô công Đa số được tổng hợp trong
nghiệp phòng thí nghiệm
8 Trong quá trình tổng hợp chỉ dùng nhiệt, không dùng Có thể được tổng hợp từ các
bất kỳ hóa chất nào dẫn xuất khác
Ở điều kiện bình thường, trên màng tế bào đều có các vùng âm điện tử và dương điện tử (anion
va cation), những vùng âm điện tử và dương điện tử này thay đổi liên tục vì các hệ thống bơm
qua màng sử dụng các âm điện tử và dương điện tử này như là năng lượng để có thể hoạt động
được. Do đó, trong một trạng thái bình thường, những vùng âm điện tử đó không đủ sức để tạo
ra một trường từ đủ mạnh để có thể kích thích hạt endo có thể nhảy tầng năng lượng để trở
thành một bẫy electron.
Ngược lại, trường hợp một màng tế bào vì lý do nào đó chưa xác định được và tích tụ một
mảng âm điện tử đủ mạnh và tạo ra một trường từ đủ mạnh, thì khi đó nếu có một hạt endo
trong không gian gần đó, nó sẽ kích thích cho hạt endo này trở thành một bẫy âm điện tử…và
như vậy hạt endo này sẽ trở thành một bẫy âm điện tử, hút các âm điện tử dư thừa trên màng
tế bào và giam các âm điện tử này trong không gian lồng của nó..
Khi nào thì hiện tượng này chấm dứt để có thể trở lại trạng thái bình thường?
khi mà số lượng âm điện tử trên màng tế bào bị bệnh chỉ còn ở mức tối thiểu, và không đủ sức
để tạo thành một trường từ đủ mạnh để có thể kích thích hạt endo, khi đó hạt Endo sẽ quay trở
lại trạng thái bình thường của nó ( ở trạng thái này nó không thể đủ sức để làm một bẫy âm
điện tử, lúc đó nó sẽ tạm thời quay về trạng thái trơ và sẽ chờ một cuộc hội ngộ tiếp theo với
một trường từ đủ mạnh khác) [16]
- Tất cả các tính chất trên đã mang lại cho vật liệu nano cacbon này rất nhiều các cơ hội mới đầy
hứa hẹn trong các ứng dụng đa chức năng y sinh nói riêng và ngành công nghiệp điện tử nói
chúng.

To understand the toxicity of C60, it is usually necessary to investigate the lethality, gross or
microscopic lesions, histopathological change, etc. However, other biological responses, such as
stimulation of the immune system, regulation of oxidative stress, and changes on the molecular
level induced by fullerenes, should generally be considered as well.
Following the inhalation pathway, C60 and its derivates can be partly deposited in the lungs and
internalized by alveolar macrophages; a pulmonary inflammatory response was elicited for a
high exposure dose of fullerenols (10 mg/kg) and fullerenols at low concentrations (1 mg/kg)
exhibited an anti-inflammatory property. Although slight changes in gene expression level were
observed, no obviously toxicity was reported [71, 72]. Only when water-soluble
polyalkylsulfonated C60 was administrated at extremely high dose (LD50=600 mg/kg) in an acute
toxicity study or at a moderately high dose (up to 60 mg/kg) via ip injection, observable
pathological or lethal toxicity was induced. However, in subsequent study of oral exposure,
fullerenes were considered to be nontoxic at a dose of 50 mg/kg, which was close to that of ip
administration [73]. Even when the dose was increased to 2,000 mg/kg, fullerite (a mixture of
C60 and C70) has not shown to cause any acute toxicity. The possible reason is that fullerenes are
not effectively absorbed through the alimentary tract [74]. Toxicity assessment of fullerenes
should therefore be considered comprehensively, including the exposure routes and the
delivered dose, etc.
After fullerenes enter into the body, they may be internalized by different types of cells, and
induce various changes inside the cells, such as viability, proliferation, inflammation responses,
and oxidative responses. Therefore, the cytotoxicity assessment of fullerenes has been one of
the most important aspects to consider when assessing the toxicity of fullerenes [75]. Many
related studies have been performed on various cell lines. C60 and quaternary ammonium
derivatized C60 inhibited keratinocytes proliferation at a concentration of 2 μM and over a
period of 8 days, which is relatively long [66]. In both human skin (HDF) and liver carcinoma
(HepG2) cells, water-soluble fullerene species are less cytotoxic than pristine C60, while
C60(OH)24 showed no cytotoxicity up to its limits of solubility, suggesting the functional groups
on the surface of a fullerene molecule can dramatically decrease the toxicity. However,
C60(OH)24 was found to induce cytotoxic morphological changes within the HUVEC cell line in a
dose-dependent way, and only maximal doses of fullerenes caused cytotoxic injury and/or
death and inhibited cell growth [76]. In nearly every fullerene-related anti-tumor study,
fullerenes were not found to have a direct influence on tumor cell viability [9, 38, 40, 42]. The
diverse toxicity results of fullerenes reveal that the sensitivity of various tissues or cells to the
nanoparticles can be different. Therefore, thorough toxicity evaluation of fullerenes before
their clinical application is necessary.
An important property of C60 molecule is its high symmetry. There are 120 symmetrical
operations, like rotation around the axis and reflection in a plane, which map the molecule onto
itself. This makes C60 the most symmetrical molecule [7]. There are two types of bond lengths
in the fullerene: C5-C5 single bonds in the pentagons and C5-C6 double bonds in the hexagons
[8]; the first is 1.45±0.015 Å and the other one is 1.40±0.015 Å. Each carbon atom forms bond
to three other adjacent atoms with sp [2] hybridization. The set of orbitals is arranged at 120-
degrees angles and is centred in the xy-plane. Hence these delocalized pi electrons stabilize the
spheroid structure by resonance [9].
A C60 molecule, also known as Buckyball or Buckminsterfullerene, is about 7 Å in diameter.
C60 molecules condense to form a solid of weakly bound molecules [10]. This crystalline state is
called fullerites. This solid is cubic, weakly bound [11] with a lattice constant a=14.71 Å and
electrically insulating. It occurs as yellow powder, which turns pink when dissolved in toluene.
On exposure to strong UV light, the Buckyballs polymerize, forming bonds between adjacent
balls. In the polymerized state, C60 no longer dissolves in toluene [12]. NMR studies of
C60 benzene solvates show free rotation at room temperature [13]. At about -13°, the balls
spin freely in their crystalline positions [14]. At lower temperature, their movements begin to
limit to certain orientations. Eventually, below -183°, the balls become completely struck [15].
Chemically the molecule is quite stable; breaking the balls requires temperature of over 1000°.
By heating fullerenes up to 1500° in absence of air, they transform to graphite.
Other than C60, fullerenes can contain between 30 to 980 carbon atoms forming different
structures with different properties and field of applications. As hexagons are added or
removed from the basic soccer ball structure, the molecule begins to lose its roundness. C70,
which has 25 hexagons, is shaped more like a rugby ball. Giant fullerenes take on a pentagonal
shape. Smaller fullerenes look like asteroids. With the loss of roundness comes a loss of stability,
C60 being the most stable of all. Other than C60 and C70, C76, C78, C84, C86 have been
isolated and studied in detail. Fullerenes have low density relative to diamond (1.65 g/cc
compared to 3.51 g/cc). Fullerenes are unstable in water. Good solvents for fullerenes are
carbon disulphide, o-dichlorobenzene, toluene and xylene [16-18]. Thin layers of fullerenes are
coloured from yellow to yellowish-green. The colour of fullerene solutions is attributed to the
pi-pi electron transition (Table 1).
Alkali-doped fullerenes
As fullerene molecule is highly electronegative, it readily forms compounds with electron
donating atoms, the most common examples being alkali metals [19]. This reaction leads to
production of an interesting class of compounds known as alkali-doped fullerides, wherein
alkali metal atoms fill in the space between Buckyballs and donate valence electron to the
neighbouring C60 molecule. If alkali atoms are potassium or rubidium, the compounds are
superconductors, and they conduct electric current without any resistance at temperatures
below 20-40 K [20], e.g., K3C60, Rb3C60.
Endohedral fullerenes
Since fullerenes are hollow with a closed shell of carbon atoms, it is possible to enclose another
atom inside. This class of fullerene derivatives is known as Endohedral fullerenes. When the
atom trapped inside is a metal, they are known as metallofullerenes [21]. Even though C60 is
the most common fullerene, few endohedral materials have been synthesized using C60 as
cage molecule, the reason being small size of the molecule. Most of endohedral materials are
made out of C82, C84 or even higher fullerenes. The atoms that form stable endohedral
compounds include lanthanum, yttrium, scandium, and some of the noble gases [22]. As it is
very difficult to open up carbon cage molecules to enclose a foreign atom inside, endohedral
material must be synthesized while formation of the cage itself [23]. (The accepted notation for
endohedral material is to use the @ symbol to show that the first material is inside the second,
e.g., La@C82 and Sc2@C84.)
Exohedral fullerenes
The most important and most versatile of all species of fullerenes is Exohedral fullerenes or
fullerene derivatives, which are molecules formed by a chemical reaction between fullerenes
and other chemical groups. Fullerene derivatives are also known as functionalized fullerenes. As
fullerenes possess the conjugated π-system of electron, two main types of primary chemical
transformations are possible on fullerene surface: addition reactions and redox reactions,
which lead to covalent exohedral adducts and salts respectively [24]. As fullerenes are insoluble
in water, numerous derivatives of fullerenes have been synthesized with improved solubility
profile.
Heterofullerenes
Heterofullerenes represent another fundamental group of modified fullerenes. They represent
heteroanalogues of C60 and higher fullerenes; one or more carbon atoms of the cage are
substituted by hetero-atoms, e.g., trivalent nitrogen or boron atom [25]. The simplest
derivative of nitrogen fullerene is aza (60) fullerene C59N. and its dimer (C59N)2.
Another significant spin-off product of fullerene research is carbon nanotubes based on carbon
or other elements [26,27]. These systems consist of graphitic sheets seamlessly wrapped into
cylinders. The diameter of nanotubes range from 0.4 to >3 nm for Single-Walled NanoTubes
(SWNT) to 1.4 to at least 100 nm for Multi- Walled NanoTubes (MWNT) [28,29]. Also nanotubes
are quite stiff and exceptionally strong, having high tensile strength. The calculated Young’s
Modulus for an individual nanotube is ≈ 0.64TPa [30]. This unique combination of diameter and
strength is potentially useful in everything from tennis rackets to space travel.
Biological Applications of Fullerenes
Fullerenes are inert, hollow and indefinitely modifiable. When administered orally in the water-
soluble form, they are not absorbed; while on i.v. injection, they get rapidly distributed to
various body tissues. They are excreted unchanged by kidney [31]. Acute toxicity of water-
soluble fullerenes was found to be quite low [32]. All these interesting properties offer
possibilities of utilizing fullerenes in biology and medicinal chemistry and promise a bright
future for fullerenes as medicinal agents. However, this possibility faces a significant problem,
i.e., natural repulsion of fullerenes to water. To overcome this limitation, a number of
methodologies are being developed. These include synthesis of fullerene derivatives having
modified solubility profile, encapsulation of C60 in cyclodextrins [33] or in calixarenes [34] or
water suspension preparations [35].
Many derivatives of fullerenes have been synthesized. There have been many patents written
on fullerenes, and fullerene patent database is growing rapidly. Fullerenes and their derivatives
are being intensively investigated as they exhibited promising preliminary activities in the
following medical streams.
Diagnostic applications
Endohedral metallofullerenes are the fullerenes with metal ion trapped inside fullerene cage.
These have shown potential applications in diagnostics. As for example, water solublised forms
like M@C82(OH)30 are being used as Magnetic Resonance Imaging contrast agents
(M=Gd3+)36, X-ray contrast agents (M=Ho3+)37 and radiopharmaceuticals
(M=166Ho3+and 170Tm2+) [38,39]. One of these derivatives, i.e., 166Ho3+@C82(OH)30 has
been extensively studied as radioactive tracer for imaging of diseased organs and for killing
cancerous tumours. The radioactive metal is trapped inside the carbon shell, which is very
stable and resistant to metabolism by body. The metallofullerenes have been found to be non-
toxic, and they stay in the body for approximately one hour, allowing imaging of circulatory
system.
AntiHIV activity
The enzyme protease specific to HIV-1 has been shown to be a viable target for antiviral
therapy. The active site of this enzyme can be roughly described as an open-ended cylinder
lined almost exclusively by hydrophobic amino acids except for two catalytic aspartic acids.
Wudl et al hypothesized that since C60 molecule has approximately same radius as the cylinder
that describes the active site of HIV-P, an opportunity exists for a strong hydrophobic
interaction between C60 derivatives and the active site surfaces. Inhibition of HIV-P in presence
of C60 was demonstrated through molecular modelling studies and experimental observations
[40]. Virus inactivation assays confirmed the activity of fullerene derivatives against HIV-1 and
HIV-2 [41,42]. A water-soluble derivative was found to be active against HIV-1 and HIV- 2
(EC50 approximately 6 μM) in acute and chronic infected human lymphocytes and also
noncytotoxic up to 100 μM in peripheral mononuclear cells and H9, Vero and CEM cells [43].
As the binding constant found experimentally was not significant in terms of affinity [40],
structural optimization of C60 derivatives for HIV-P interaction was investigated. On this basis,
some authors proposed ideal inhibitors, in which two ammonium groups at distance of 5.5 Å or
5.1 Å are directly linked to C60. Molecular modelling studies on these derivatives showed that
they could fit well inside the HIV-P cavity and can interact with carboxylic residues of aspartatic
acid [44].
DNA photocleavage
A water-soluble fullerene carboxylic derivative was found to be cytotoxic when exposed to
visible light. In an initial experiment, C60 carboxylic acid was incubated with cultured cells for
three days. No biological activity was observed. However on exposure to low power visible light,
the compound killed the cells. It was later shown that cytotoxicity of C60 derivatives was
mediated by its ability to cleave DNA [45]. Hence other potential application of fullerenes is
related to the easy photoexcitation of fullerenes [46] (Fig. 3) on the basis of which fullerene-
based photodynamic compounds are being developed for the treatment of cancer.

In this field, many fullerene conjugates with different units possessing biological affinity to
nucleic acids or proteins are being investigated for anticancer activity. In particular, conjugate
between C60 and specific agents that interact with nucleic acids, such as acridine [47] or
complementary oligonucleotide [46,48], have been synthesized with the objective of increasing
both cytotoxicity and sequence selectivity. However, DNA cleavage was found to occur at
guanine residue without any significant sequence selectivity [45]. A C60-PEG conjugate
irradiated by light strongly induced tumour necrosis without any damage to the overlying
normal tissue, with complete cure achieved by a C60-PEG dose of 0.424 mg/kg and irradiation
power of 1011 J/m2, making it an excellent candidate for photodynamic tumour therapy [49].
Cytotoxicity of dendritic C60 monoadduct and malonic acid C60 trisadduct was recently
investigated on Jurkat cells, and on exposure to UV light, the cell number was found to reduce
to approximately 19% within two weeks [50].
Free radical scavenger
Free radicals have been linked to onset and progression of foetal neurogenerative diseases like
amytrophic lateral sclerosis, commonly known as Lou Gehrig’s disease. Fullerene compounds
with their unique cage structure, combined with high number of conjugated double bonds (30)
in the core, interact with biomolecules and have avid reactivity with free radicals. Buckminster
fullerenes, for example, are capable of adding multiple radicals per molecule; the addition of as
many as 34 methyl radicals to a single C60 sphere has been reported. Hence they are regarded
as ‘‘free radical sponge’’[51]. However to improve native solubility profile of fullerenes, several
fullerene derivatives have been prepared, and these are reported to possess protective effect
in various systems. A water-soluble C60 derivative, fullerenol, has been shown to scavenge free
radicals in vitro [52] and in vivo [53]. Fullerenol has been shown to block hydrogen peroxide-
induced inhibition of population spikes in rat hippocampus [53]. Carboxy-fullerene, another
water-soluble derivative of fullerene, has been shown to prevent apoptotic injury evoked by N-
methyl D-aspartate (NMDA) without interfering with NMDA-induced Ca2+ influx in cortical cell
cultures [54] and has been found to have effective neuroprotective antioxidant activity in
vitro and in vivo (hundreds of times more powerful than Vitamin E). As a free radical scavenger,
the protective effect of fullerene derivatives has been demonstrated in various systems,
including reduced injury on ischemia reperfusion intestine [55], protecting cell types from
undergoing apoptosis [56,57], reduced free radical level in organ perfusate [58], and having
neuroprotective effect [59,60].
Additional factors that fullerene derivatives have shown promising results are in pretoxicity
studies and the fact that water as wall as lipid soluble derivatives are possible greatly increases
their usefulness as antioxidant in health and personal care products, e.g., skin creams, burn
creams and nutritional supplements.
Antimicrobial activity
Fullerenes were anticipated to possess antibacterial activity based on the hypothesis that
C60 could produce membrane disruption by insertion into phospholipids bilayers [61]. The
resulting membrane disorder, causing altered permeability, leads to the release of metabolites
and cell death. As for example, monomethoxy triethylene glycol (mTEG) substituted
fulleropyrollidines showed complete inhibition of Mycobacterium avium at dose 260 μg/ml and
M. tuberculosis at dose ≅ 50 μg/ ml [62]. The plausible explanation for the antitubercular
activity has been proposed that presence of carbon cage destabilizes the tubercular cell wall by
intercalation in the hydrophobic part. The affinity of carbon cage for the hydrophobic
constituents of Mycobacterium cell wall, i.e., ceramide has been further proved by
administration of intraperitoneal injection of mTEG fullerene derivative to mice. The derivative
was found to be deposited on Tyson’s gland, which is again a tissue rich in ceramide [63].
Antimicrobial activity of another fullerene derivative, i.e., carboxyfullerenes has been
intensively investigated. Carboxyfullerenes were found to inhibit E. coli-induced meningitis by
reducing the damage caused by infiltrating neutrophils on the blood brain barrier but not by
direct inhibition of E. coli [64]. Moreover, it was found that the carboxyfullerenes inhibited the
growth of Streptococcus pyrogens strain A-20 (MIC 5-50 mg/l) [61]. Since these derivatives
exhibited varying effects on Gram+ve and Gram-ve bacteria, further investigations led to the
observations that carboxyfullerenes can insert into cell wall of Gram+ve cocci, disrupt the cell
wall structure and cause bacterial death. The cell wall of Gram-ve organisms, having an outer
membrane consisting of lipoproteins, lipopolysachharides and phospholipids, is not susceptible
to access by fullerenes [65]. This suggests that carboxyfullerenes could be considered as new
antimicrobial agents against Gram+ve cocci.
Osteoporosis
To date, biphosphonate compounds and fluoride anions are the drugs used in the treatment of
osteoporosis and other bone disorders. However, the former are not absorbed orally and latter
are fairly toxic. Advantage is being taken of the preferential localization of fullerene derivatives
in bones. Hence polyfluoro biphosphonated fullerene derivatives are being developed as
bimodal drug for osteoporosis therapy [66].
However, the use of fullerenes as biological or pharmacological agents requires the
measurement of dosage and serum levels by sensitive and simple immunological procedures.
This in turn requires that specific antibodies to fullerenes should be available. The requirement
led to the question of the ability of these carbon-made spherical structures to induce the
production of specific antibodies. However, it was determined that immunization of mice with a
water-soluble C60 fullerene derivative conjugated to bovine thyroglobulin yielded a population
of fullerene-specific antibodies of the IgG isotype, showing that the immune system was diverse
enough to recognize and process fullerenes as protein conjugates [67]. A monoclonal fullerene-
specific antibody has been isolated and the sequences of its light and heavy chains have been
reported [68]. These findings further substantiate the practical utility of fullerenes as
therapeutic agents, diagnostic agents and as imaging vectors.
Conclusions
The exponential increase in patent filing and publications indicate growing industrial interest
that parallels academic interest. The discovery of fullerenes has been compared to the
discovery of benzene by many researches. The hydrophobic spheroid structure and radical
sponge character of the fullerene molecule is responsible for activities in many fields. In
addition to some of the important medical applications as mentioned above, fullerenes are
being intensively investigated for practical utilization in a number of other fields, e.g., as room
temperature superconductors, as component of solar batteries, for production of synthetic
diamonds and as lubricants.
However, there are two important facts that are considered to be barriers to fullerene
applications. The first one is relative insolubility and instability of fullerenes to water. This
however is being successfully overcome with the development of numerous water-soluble
fullerene derivatives. The second and more important limitation is the high cost of fullerenes.
Compared to gold that is available at the cost of $10/g, the cost of fullerenes, as listed on web,
varies from $45 to $1000/g, depending on degree of purification. This high cost, which is mainly
attributed to very high temperature involved in the production process, is restricting the
numerous potential applications of fullerenes. One possible development that may take place
in near future is creation of modest demand of fullerenes in medicine, as medicine is
considered to be the cost-insensitive application. This in turn will encourage larger production
leading to lower prices and still more demand. The preliminary biological investigations of
fullerene derivatives as discussed above do promise encouraging results. Hence research efforts
in both chemistry and biology should be to further investigate the potential applications of
these interesting spheroidal molecules.

Introduction
A sample absorbing a single ultraviolet photon may emit a single electron having energy and
momentum distributions that reflect the spectral properties of the material1. It is also possible,
though usually less probable, that two electrons escape. How can one photon “kick out” two
electrons? For few-electron atoms, it is established2 that the Coulomb repulsion plays a key
role. A possible scenario is that the photon is absorbed by one bound electron that approaches
the other electron while undergoing multiple scattering from the residual ion or the other
electron. Mediated by electron-electron Coulomb interaction, the two electrons exchange
momentum while leaving the sample and interacting mutually and with the residual ion, in
principle to infinite distances. This physical picture, often referred to as knock-out mechanism,
dominates for photon energies close to double ionization threshold, whereas for larger photon
energies different processes (e.g. shake-off ) become important3. When detecting the two
electrons in coincidence (called double photoemission (DPE) spectroscopy4,5,6), depending on
the selected energies and angles, one may zoom into some of these processes, albeit with
restrictions imposed by symmetry2,7.
The situation changes with a growing number of electrons in the system. The effective electron-
electron interaction is not even known a priori as it is determined by the dynamic behavior of
its active surrounding, meaning that the e–e interaction builds up during the photoexcitation
process. Thereby, dimensionality is a key factor8. In fact, for electronic systems strongly
confined to one dimension (e.g., a one channel quantum wire) e-e interaction gives rise to a
new form of excitations (Luttinger liquid9) with features distinct from those akin to Fermi
liquids, i.e. most three-dimensional systems. As DPE experiments are available for weakly and
moderately correlated surfaces and bulk materials (e.g. Cu, NiO or CoO10,11,12), it is valuable
to consider DPE for nano-sized systems that bridges the extended and atomic cases.
A possible scenario of DPE is that the photon excites one electron which senses its environment
for accessible scattering channels (elastic, phononic, magnonic, etc). DPE at a fixed incident
photon energy via the selection of the energy sharing and relative angles between the two
escaping electrons zooms into those channels, where electron-electron (e–e) interaction is
operational. The focus here is on e–e interaction mediated by charge density fluctuations in
confined geometry. On the other hand, electronic correlations are at the heart of diverse
fundamental phenomena such as superconductivity and plasmon formation which underlines
the relevance of the information encoded in the DPE spectra. Theoretically, the treatment of
two-particle correlations is a central problem in many-body physics13,14,15,16.
For the electron gas in particular, focus was put on two aspects affecting the two-particle
interaction. 1) Long wave-length density fluctuations which are characterized by the presence
of classical excitations (plasmons) and are well captured, for instance by the time-dependent
density functional theory (TDDFT) or the random phase approximation17,18. 2) Short wave-
length effects (exhibited in the on-top pair distribution function19,20,21) which are captured by
the ladder diagrams22,23. Exploiting the tunability of synchrotron radiation, DPE (cf. Fig. 1(a))
can be tuned to an energy region where the dynamic and non-local field of collective
excitations (plasmons) is the main driving for secondary electron emission whilst short-range
effects govern the formation of two-particle scattering states.

The emerging physical picture is illustrated in Fig. 2(a): (i) Photoabsorption promotes a valence
electron to a high-energy state. (ii) This electron scatters inelastically from charge-density
fluctuations (plasmon creation) that (iii) decay emitting a second valence electron (whose
energy and angular correlations with the first one is measured in a coincidence set up, revealing
so how charge-density fluctuations mediate e–e interaction). This three-step mechanism (3SM)
emerges from a diagrammatic nonequilibrium Green’s function (NEGF) approach as detailed in
the supplementary information. It is already clear at this stage that DPE is qualitatively different
from SPE in that, a) it delivers information on e–e interaction mediated by charge-density
fluctuations, and b) as these plasmonic excitations are triggered by an electron a multitude of
modes, e.g. volume plasmons, are involved.

In Fig. 1(b) the electron pair coincidence yield versus the binding energy of the doubly
charged ion is reported and compared with the Auger spectrum. The binding energy of the
latter is determined by the energy of the secondary electron and the carbon 1 s core level (see
methods). The Auger process, which one might expect to be comparable to DPE when plotting
as a function of the binding energy, can be interpreted in terms of the joint density of states
(JDOS) as determined by the convolution of the density of occupied states of the neutral
system, , and that of the ionized molecule, , . Our ab initio calculations in Fig.
1(b) confirm this picture (note, these same and are also part of DPE and are
calculated with the same code). For plasmon-mediated DPE the situation is different. As
inferred from Fig. 2(a), (ii), the spectral width of the plasmon modes is a determining factor for
the width of the DPE spectrum. Which mode is active (and what is its multipolar nature) is set
by the momentum balance that in turn points to the momentum region of the involved
plasmons. The full ab initio calculations of multipolar plasmons in C60 in ref. 30 enter as a part
(i.e. steps (ii)–(iii) Fig. 2(a)) of our DPE calculations.
The electron pair coincidence yield is calculated following the derivation in the supplementary
information. From Fig. 2 one infers that the non-local, frequency dependent screened electron-
electron interaction W = v + vχv is a central quantity for DPE (v is the bare Coulomb interaction).
As expected from the scheme in Fig. 2 the density-density response function χ(r, r′; ξ) is also the
key factor for the electron energy loss experiments31,32,33 and also for the screening of the
optical field34,35 by charge-density fluctuations in SPE (in our experiment this effect is
negligible because the optical frequency is higher than the relevant plasmon resonances). We
write the effective e-e interaction in the form
Here, ξ denotes the frequency dependence, while λ represents a screening parameter discussed
below. The collective modes are well characterized by the multipolarity L and a radial quantum
number ν30,36. We account for symmetric surface (SS), the anti-symmetric surface (AS) and
volume (V) modes (L = 0). The Lehmann representation of the response function is expressible
as
are the spherical harmonics. are known as fluctuation densities, which can be
interpreted as the spatial distribution of the density oscillation associated to a particular
plasmon (Fig. 2(b)). The corresponding frequency spectrum is represented by Bν,L(ξ). For the
radial profiles Rν,L(r) and plasmon spectra Bν,L(ξ) we utilize our recent approach from
ref. 30 that yielded EELS spectra in very good agreement to experiments33. The static part in eq.
(1) is written as . Previous calculations37 provided an insight into the value of λ. The two-
electron coincidence yield, averaged over the initial orientations of C60, reads

The momenta of the two photoelectrons are denoted by k1 and k2. The sum over n runs over
all occupied states of the singly ionized molecule and is the corresponding spectral
elements . Inspecting eq. (3) one identifies the steps (i)–(iii) sketched in Fig. 2(a). Note, due
to rearrangement of the ionic core, the energy levels of the neutral molecule ( ) are
lowered by Δ when removing one electron, such that the IP increases [see Fig.
2(a)]. Overall energy conservation follows from the restrictions (i) , (ii) , and (iii) .
states . Hence, DPE in the present case adds new aspects to DPE from, e.g., atomic targets,
and is a useful sensor for the e–e interaction mediated by charge-density fluctuations.

which is mainly responsible for the DPE signal at photon energies eV, is suppressed by the
energy selectivity of the plasmon excitation. In particular, the plasmon giving rise to the
emission of the second electron at stage (iii) needs to provide sufficient energy to promote a
certain initial state of the molecule [Fig. 3(c)] to the continuum. Hence, the limited
spectral width of the SS plasmon modes suppresses the emission from deeper σ states [Fig.
3(b)]. A test calculation replacing the plasmon spectra Bν,L(ξ) by a constant produced a
significantly broader DPE spectrum. This confirms the picture outlined above.
Our theory permits also to selectively include different plasmonic modes in the calculation. It is
known that plasmon excitation upon photoabsorption obeys optical dipole selection rules and
allows for exciting the SS plasmon with L = 1 only (the dipolar resonance mostly addressed in
the literature and manifested e.g. in plasmon-assisted SPE40). On the contrary, the electron
scattering (as in EELS) transfers a finite momentum meaning that SS and AS plasmons with any
multipolarity can be excited30,33,36. In agreement with this Fig. 3(d) underpins the significant
contribution of the AS plasmons and thus substantiates the physical picture of the 3SM,
according to which the step ii) can be regarded as an inelastic electron scattering event that is
inherent to the plasmon-assisted DPE process. Similarly, SS dipolar plasmon transitions play
only a minor role, while the multipolar plasmons are responsible to a large extent for the
coincidence yield [Fig. 3(e)]. All these facets endorse that DPE mediated by charge-density
fluctuations as the predominant channel for e-e correlations represents a new facet to the
information what is extractable from SPE and Auger spectra.
To summarize, an ab initio scheme for this process has been implemented with results in line
with the first DPE experiment resolved with respect to the electron pair energies. We identified
the dominant pathway as the following: a valence electron absorbs the photon and rescatters
inelastically from multipolar collective modes that mediate the coherent emission of a second
electron. The dwell time for this quasi-resonant scattering may be accessed by attosecond
time-delay experiments41. For plasmon-assisted DPE the average electronic density plays a
decisive role. For metals the plasmonic energies (which can be estimated using a classical
expression with rs being the Wigner-Seitz radius) are too low for plasmons to lead to a
direct electron emission, although these modes may likely contribute to the loss channel for
DPE. In contrast, for confined systems such as Carbon-based fullerenes the density is much
higher (rs ≈ 1.0 aB) resulting in plasmonic peaks in the XUV range. Thus, energy- and angle
resolved DPE experiments open the opportunity to explore different regimes of electronic
correlation, including Coulombic scattering, local field effects and dynamical screening.
COVID-19 nhanh chóng được phát hiện là do một loại coronavirus gây ra sau đó được đặt tên là

người và là loại coronavirus thứ ba gây viêm phổi.Trong đó COVID-19, viêm phổi thường phức
tạp do nhiễm vi khuẩn Streptococcus pneumoniae , thường nhắm vào đường hô hấp trên và có
thể gây tử vong
- Các Endo fullerene có khả năng kháng vi rút, ức chế sự xâm nhập của vi rút, thay đổi hình thái
và chức năng của nó, đồng thời ngăn chặn sự nhân lên của vi rút. Các nghiên cứu về cơ chế hoạt
động kháng vi rút cho thấy rằng khối cầu cacbon C 60 phù hợp với vị trí hoạt động của một số
enzym của virus
Trong một bài báo cáo khoa học người ta đã phát hiện: Vi-rút cúm A là một vi-rút mạch đơn
mang điện tích âm, thuộc nhóm Baltimore V. Tuy nhiên, nó là một vi-rút RNA có vỏ bọc giống
như SARS-CoV-2. Cơ chế hoạt động kháng vi rút của Endo fullerene có thể là do thành phần
lipid của màng vi rút có khả năng tạo lớp màng bề mặt. Các glycoprotein bề mặt được xác định
là bị mất tính toàn vẹn của vỏ virus khi chúng tiếp xúc với hạt Endo Fullerene bằng cách phá vỡ
cấu trúc vỏ lipoprotein (protein S), và những phức hợp này hợp nhất với các tập hợp giống
virion.[21]
Các nhà nghiên cứu cho biết các nghiên cứu chủ yếu cho rằng hoạt động kháng khuẩn của CBN
là do các cơ chế vật lý và hóa học như phá vỡ màng, chuyển điện tử và gây ra sự rối loạn oxy
hóa bởi các loại oxy phản ứng. Các cơ chế này đều được đặc trưng bởi nguy cơ kháng kháng
sinh thấp.
Tính kỵ nước cao của fullerene nguyên sinh có nghĩa là các dẫn xuất fullerene kháng virus có thể
được tổng hợp để tạo ra các loại thuốc ưa nước dễ phân tán trong nước và ức chế sự xâm nhập
của virus, thay đổi chức năng của virus và ngăn chặn sự nhân lên của virus
Một nghiên cứu được thực hiện vào năm 2011 cho thấy các dẫn xuất C70-fullerene hòa tan cao
trong nước và có tác dụng diệt virus chống lại HIV và vi rút cúm. Gần đây, một loạt các dẫn xuất
fullerene mới đã chứng minh khả năng ức chế vi rút viêm gan C.
Một nghiên cứu năm 2019 về các chấm carbon đã được chức năng hóa cũng đã chứng minh
hoạt tính kháng vi-rút chống lại nhiễm trùng coronavirus ở người trong tế bào gan của con
người. Các dấu chấm làm gián đoạn sự xâm nhập và sao chép của virus, nguyên nhân là do sự
tương tác của chúng với thụ thể dipeptidyl peptidase 4 (DPP4) của tế bào chủ.[22]

- Trên thế giới các nhà khoa học đã nghiên cứu rất nhiều về sự tĩnh điện bề mặt của
Coronavirrus 19 để từ đó có những ứng dụng phòng tránh. [29]
Ở môi trường ngoài cơ thể vật chủ: Người ta đã thấy rằng tất cả các virus có khối lượng, điều
đó có nghĩa nó có electron, proton và lõi. Nó di chuyển trên không khí và các bề mặt. Vì vậy, có
nghĩa là nó cũng có một điện tích. Tất cả các bề mặt mà virus có thể sống sót đều mang điện
tích âm tự nhiên. Nước, hydro, giọt nước nhỏ, virus…do đó chúng ta có thể sử dụng các bề mặt
tích điện dương là một biện pháp phòng ngừa tốt.
Cấu trúc của virus SARS-CoV-2 được tạo nên từ hơn 20 phân tử protein. Mặc dù virus SARS-
CoV-2 sử dụng nhiều loại protein khác nhau để sinh sôi và xâm nhập tế bào, nhưng protein gai
(S) là protein quan trọng nhất mà virus sử dụng để gắn vào thụ thể trên bề mặt tế bào vật chủ.
Trong vỏ của virion này là nucleocapsid (là thành phần cấu trúc chính của virion và tham gia
vào quá trình sao chép, lắp ráp và điều hòa miễn dịch của virus, đóng vai trò quan trọng trong
vòng đời của virus.) . Các coronavirus có các nucleocapsid đối xứng theo hình xoắn ốc, điều này
không phổ biến ở các virus RNA cảm nhận dương tính, nhưng phổ biến hơn nhiều đối với các
virus RNA cảm nhận âm tính.[28]
- Các nhà khoa học cũng phát hiện một vị trí mang điện tích dương nằm cách vị trí gắn kết trên
protein gai của virus SARS-CoV-2 khoảng 10 nanomét. Khu vực có điện tích dương này không
chỉ cho phép các protein của virus gắn kết chặt chẽ với nhau, mà còn giúp virus bám vào các
protein thụ thể mang điện tích âm trên tế bào chủ của người (được gọi là ACE2) .
Với phát hiện này, người ta đã thiết kế một phân tử mang điện tích âm để có thể hút các điện
tích thừa trên, từ đó ngăn chặn quá trình gắn kết của SARS-CoV-2 với tế bào vật chủ hoặc ngăn
chặn sự lây lan của virus

In the case of the 3rd-degree burns, they are particularly susceptible to bacterial infection as
the barrier function of the skin is destroyed, the dead tissue is devoid of host-defense elements,
and a systemic immune suppression is a worrying consequence of serious burns. Furthermore,
the lack of perfusion of the burned tissue means that systemic antibiotics are generally
ineffective.116 Although excision and skin grafting is now standard treatment for the 3rd-
degree burns,117 superimposed infection is still a major problem. Patients with Gram-negative
burn infections have a higher likelihood of developing sepsis than Gram-positive infections.
Topical antimicrobials are the mainstay of therapy for burn infections and PDT may have a
major role to play in the management of this disease.118
We found in previous study a decacationic fullerene LC17 ( C70[>M(C3N+6C3)2]) was effective
at mediating the photokilling of Gram-negative bacteria in vitro.66 We synthesized a new
compound C70[>M(C3N+6C3)2][>M(C3N6C3)2] (LC18) with the same decacationic side chain
plus an additional deca-tertiary amine groups against Gram-negative bacteria. A mouse model
of the third-degree burn infection with bioluminescent Gram-negative bacteria was used to test
the in vivo effectiveness of the therapeutic approach using the UVA excitation.119 The data
shown in Figures 10 and and1111 suggested that the attachment of an additional
deca(tertiary-ethylenylamino) malonate arm to C70, producing LC18, allowed the moiety to act
as a potent electron donor and increased the generation yield of hydroxyl radicals under UVA
illumination. This is consistent with the reported phenomena of photoinduced intramolecular
electron transfer from the tertiary amine moiety to the fullerene cage in polar solvents,
including water, at the short excitation wavelength. With the availability of ten tertiary amine
moieties, each capable of donating one electron to the C70 cage, LC18 may function as an
electron-rich precursor for hydroxyl radical production that demonstrated a new approach in
enhancing HO• radical killing of pathogenic bacteria in contrast to the more common 1O2-
killing mechanism.

III. CƠ CHẾ HOẠT ĐỘNG CỦA GỐC TỰ DO VÀ SỰ HÌNH THÀNH TẾ BÀO UNG THƯ
1. Gốc tự do là gì?
Gốc tự do là phân tử, ion hay một nguyên tử có số một electron hay có số electron lẻ nên
thường không ổn định dễ dàng phản ứng với các phân tử khác trong tế bào.[30]
Gốc tự do có thể mang điện tích dương, âm hoặc không mang điện và chúng đều giữ vai trò
quan trọng trong hệ thống sinh học. Chúng có lớp điện tử ngoài cùng chứa một điện tử không
ghép cặp (hay gọi là điện tử đơn độc), do có điện tử không ghép cặp ở lớp ngoài cùng nên gốc
tự do rất không ổn định, chúng luôn có xu hướng cướp điện tử của các nguyên tử hay phân tử
khác để trở về trạng thái ổn định, nhưng lại biến các nguyên tử hoặc các phân tử này trở thành
gốc tự do, làm cho cấu trúc tế bào bị thay đổi và bị phá vỡ.
Các gốc tự do được sinh ra trong cơ thể chủ yếu từ hai nguồn:
- Nguồn gốc nội sinh ( sinh lý tế bào): được tạo ra thường xuyên do chuỗi hô hấp tế bào. Chuỗi
hô hấp tế bào là quá trình chuyển đổi năng lượng cho sự hoạt động của các tế bào, năng lượng
được chuyển đổi chủ yếu từ carbohydrate
- Nguồn ngoại sinh: Do các tác nhân phóng xạ, phản ứng viêm, căng thẳng, mệt mỏi kéo dài,
thuốc lá, ô nhiễm môi trường và một số tác nhân khác.
Đó chính là lý do vì sao, mỗi ngày có hơn 10000 gốc tự do tấn công vào cơ thể chúng ta.
Một số loại gốc tự do nguy hiểm gây hại cho cơ thể như: Superoxide, ozone, hydrogen peroxide,
peroxy lipid, hydroxyl radical gây ra nhiều tổn thương tế bào. Các gốc tự do có hoạt tính rất
mạnh nếu tăng quá mức sẽ gây ra những tổn thương đến tế bào, làm tăng nguy cơ bệnh tật. Và
khi tấn công vào cơ thể chúng ta, gốc tự do vì chưa tạo được thành cặp, không cân bằng về điện
tử do đó nó rất bất ổn, dễ dàng tạo ra phản ứng chiếm đoạt điện tử của các phân tử khác để
tạo thành cặp và lần lượt tiếp tục tạo ra chuỗi gốc tự do mới gây rối loạn hoạt động bình
thường của tế bào.
2. Những ảnh hưởng của gốc tự do đối với cơ thể
Gốc tự do có thể dễ dàng phản ứng với các thành phần của tế bào, qua đó các gốc tự do gây ra
sự rối loạn hoạt động bình thường của tế bào, phá hủy tế bào và đẩy nhanh quá trình lão hóa
tự nhiên của cơ thể. Chính vì nguy hại như vậy, gốc tự do được xem là “sát thủ giấu mặt” gây ra
quá trình lão hóa và phần lớn các bệnh tật. Y học hiện đại đã thống kê, sự tấn công của gốc tự
do gây ra hơn 60 loại bệnh khác nhau, trong đó đặc biệt nguy hiểm là các bệnh ảnh hưởng đến
não bộ như suy giảm trí nhớ, sa sút trí tuệ, Alzheimer, đột quỵ…[31]
Nhiều nghiên cứu thấy sự liên quan của các gốc tự do đối với sự hình thành một số bệnh lý gồm:
- Bệnh lý ở hệ thần kinh trung ương như: Alzheimer, Parkinson, các chứng mất trí nhớ
- Bệnh lý tim mạch do tắc động mạch gây ra.
- Rối loạn viêm và hệ thống miễn dịch: Các bệnh tự miễn thường gặp như viêm khớp dạng
thấp, lupus ban đỏ
- Gốc tự do là tác nhân gây đột biến gen khi tấn công vào các ADN và tấn công vào các tổ chức
màng, các chất sinh học trong tế bào góp phần làm tăng nguy cơ ung thư và làm cho bệnh cảnh
ung thư nặng nề thêm.
- Đục thủy tinh thể và chứng suy giảm thị lực liên quan đến tuổi tác
- Thay đổi về ngoại hình do sự lão hóa gây ra như da mất độ căng bóng, đàn hồi, xuất hiện
nhiều nếp nhăn, tóc bạc, rụng tóc, móng tay yếu.
Liên quan tới bệnh lý đái tháo đường ở người già.
Bên cạnh những ảnh hưởng tiêu cực đến cơ thể thì sự hình thành các gốc tự do nội sinh cũng
rất cần thiết cho cơ thể, giúp cho quá trình tổng hợp và phân hủy các chất trong quá trình
chuyển hóa tế bào.
3.Tác hại của gốc tự do khi nó tấn công vào cơ thể
Khi xâm nhập vào cơ thể, gốc tự do sẽ bắt đầu phá hủy sức khỏe của chúng ta theo trình tự sau:
Đầu tiên, gốc tự do sẽ tấn công vào màng tế bào. Chúng sẽ oxy hóa màng tế bào gây khó
khăn, trở ngại cho việc thải chất độc và hấp thu chất dinh dưỡng.
Sau khi đánh bại lớp màng tế bào, các gốc tự do tiếp tục tấn công vào các ty lạp thể, phá
vỡ nguồn cung cấp năng lượng cho tế bào.
Sau cùng, bằng cách oxy hóa, gốc tự do sẽ làm suy yếu kích thích tố và các enzym khiến
cơ thể không tăng trưởng được.[30][31]
Gốc tự do là kẻ thù đặc biệt nguy hiểm đối với sức khỏe con người. Chúng là nguồn gốc của sự
lão hóa và là tác nhân của hơn 100 loại bệnh tật ảnh hưởng đến chất lượng cuộc sống con
người.
Các bệnh do gốc tự do có thể phát sinh trên hầu hết các bộ phận của cơ thể như não, mắt, da,
hệ miễn dịch, tim, mạch máu, phổi, thận, đa cơ quan và khớp. Và gốc tự do cũng là nguyên
nhân hàng đầu gây nên bệnh ung thư do nó “cướp” điện tử, làm tổn thương màng tế bào, phản
ứng mạnh với các phân tử protein, DNA và các axit béo, dẫn đến những rối loạn trong hoạt
động của tế bào, gây tổn hại, rối loạn và làm chết tế bào
4.Cơ thể chúng ta chống lại gốc tự do bằng cách nào?
Nếu như phải chọn một cỗ máy thần kỳ nhất trên thế giới, thì có lẽ cơ thể con người chính là
lựa chọn đầu tiên.
Vì sao? Bởi cả cơ thể con người là một hệ thống với các bộ phận liên kết chặt chẽ với nhau. Hệ
thống này làm việc ngày đêm không ngừng nghỉ để duy trì sự sống cho con người. Nếu có bất kỳ
sai phạm nào xảy ra, cơ thể chúng ta sẽ phát hiện ra ngay và tìm cách khắc phục.
Khi gốc tự do tấn công vào chúng ta cũng vậy, cơ thể sẽ có biện pháp phù hợp để loại bỏ chúng.
Khi bị các gốc tự do xâm nhập vào và phá hủy tế bào, gây tổn hại cho DNA thì cơ thể sẽ tiết ra
một loại enzym có chức năng cắt và loại bỏ những phần AND bị hỏng, tiếp theo sẽ tổng hợp lại
và gắn các DNA trở lại vị trí cũ, từ đó sửa chữa, khắc phục những sai phạm trên DNA.
Trường hợp gốc tự do phá hủy các phân tử protein, cơ thể sản xuất ra enzym cellular protease
có khả năng nhận diện protein hỏng và phân hủy nó để tạo ra nguyên liệu cho quá trình tổng
hợp protein mới.
Bên cạnh đó, các gốc tự do có thể tạo ra các chuỗi phản ứng oxy hóa lipid khác nhau để tạo ra
các chất oxy hóa mới và chúng tiếp tục gây tổn hại đến cơ thể. Những loại gốc tự này đều
được vitamin E, vitamin C, vitamin A, Selence, Glutathione Peroxidase và các chất bảo vệ khác
trong cơ thể chống lại và tiêu diệt chuỗi phản ứng sinh ra các chất oxy hóa mới.
Tuy nhiên, nếu có quá nhiều gốc tự do tồn tại trong cơ thể, bộ máy bên trong chúng ta không
thể tự loại bỏ hết được. Và nếu một lỗi nhỏ không được khắc phục, thì hậu quả sẽ ngày càng
lớn và cuối cùng, sức khỏe, sự sống của chúng ta sẽ bị đe dọa.
Vì lý do đó, mà chúng ta phải tìm mọi cách chống lại gốc tự do, hạn chế sự tấn công của nó vào
cơ thể, đồng thời bổ sung cho cơ thể những chất chống oxy hóa để khử các gốc tự do.
Ung thư có thể xuất hiện ở gần như bất kỳ đâu trong một ngàn tỷ tế bào của cơ thể. Bình
thường, các tế bào lớn lên và phân chia để hình thành tế bào mới, là cách thức mà cơ thể người
sinh trưởng và phát triển. Tất yếu, các tế bào cũ sẽ dần già đi hoặc bị tổn hại, chết đi, và được
thay thế bởi các tế bào mới.[35]
Khi ung thư xuất hiện, quá trình tự nhiên bị phá vỡ. Các tế bào ngày càng trở nên bất
thường, các tế bào già cũ không chết đi mà tiếp tục phát triển, liên tục sản sinh các tế bào mới.
Chúng cứ thế nhân lên không kiểm soát, và cuối cùng tạo thành khối bất thường mà chúng ta
gọi là khối u.
Rất nhiều loại ung thư hình thành khối u rắn, đa số có bản chất là các khối mô. Các loại ung
thư máu, ví dụ như leukemia, lại thường không xuất hiện u rắn.
Các khối u trong ung thư có tính chất ác tính, có nghĩa là chúng có khả năng xâm lấn ra
xung quanh. Trong quá trình phát triển lớn lên, các tế bào ung thư có thể đi xa tới các vùng
khác của cơ thể thông qua hệ tuần hoàn hoặc hệ bạch huyết, hình thành khối u mới tách biệt
hoàn toàn với khối u ban đầu, gọi là di căn.
Không giống các khối u ác tính, u lành tính không có tính xâm lấn, dù đôi khi u lành tính cũng có
kích thước rất lớn. Khi loại bỏ u bằng phẫu thuật, u lành tính thường không tái phát (trong khi u
ác tính rất hay tái phát). Tuy nhiên không phải lúc nào u lành tính cũng vô hại, ví dụ như trường
hợp u não lành tính có thể đe dọa tính mạng bệnh nhân
2. Ung thư là một bệnh lý liên quan tới gen, phần lớn do gốc tự do tạo ra:
Bởi ung thư hình thành khi có sự thay đổi bất thường trên gen, khiến cơ chế kiểm soát hoạt
động của tế bào (đặc biệt là kiểm soát tế bào sinh trưởng và phân chia) bị rối loạn.[35][36]
Sự thay đổi về gen sinh ung thư có thể là di truyền từ cha mẹ sang con cái, cũng có thể là mới
phát sinh trong cuộc đời của một cá nhân, xuất phát từ các lỗi xảy ra trong quá trình phân chia
tế bào hoặc từ sự tổn hại ADN gây ra bởi những phơi nhiễm nhất định ngoài môi trường. Những
yếu tố phơi nhiễm gây ung thư có rất nhiều, bao gồm các yếu tố vật chất (như hóa chất trong

Ung thư của mỗi người hình thành từ sự tổng hợp của những thay đổi trên gen. Khi ung
thư tiến triển, các biến đổi sẽ tiếp tục xảy ra thêm. Thậm chí trong một khối u, các tế bào khác
nhau lại có những thay đổi về gen khác nhau.
Rất nhiều các nghiên cứu khoa học đã chỉ ra tế bài ung thư có bề mặt là tổ hợp các đám mây
điện tử electron, do được hình thành từ các gốc tự do gây nên sự dịch chuyển điện tích của bộ
gen dẫn đến sự sai lệch, tạo nên các hiện tượng tĩnh điện của bề mặt tế bào ung
thư[36][37][39][40]
Thông thường, các tế bào ung thư có nhiều sự thay đổi về gen (ví dụ như đột biến DNA) hơn
các tế bào bình thường. Những thay đổi này và tác động này là sự phát triển của bệnh ung
thư.[30][31][32][33][34][35]

Các thay đổi về gene hình thành ung thư thường ảnh hưởng lên ba loại gen chính: tiền - gene
sinh ung (proto - oncogene), gen ức chế khối u (tumor suppressor gene), và gen sửa chữa ADN.
- Tiền - gene sinh ung có liên quan tới sự phát triển và phân chia của tế bào bình thường. Tuy
nhiên, bằng một cách nào đó, gene này bị biến đổi hoặc hoạt động quá mức bình thường, nó có

này có xu hướng xuất hiện thêm nhiều đột biến tại các gen khác, cuối cùng ung thư hóa là kết
quả của những đột biến đã xảy ra.
Sự phát triển của sinh học phân tử ngày nay đã giúp ích rất nhiều cho các nhà khoa học khám
phá ra nguyên nhân gây ra ung thư, đó chính là đột biến. Những đột biến nhất định xuất hiện
trong rất nhiều loại ung thư, do đó, ung thư đôi khi còn được phân loại dựa trên những sự biến
đổi về gene đã gây ra chúng, chứ không chỉ phân loại dựa trên nơi khởi phát ung thư hay dựa
trên loại tế bào ung thư , Sự khác biệt giữa tế bào ung thư và tế bào bình thường [33][34][35]
Bất kỳ bệnh ung thư nào cũng đều có sự xuất hiện của các tế bào bất thường, vốn là những
tế bào bình thường của cơ thể bị nỗi ADN khi nhân đôi không sao chép được ADN, tế bào phân
chia lỗi gien P53, gien này khi nhân lên mất kiểm soát, xâm lấn các mô xung quanh và có thể di
căn đi xa.
Ung thư có khả năng xâm lấn ra xung quanh, và đặc biệt, hình thành khối u mới ở một vị
trí cách biệt hoàn toàn so với vị trí của khối u ban đầu, gọi là ung thư di căn. Quá trình tế bào
ung thư di chuyển tới vị trí khác để hình thành khối u mới gọi là di căn.
Tên của ung thư di căn cũng như loại tế bào ung thư di căn hoàn toàn giống với ung thư
ban đầu. Ví dụ, ung thư vú tạo các khối u di căn tại phổi, sẽ có tên gọi là ung thư vú di căn,
không phải ung thư phổi.
Về mặt giải phẫu bệnh, các tế bào ung thư di căn thường giống như tế bào ung thư ở
khối u ban đầu. Hơn nữa, ở cấp độ phân tử, tế bào ung thư di căn và tế bào ung thư ở khối u
ban đầu cũng giống nhau, chẳng hạn như có cùng sự biến đổi trên nhiễm sắc thể. Quá trình
điều trị có thể giúp kéo dài sự sống cho những bệnh nhân bị ung thư di căn. Thông thường mục

Không phải cứ có sự thay đổi của mô tế bào nghĩa là đã bị ung thư, mặc dù có những thay đổi

những thay đổi của mô tế bào không phải là ung thư, nhưng cần được theo dõi trong một số
trường hợp:

nếu độ bất thường càng lớn, thì nguy cơ tiến triển thành ung thư càng cao. Một số loại loạn sản
cần được theo dõi và điều trị, ví dụ: một nốt ruồi bất thường trên da (loạn sản hắc tố) có thể
tiến triển thành ung thư hắc tố (dù phần lớn trường hợp sẽ không xảy ra như vậy).
- Trường hợp nghiêm trọng hơn nữa là carcinoma tại chỗ (carcinoma in situ). Dù đôi lúc nó
được gọi là ung thư, nhưng về bản chất nó không phải ung thư, bởi những tế bào bất thường
không xâm lấn quá khỏi mô khởi đầu, nghĩa là nó không xâm lấn ra xung quanh như các tế bào

Electrophoretic Mobility
Cells in suspension in an electrolyte solution like saline normally have a negative surface charge
and can therefore be induced to flow along an electrochemical gradient.
Cell electrophoretic mobility – the ratio of cell velocity to electric field strength, in cm^2 V^(-1)
s^(-1) – depends on the cell cycle stage, peaking around mitosis. So you might expect all rapidly
dividing cells, including tumor cells, to have high electrophoretic mobility more of the time.[1][4]
In samples from healthy patients and patients with chronic lymphocytic leukemia, n = 19,
electrophoretic mobility did not significantly differ between cancer and normal lymphocytes.[2]
In a study of blood cells taken from 700 patients (some healthy, some with various diseases
including both blood cancers and solid tumors), all cancers significantly _reduced _the
electrophoretic mobility of red blood cells, indicating that the RBCs became less negatively
charged. Benign tumors had no effect.[3]
Leukaemic mouse lymphocytes in saline had significantly lower electrophoretic mobility than
normal mouse lymphocytes.[5]
In a study of multiple human tumor samples, epithelial tumor cells did not have higher
electrophoretic mobility than normal cells, but connective tissue tumor cells (sarcomas and
myomas) did.[6]
Another study on different cell lines found that an epithelial cancer cell line (HeLa) was not
significantly different in electrophoretic mobility than normal cells, but that a carcinoma cell
line (Ehrlich ascites tumor) had significantly higher electrophoretic mobility.[7]
At physiologic pH, breast cancer cells have higher electrophoretic mobility (and thus more
negative surface charge) than healthy fibroblasts.[13]
Hamster kidney and liver tumor cells, which are carcinomas, have significantly higher
electrophoretic mobility than their healthy counterparts. The MCIM sarcoma becomes more
electrophoretically mobile as it gets closer to metastasis.[14]
Solid tumor cells are also less adhesive than healthy cells from the same tissue type; this is what
you would expect if they were more negatively charged and thus repelled each other
electrostatically more. Blood cells, healthy or cancerous, are already not very adhesive, and
accordingly they are more electrostatically mobile.[14]

In this report we present a theoretical study (Density Functional Approximation), focusing on
the stability and the free radical scavenger capacity of metal-fullerenes. We also denominate
free radical scavenger capacity as antioxidant or antiradical. Two different fullerenes have been
considered for this investigation: C60 and C82. Three transition metals were included to form
the metal-fullerenes: copper, silver and gold (one atom and a cluster formed with four atoms).
We analyzed the free radical scavenger capacity considering two mechanisms: electron transfer
(ET) and radical adduct formation (RAF). From this report, it is evident that the presence of
metals considerably improves the antiradical capacity of fullerenes. This may be useful for
future applications of these systems.
As the cancer cells are known to express more glucose receptors, Otake et al. exploited this fact
and synthesized group of C60-glucose conjugates which also proved to be more soluble. These
conjugates demonstrated selective phototoxicity compared to fibroblast cells thus suggesting
the significance of targeting glucose receptors.103 The PDT effect in vivo was investigated in
human-melanoma (COLO679)-xenograft bearing mice by injecting C60-(Glc)1 (0.1 or 0.2
mg/tumor) intratumorally followed by irradiated with 10 J/cm2 UVA 1ight. The drug-light
interval was 4 h. Tumor growth was suppressed better with the higher dose than the lower
dose.

2.4. Molecular Mechanism of Tumor Encaging: MMP Inactivation and Collagen Stabilization
As described in Figure 2, MMP deactivation and collagen stabilization contributed to tumor
encaging by Gd@C82(OH)22. On the one hand, Gd@C82(OH)22 inhibited the matrix-degrading
function of MMPs and thus prevented the proteolytic degradation of extracellular matrix in
tumor progression [42]. On the other hand, Gd@C82(OH)22 acted as a bridge to facilitate the
formation of collagen oligomers or microfibrils, inducing the enhanced stability and rigidity of
collagen layers.

3.2. Failure of Broad-Spectrum MMP Inhibitors: The Dilemma of Classical Drug Design
Preventing ECM degradation by MMPs is considered as a new route to fight cancer. In the past
two decades, many pharmaceutical companies have made great efforts to develop many MMP
inhibitors [60]. Unexpectedly, these MMP inhibitors were disappointing in clinical trials, either
with no efficacy or with severe adverse reactions [61]. The key reason for the failure lies in the
lack of selectivity towards the specific MMPs by the designed MMP inhibitors [62]. In fact, the
substrates of these MMPs are much more than extracellular matrix and different MMP
isoforms play diversified roles in different stages of the tumor [63]. By proteolysis, some of the
MMP isoforms can activate growth factors, for instance, angiostatin that inhibits tumor
angiogenesis and metastasis [64]. The broad-spectrum MMP inhibitors target all MMPs, even
those tumor-suppressing MMP members. This tribulation calls for a new generation of MMP
inhibitors with enhanced selectivity and specificity. However, chemically, it is challenging to
design the highly specific MMP inhibitors due to high similarities in MMPs’ active sites. In fact,
low selectivity is very common to small-molecular drugs which are traditionally designed
according to the key and lock principle [65]. The key and lock principle, firstly put forward based
on the model for enzyme-substrate interaction, mainly focuses on the active sites of enzymes.
The specific complementary geometric shapes are suggested to be possessed by enzyme and
substrate. Like a key into a lock, only the correct size and shape of the substrate (the key) could
fit into the active site (the keyhole) of the enzyme (the lock). Following this, the key and lock
principle extends its application to drug design. The designed small molecule drugs, with the
correct geometric shapes and chemical properties, occupy the active sites and impact the
function of biomolecules and ultimately disturb the associated biological process [66]. Despite
the success in conducting the development of some molecule-targeted drugs, this canonical
principle also encounters the dilemma where off-target occurred or low selectivity is very
common. By strong chemical bonds, the interaction between small-molecular drugs and the
active sites, in most instances, is compounded by the presence of high similarities in active sites
possessed by a family of proteins and generated indiscriminate inhibitions. Therefore, a new
strategy of drug design is urgently expected.

Third, the structure of nanomaterials is a key determinant factor in the biological effects of
nanomaterials. Previously, the authors compared the toxicity of Gd@C82(OH)22 and other
carbon nanomaterials to illustrate the role of the structure in determining biological effects. For
example, after respiratory exposure to the single-wall carbon nanotubes (SWCNTs) for 72 h, the
spontaneously hypertensive rats developed pulmonary inflammation and peripheral vascular
thrombosis [76]. Carbon nanotubes were observed to destroy the membranes of human gut
bacteria [77]. Moreover, the cytotoxicity of carbon nanomaterials with different structures was
compared, including single-wall nanotubes (SWNTs), multi-wall nanotubes (MWNTs) and
fullerene (C60) on alveolar macrophages, and C60 was found to induce the smallest cytotoxicity
[78]. Later, the biological effects of fullerene derivatives Gd@C82(OH)22 and C60(OH)20 were
compared, especially the anti-tumor activity. Resultantly, C60(OH)20 was found to possess an
attenuated anti-tumor activity compared with Gd@C82(OH)22, which was finally tracked to the
differences in the number and distribution pattern of hydroxyl groups on the carbon cage of
these two different fullerene derivatives [79]. A smaller number of hydrogen bonds formation
happened when C60(OH)22 interacted with collagen peptide triplex. Compared with C60(OH)22,
Gd@C82(OH)22 had a smaller percentage of carbon atoms functionalized by hydroxyl groups
and exposed more of the carbon cage surface, being more hydrophobic. The negatively-charged
fullerenol cage facilitated the electrostatic interaction with collagen. The stronger hydrophobic
and electrostatic interaction facilitated the absorption of Gd@C82(OH)22 on collagen
molecules, allowing a more favorable formation of hydrogen bonds with collagen molecules.
Due to the flexibility and diversity of interacting with certain molecules, nanomedicine, such as
Gd@C82(OH)22, seems to be a transformable and dynamic key that could match several locks
depending on the surface characteristics of the target biomolecules. In detail, the canonical key
and lock principle requires the existence of the complementary geometric shapes between the
molecular drug (the key) and the target biomolecules (the lock) to fit into the active site (the
keyhole). If the molecular drug cannot match the active site of the target biomolecules, off-
target happens and no drug efficacy generates. In contrast, designed nanomedicine could
intelligently detect the surface characteristics and selectively interact with the unique surface
area of target biomolecules, thus enhancing the target specificity. Depending on the surface
characteristics of the target biomolecules, nanomedicine, just like a transformable and dynamic

suggesting the target-tailoring ability of nanomedicine. Figure 4 demonstrates a brief history of
MMP-based research from the discovery of the first MMP in 1962 [80], the consecutive
identifications of MMP family members and their biological functions [81], the development of
MMP inhibitors to novel MMP-inhibiting nanomedicine Gd@C82(OH)22.

A brief history of MMP-based discovery: From MMP inhibitors to Gd@C82(OH)22. After the
discovery of the first MMP in 1962 [80], the intercellular regulation of MMP family was also
explored, particularly in tumor cells [56,81]. Subsequently, the development of MMP inhibitors
was initiated to achieve tumor confinement. However, due to their low selectivity and severe
side effects, these MMP inhibitors failed in clinical trials and drug development was abandoned.
The emergence of Gd@C82(OH)22 may bring the light for the development of selective MMP
inhibitors owing to the unique physicochemical characteristics of nanomaterials [42,50]. The
