CULTURE MEDIA
ial media are used for cultivation of bacteria fren clinical sainples.
6 like carbohydrates, proteins, electrolytes && water which pre
vi nuitrle
esseritin! fir the growth & multiplication of bacteria.
CLASSIFICATION
(Culture media can he classified in two ways!
1) Based on the consistency. 08
.iquid
+ Semi solid media (01.2 ~
+ Solid media (2%)
2) Based of their use, ms
- Simple media eg. Peplone water, Nutrient broth, Nutrient ager-
Enriched media eg. Blood agar, Chocolate agar, [,oefiler’s serum slope,
Dorset egg medina, Milk agar, Glucose broth, Brain heart Infusion broth,
Enrichment medin eg. Selenite F broth, Tetrathionute bros, Alkuline
peplone water. Bile broth,
Selective media eg, Potassium teDurite agar, Wilson & Blair medium,
Lowenstein Jensen mediutti. TIiesulphate citrate Bile sucrose agar (TCI
Sulmonelia Shigelli agar. Deoxy cholate citrate agar,
Differential media eg. MacConkey's azar, Xylose Lywine Decarboxs lase
medium (XLD), Eosin Melliyfeng Blue agar.
Transport meillu eg. Stuart's transport medium, Amie's trunsport medium,
Cary Blair medium, Venkalneaman Ramakrishnan medium.
Binchetnieal media eg. Triple sugor iron agnr, Simman’s eitrate medium,
Christensen’s Urcase medium, Sugar fermentation medium,
Methy] red — Voges Provkauer broth,
Anaerobic media — Robertson's Cooked ineal medium,
Thioglycolline broth.

PRINCIPLE: This medium determines the ability of an organism to utilize citrate as
sole source of carbon.
KEY INGREDIENTS: Sodium citrate - Source of carbon.
Ammonium dihydrogen phosphate- source of nitrogen

Green colour Blue colour
RESULT: Interpret after incubation for 24 — 48hours at 35 -37°C
Positive test: Growth with or without intense blue colour.
eg: Klebsiella pneumoniae, Citrobacter species.
Negative test: No growth and no change in colour,
eg: E. coli, Shigella species,

ROBERTSON’S COOKED MEAT MEDIUM (CM)
TYPE: Anaerobic medium.
KKEY INGREDIENT: Minced meat, Peptone.
STERILIZATION: Autoclaving [Fresh medium preferred]
PRINCIPLE: Minced meat provides amino acids, nutrients and glutathione, a reducing,
substance promotes the growth of anaerobes.
USES: 1) Cultivation of facultative anaerobes, obligate anaerobes
especially pathogenic Clostridia.
2) To determine proteolytic (blackening of meat and foul odour) and
saccharolytic (pink color of the meat and sour odour) activity of
bacteria.
3) Maintenance of clostridia.
4) Recovery medium for spores.
