What is Vitamin D3?

Vitamin D3, also known as cholecalciferol. It is a fat-soluble vitamin that


plays a crucial role in maintaining bone health and supporting the
immune system.

It is produced in the skin upon


exposure to sunlight.

Molecular Formula of cholecalciferol is C27H44O


Health Benefits
Vitamin D3 has numerous health
benefits, including supporting
Bones strength regulating mood , and
enhancing immune function .

Its deficiency is linked to increased risk


of various health conditions .
Synthesis of vitamin D3
Vitamin D3 is synthesized in the skin upon UVB exposure.

The UVB exposure of provitamin D3 (7-dehydrocholesterol) in the


skin breaks the B-ring to form previtamin D3, which undergoes
thermally induced rearrangement to vitamin D3.

Vitamin D3 is transported to the liver where it is hydroxylated at C-25


by the enzyme 25-hydroxylase producing 25OHD3, which is the major
circulating form in vertebrates.

The 25OHD3 is hydroxylated a second time at C-1 in the kidneys to


the active metabolite 1,25(OH)2D3.
Sources of Vitamin D3
The primary source of Vitamin D3 is
sunlight. It can also be obtained from
certain foods such as fatty fish, egg yolks, and
fortified dairy products. Supplements are
recommended for individuals with limited sun
exposure.
Sunlight Exposure

Sunlight is the primary source of Vitamin D3.

However, it is important to practice sun safety to


minimize the risk of skin damage and skin cancer.

Balancing sun exposure for adequate Vitamin D3 synthesis and skin


protection is essential.
Vitamin D3 Deficiency
Vitamin D3 deficiency is associated
with an increased risk of osteoporosis, depression,
immune disorders, and
other health conditions.

It is important to recognize the signs and symptoms


of deficiency and take appropriate measures.

Rickets is a condition of softening of the bones


that occurs in growing children. It happens
when the bones can not take up enough
calcium and phosphorus to make hard,
healthy bone.
Metabolization
Vitamin D3 produced in the epidermis must be further metabolized to be active.
The first step, 25-hydroxylation, takes place primarily in the liver, although other tissues have this enzymatic activity as well.
there are several 25-hydroxylases.
25OHD is the major circulating form of vitamin D.
However, in order for vitamin D metabolites to achieve maximum biologic activity they must be further hydroxylated in the 1α position by the
enzyme CYP27B1; 1,25(OH)2D is the most potent metabolite of vitamin D and accounts for most of its biologic actions.
The 1α hydroxylation occurs primarily in the kidney, although as for the 25-hydroxylase, other tissues have this enzyme.
Vitamin D and its metabolites, 25OHD and 1,25(OH)2D, can also be hydroxylated in the 24 position.
This may serve to activate the metabolite or analog as 1,25(OH)2D and 1,24(OH)2D have similar biologic potency, and
1,24,25(OH)3D has activity approximately 1/10 that of 1,25(OH)2D.
However, 24-hydroxylation of metabolites with an existing 25OH group leads to further catabolism.
Current Research
Vitamin D3 plays a crucial role in the body, primarily
in maintaining bone health by aiding in the absorption
of calcium and phosphorus.
It also supports immune system function, regulates
cell growth, and contributes to overall cardiovascular
health.
Research suggests that adequate vitamin D levels may
reduce the risk of certain chronic diseases such as
cancer,diabetes,and heart disease. and promote overall
well-being.
However, excessive intake can have adverse effects,
emphasizing the importance of maintaining a balance.
