How does splicing work?
• A bacterial cell contains :
1) Bacterial (Chromosomal) DNA
2) Plasmid DNA
What is a Plasmid?

• is a small, extrachromosomal DNA molecule within a cell that is


physically separated from chromosomal DNA and can replicate
independently. They are most commonly found as small circular,
double-stranded DNA molecules in bacteria)
What are restriction enzymes:
• Restriction enzymes are DNA-cutting enzymes. Each enzyme
recognizes one or a few target sequences and cuts DNA at or near
those sequences.
What are sticky ends?
• sticky' end is produced when the restriction enzyme cuts at one end
of the sequence, between two bases on the same strand, then cuts
on the opposite end of the complementary strand. This will produce
two ends of DNA that will have some nucleotides without any
complementary bases.
What is DNA ligase?
• DNA ligase is an enzyme which can connect two strands of DNA
together
What is Recombinant DNA ?
• Recombinant DNA molecules are DNA molecules formed by
laboratory methods of genetic recombination to bring together
genetic material from multiple sources.
