• Vitamin-B12 is water soluble,
heat stable and red in colour.
• Four Pyrrole rings are co-ordinated
with a cobalt atom ,is called a
corrin ring.
• The 5th valency of cobalt is covalently
linked to a substituted benzimidazole
ring. This is then called cobalamin.
• The 6th valency of cobalt is satisfied by
any of the following group – cyanide (
, Hydroxyl , Adenosyl , or methyl.
(2) Isomerisation of Methylmalonyl CoA to Succinyl
CoA :-

• In B12 deficiency , Methylmalonyl CoA accumulates and methyl


malonic aciduria occurs.
Recommended Dietary Allowance
•(RDA)
For Adults : 1-2 microgram per day
• For Children : 0.5-1.5 microgram per day
• For females during pregnancy and lactation : 3 microgram per day

Dietary Sources
• In Vegetarian diet : Curd , Milk
• Animal origin : Fish , Pork , Chicken
Deficiency
Symptoms
• Most Important disease associated with Vitamin-B12 deficiency is pernicious anemia.
• Characteristics :-
1. Low haemoglobin levels
2. Decrease number of erythrocytes
3. Neuronal degeneration and demyelination of nervous system
• Causes :-
1. Autoimmune destruction of gastric parietal cells.
2. Gastrectomy : individual becomes Intrinsic factor deficient.
3. Dietary deficiency of B12 seen among the strict vegetarian of low socioeconomic group.
• Treatment :-
1. Vitamin B12 is administered in therapeutic doses (100-1000) microgram
intramuscularly.
