Highlight of Enzyme Analysis

• Representative sample and homogeneity of the feed is

important for the result
• For extraction, take a representative sample. If facing high
CV extract at least 50 g or more
• Treat the enzyme standard vials the exactly the same way
as the sample vials (way of pipetting, incubation, etc.)
• All enzyme should be dissolved from its product form and
released into the liquid (buffer). Enzyme show the tendency
to bind to insoluble feed components
Highlight of Enzyme Analysis

• Enzyme inhibitors and activators should not be extracted,

and if extracted, inactivated
• Extraction solution should be ideally cheap and easy to
prepare. It should always contain a detergent
Protease Analysis

• Use eppendorf tube instead of test tube

• For feed samples, use 3 kinds of glycine buffer.
• For premix samples, use buffer with 1.5 % EDTA.
Xylanase Analysis

• Use blank : Corn meal, blank feed or feed extract

• Incubation time for feed : 180 minutes + 45 minutes at RT
For premix : 90 minutes + 45 minutes at RT
Rapid Lab

• It is a equipment to detect the 3D’s structure of Hi Phos

• The analysis used cassette with barcode
• The feed sample is extracted with water and 0.02 % Tween for 20
minutes. 50 µL feed extract is mixed with 950 µL MICT buffer
(part of the kit).
100 µL of the mix is pipetted and is poured to the conjugate tube.
Mix by pipetting 4 x. The whole mix is taken out and is loaded to
the cassette.
• Leave for 30 minutes (Rapid read) or leave until dry for 4 hours
(Standard read)
Obstacles

• Clean combitips and eppendorf tube racks at RL Balaraja

are limited in number

• Spectrophotometer microplate
