ENZYME TRANSESTERIFICATION :
BURKHOLDERIA CEPACIA LIPASE
Ethanolysis with immobilized lipase in free
solvent system
Efficient in production of biodiesel of vast
range of vegetable oil
Burkholderia cepacia lipase can be reused
due to the immobilization
Slow reaction
OBJECTIVES
Ethanolysis of Jatropha curcas L. oil by
using enzyme catalyst which is
Burkholderia cepacia

To determine the biodiesel production
through measurement of viscosity
METHODOLOGY
Analysis
Biodiesel a.k.a fatty acid ethyl ester conversion
measured using Brookfield Viscometer LVDV-E

Material
5 batch reactors with Jatropha curcas L. oil, ethanol
and enzyme and repeated one more time

Data
Taken in 4 different parameter which are molar ratio,
enzyme content (wt% of oil), reaction time and water
content (wt% of oil)
PARAMETERS
Molar ratio Enzyme (wt% of oil) Reaction time (hr) Water content(wt% of oil)
Temperature 40 C

TRANSESTERIFICATION MECHANISM
Ethanol was deprotonated by the enzyme catalyst
to become nucleophile.
The breakage of triglyceride accompanied by the
exchange of acyl acceptor, ethanol R group to
form 3 ethyl ester and 1 glycerol

The existence of heat as well as enzyme catalyst
only increase the reaction rate and does not
change form in the end of the process

CONCLUSION
Immobilization of lipase based on the experiment
results shows that the optimum parameters are
1:6 molar ratio, 60 hours, 50% enzyme
percentage and 10% water content
The best viscosity value obtained was 10.68 cP.
The calcium alginate beads were appear as
appropriate support for the ester conversion.
