ANALGESIC AND ANTIPYRETIC ACTIVITY OF THE LEAVES

OF MUKIA MADERASPATANA

INTRODUCTION:

Tamil Siddha Vaidyans widely using the juice obtained


from the leaves of mukia maderaspatana in treating fever, headache
and muscular pain. Based upon the information available mukia
maderaspatana was selected for analgesic and antipyretic activity.

OBJECTIVES:

The objectives of the present study is based on the following:

• Authentication of plant
• Pharmacognostic evaluation
• Successive solvent extraction
• Pharmacological screening of
 Analgesic activity
 Antipyretic activity

MATERIALS AND METHODS:

The plant was collected from the villages in Sivagangai


district of Tamilnadu and authentified by Botanical survey of India,
Coimbator the solvents used for extraction purchased are purchased
from SD fine chemicals the standard drug celecoxib procured from
unichem lab, Goa and Paracetemaol from Pharmafabricon, Madhurai.
The raw data of the present pharmacological study were
subjected to simple statistical analysis to draw meaningful
interpretation and conclusions for the + SEM and student t-tests were
calculated to examine statistical significance.

SUCCESSIVE SOLVENT EXTRACTION:

About 200 gms of powdered leaf of Mukia maderaspatana was


evenly packed in the soxhelet apparatus and was first extracted with
petroleum ether (60-800 c). This extraction was carried out for 48
hours. Then the marc was subjected to successive extraction with
chloroform and methanol respect. The chloroform and methanol were
taken up for the Pharmacological screening.

PHARMACOLOGICAL SCREENING:

The leaf extract of Mukia mederaspatana was evaluated for the


locomotor activity in albino mice. There is no change in locomotor
activity especially in CNS initiate the screening of non opioidal type
of analgesic activity.

WRITHING IN ALBINO MICE

Over night fasted albino mice (20-30 gms) of either sex were
selected and water ad libitum. They were divided into 4 groups each
consisting of six animals. The first group served as a solvent control
which received 1ml/100gms of water. The second group was treated
with celecoxib [9mg/kg]. P.O. The other groups were treated with
chloroform and methanol extracts of the leaf at a dose level of
25mg/kg.P.O.after 30 min of drug treatment 5% of acetic acid was
injected I.P. The number of wriths calculated by using the formula
100t1/t2*100.

INDUCED PYREXIA IN ALBINO RATS

Experimental pyrexia was induced in rats by administering


20% suspension of Brewer’s yeast 1 ml/ 100 gm. Sc .This experiment
was carried in four groups of 6 rats each (180 – 200 gms).Group 1
served as a solvent control and group 2 received Paracetomol as a
positive control. Remaining groups received chloroform and
methanol extracts [25mg/kg].P.O. Rectal temperature was measured
and total reduction in temperature was calculated.

RESULT AND DISCUSSION:

1. ANALGESIC ACTIVITY:
Chloroform and methanol extracts showed percentage
protection of 63.09% and 68.24% respectively possess almost
equipotent analgesic activity when compared with celecoxib
[76.18%].

2. ANTIPYRETIC ACTIVITY:
Chloroform and methanol extracts showed a total reduction in
temperature of 1.150c and 1.2750c possess almost equipotent
antipyretic activity as that of Paracetamol [10c].
