In order to correctly achieve the antibiogram, we have to know if our bacteria is

gram-positive or gram-negative, knowing this, choose the Macconkey or Mueller


Hinton culture medium, in this case Macconkey was chosen because it was known
that the microorganism was gram-negative.

Subsequently, the sensitivity discs were made with the filter paper and then with a
pencil the initial of each antibiotic was put on them, A for ampicillin, T for
trimethoprim and B for blank, the latter is not an antibiotic, but it is important to
have a blank to corroborate results.

Having this, we proceed to make divisions in the Petri dish to identify where the
target and each antibiotic will go. In the middle of each division the first letter of the
target and the medicine is placed.

Once this part was concluded, we concentrated on taking out the antibiotics, it was
known that ampicillin was only a drop and trimethoprim was .1 g in 2ml of distilled
water.

Next, you take the sensidisks to put a drop of the antibiotic on them, the one with
the letter A for ampicillin and the one with a T for trimethoprim, and you have to let
it rest.

While the sensididiscs are absorbing the antibiotic, the striatum is made. You have
to turn on the burners and sterilize the area, open the Petri dish where you have
the desired microorganism, now with a bacteriological loop take the sample and
make a massive streak in the agar, finishing this with some dissecting forceps,
grab the sensidisc with the letter B and place it in its corresponding place.
We take the other sensidisks and repeat the same procedure as with the white
one.

Finally, the Petri dish is closed and sealed, the burners are turned off and it is taken to
incubate and the results can be seen later.
