LAB 3: GRAM STAINING OF YEAST CELLS.

Carry out the following technique.

1. Using a sterile inoculating loop, prepare a smear on a clean dry slide.


2. Air dry and heat fix the smear.
3. Dip the smear in crystal violet for 1 minute.
4. Gently wash the slide with tap water and drain the slide again a paper towel (Do
not blot).
5. Dip the smear in Gram’s iodine for 1 minute.
6. Gently wash the slide with tap water and drain carefully.
7. Wash with 95% alcohol for 30 seconds.
8. Wash with tap water immediately after the 30 seconds and drain.
9. Dip the smear in Safranin for 30 seconds.
10. Wash with tap water, drain, blot and air dry.
11. Examine under oil immersion.
12. Make well labeled diagrams of the observed cells and answer the questions given
in the answer sheet.
