Monochromatic staining
• Principle:
• Staining with single dye solution contain salts.
• Auxochrome and chromophore.
• Auxochrome responsible for the dissociation of
the dye and a chromophore responsible for the
colour of the dye.
• In basic dye chromophore is positively charge
and the cell have negative charge.
• Ex. Methylene blue, crystal violet etc.
