Areas of Research Interest: Extinction as a Therapeutic Tool for Post-Traumatic Stress Disorder (PTSD) and for various Mood Disorders. We use Early Maternal Separation Stress as an Animal Model to Unravel the Neural circuits and the cellular mechanisms underpinning passive (extinction) and active (emotion regulation) strategies to regulate adult behavioral changes. We use Pavlovian Classical Fear conditioning as an animal model. Above figure represents the effect of Maternal Separation Stress on the cued fear conditioning and subsequently on the neural activities of hippocampus, medial prefrontal cortex and amygdala during presentation of conditioned stimulus. Left figure is from normal control and right from Maternal Separation stressed group. Recently, vasocerebral accidents and their therapy have become a central focus of research. Significant efforts have been made to develop animal models that will mimic the physiological and pathophysiological consequences associated with stroke in humans. I am interested in brain plasticity mechanisms and functional recovery following ischemic injury. We infuse Endothelin-1 to induce vasoconstriction at the stem of the MCA leading to a focal lesion limited to the cortical area of interest, without performing a craniectomy, leaving the tissues in place to more closely replicate the physiological state of the post brain infarct. Developing a focal cerebral ischaemic

model in rat would enable a method for the continuous observation of neuron specific electrical properties that will register fluctuations of cortical excitability and determine if such changes vary according to the location or type of lesion. By establishing the degree of inter-subject variability regarding the electrical state of the post-ischemic cortex, subject-specific treatment protocols can be tailored to reverse damaging excitability changes as they occur. Because previous studies limit electrophysiological characterization to discrete time points after the onset of ischemia, the temporal dynamics of local electrophysiological properties induced by infarction remain uncertain. With this, we would be able to demonstrate a method to monitor the continuous electrical signals of cortical neurons subject to ischemia and to document the dynamic electrophysiological responses of neuron clusters in the infarct zone and the penumbra by unit neural activity before, during and after infarction and also to establish the possible role of fear related stimuli in reestablishing the neural connections related to attention circuits. Understanding the continuous electrophysiological profile of the post-ischemic cortex would also provide new insights in to cortical plasticity during recovery or help to elucidate new strategies for acute and chronic clinical intervention based on known neural response patterns.
