• Describe radial immunodiffusion (RID), and explain how it is used to
quantify the concentration of antigens or antibodies
• Determine the principle behind the immunoelectrophoretic (IEP)
assay, including the roles of electrophoresis and immunodiffusion.
Also, illustrate five advantages and limitations of the IEP assay
compared to other immunoassays.
Immunodiffusion
Immune precipitates can form not only in solution but also in an agar
matrix.
When antigen and antibody diffuse toward one another in agar, or
when antibody is incorporated into the agar and antigen diffuses into
the antibody-containing matrix, a visible line of precipitation will
form.
Immuno-diffusion is a technique for the detection or measurement
of antibodies and antigens by their precipitation which involves
diffusion through a substance such as agar or gel agarose.
Simply, it denotes precipitation in gel.
Immunodiffusion
• It refers to any of the several techniques for obtaining a precipitate
between an antibody and its specific antigen.
• This can be achieved by:
b) Letting both antibody and antigen migrate through the gel from
separate wells such that they form an area of precipitation.
