Immunoassay
1- Competitive ELISA
2- Sandwich ELISA (also called direct ELISA)
Indirect ELISA -3
Direct Elisa
The simpliest
Ag (sample) directly
adsorbed on plate
Enzym conjugated
antibody + substrate
: Application
Screening antigen (immunoglobulin) in Fetal Calf serum
FCS was diluted in coating buffer & coated on plate -
washed the plate, add specific enzyme-conjugated calf-Ab
Washed the plate, add substrate -
Competitive ELISA

Elisa sandwich (capture
antigen)
Specific Ab adsorbed on plate to catch
Ag
Ag in the sample were added
2nd labelled Ab were added
Addition of substrate will produce
collor
OD depend on Ag concentration
Mo Ab were usually used for capture
& 2nd Ab
Capture & 2nd Ab (if it is MoAb)
should have different epitopes on the
same Ag (except particulate Ag which
have several identic epitopes may use
same MoAb)
Elisa sandwich for Ab
)Capture Ab(
Anti Ig Ab were adsorbed on
plate
Ab in the sample were catch
& indicator system were Ag-
labelled/2nd Ab
This assay were usually used
to determine isotype (exc. To
measure IgM, were used
anti IgM-Ab
Indirect ELISA
The protein antigen to be tested for is added to
each well of ELISA plate, where it is given
time to adhere to the plastic through charge
interactions
A solution
of non-reacting protein is added to
block any plastic surface in the well that
remains uncoated by the protein antigen
Indirect ELISA-Cont
Then the serum is added, which contains a
mixture of the serum antibodies, of unknown
concentration, some of which may bind
specifically to the test antigen that is coating the
well.

The standards concentrations is specified on
the x-axis and the reading of each standard is
specified on the y-axis and the standard curve
is drawn
Results-cont
Thisstandard curve is used to determine the
unknown concentration of each sample by
finding the opposite concentration to the
