 TITTLE:GRAM’S STAINING
 AIM: to differentiate between grams positive bacteria and grams
negative bacteria
 METHOD: grams staining technique
 MATERIALS: microscope, slide, crystal violent, lugol’s iodine,
acetone, neutral red, Bunsen burner, wire loop, staining rack, culture
plate, cotton wool, pipette,
 PRINCIPLE: the acidic component of a cell have affinity to basic
dye and the basic component of the cell have affinity to acidic dye.
SMEAR MAKING WITH SOLID
CULTURE
 A drop of normal saline was placed at the center of
a clean grease, free, slide,
 A wire loop was sterilized and allow to cool
 A colony of the sample was picked and emulsified
in the drop of normal saline evenly
 It was allowed to air dry
PROCEDURE

 The dried smear was pass over a Bunsen  It was decolourized with acetone briefly
burner flame three times
 It was rinsed with clean water
 It was allowed to cool
 It was counter stained with neutral red for 120sec
 It was placed on a staining rack  It was rinsed with clean water
 It was covered with crystal violent for 60sec  The back of the stained smear was clean with dry cotton
 It was rinsed with clean water wool

 It was covered with lugols iodine for 60sec  It was allowed to air dry
 It was examined microscopically using x100 objective
 It was rinsed with clean water

RESULT

 Gram’s positive cocci seen (blue or


violet).
CHALLENGES
 The following are the
 At the collection bench , I was unable to collect blood
from most of the infants effectively
 Most test I feel I can carry out, but I was not allowed to
do.
 Only few staff were ready to assist us in term of teaching
on bench
 I was told that most of the work are for scientist and not
technicians.
RECOMMENDATION

 I strongly recommend our school authority to


maintain the posting of our students to the
facilities because the hospital is well
equipped, there is orderliness of work,
experienced scientists and technicians and the
work load is much.
CONCLUSSION

 Despite the challenges we faced, I was able to conclude


my practical posting. And through the help of God, I was
able to learn a lot from the various department and I can
carry out some investigations in those various department.
I therefore wish to thank the unit staffs and their head,
HODs, admin, who made sure that we didn’t go back the
way we came.
