CSI 251 CLINICAL MICROBIOLOGY III

WORKFLOW OF
BACTERIOLOGY
IDENTIFICATION PROCESS

LEARNING OUTCOMES
At the end of this lecture, student should
be able to:
1. Know the workflow in the Bacteriology
laboratory.
2. Classify the sample received with
appropriate testing.
3. Identify the tests that should be
carried out according to the first day,
second day and third day.
1. Sample types
The type of sample that could be send to the
laboratory are:
Stool
Urine
Blood
Pus
CSF
Other body fluids
2. Examined the sample quality
Medical Laboratory Technologist (MLT)
MUST examine the sample quality
according to the following criteria:
Examine sample received and request
form
Amount of sample
TIME
Container used
WORKFLOW
e.g. Stool sample

First day
Culture the specimen on Blood agar and
MacConkey
Incubate for overnight at 37°C (12-24 hours)
Second day
Examine the culture plate
Perform gram stain
Read the culture morphology on blood agar
Second day (continued)
Lactose fermenter or non lactose fermenter
Perform biochemical testing e.g. IMViC ,TSI,
urea, motility and sugar fermentation.
Perform catalase, oxidase, coagulase and
Dnase testing.
Second day (continued)
Perform 3D culture on Muller Hinton agar
Place the antibiotic disc on the culture
Incubate the biochemical testing for
overnight at 37oC
If no grow observed from first day, re-
incubate the culture.
Third day
Identify the organisms from
biochemical testing
Read the AST zone inhibition
State the organism identifed and
antibiotic suggestion.
Release the result to the ward
