ALLOSTERIC

REGULATION
What is
Allosteric
Enzyme?
Allosteric enzymes are enzymes that have an
additional binding site for effector molecules other
than the active site. The binding brings about
conformational changes, thereby changing its
catalytic properties. The effector molecule can be an
inhibitor or activator.
What is
Allosteric
Regulation?
Allosteric Regulation refers to the process for
modulating the activity of a protein by the
binding of a ligand, to a site topographically
distinct from the site of the protein, in which the
activity characterizing the protein is carried out,
whether catalytic (in the case of enzymes) or
binding (in the case of receptors) in nature.
These enzymes, which include some of our key
metabolic regulators, are often given the name
of allosteric enzymes.
There are also allosteric activators. Some
allosteric activators bind to locations on an
enzyme other than the active site
How does Allosteric
Regulation Works?
2 Types of Allosteric Mechanism
Homotropic Regulation: Heterotropic Regulation:
Here, the substrate  When the substrate
molecule acts as an and effector are
effector also. It is
different. The effector
mostly enzyme
may activate or inhibit
activation and also
the enzyme, e.g.
called cooperativity,
e.g. binding of oxygen binding of CO2 to
to haemoglobin. haemoglobin.
In allosteric inhibition,
the binding of the
molecule to the allosteric
site causes a shape change
that reduces the affinity of
the enzyme for the
substrate.

In Allosteric Activation, it
increases the rate of
reaction.

Transcarbamoylase Carboxylase
(ATCase)
