How to do the Microscopy Calibration with PAX-it

1. Add the Magnification into the software. Go to Database, click Utilities and Select Setup Database tab.

2. Click Hard Drive button

3. Select the field Field #3: Magnification, click the Configure Lookup Table button, and then click Add button and type in the Microscope Magnification you want to calibrate. Finally click OK.

4. Click the PaxCam button and choose the right camera you are going to use. Put the standard scalar on the sample stage and focus it to the best condition. And then click Capture button to take a picture. You could also Check Preview and Make sure you are choosing the right camera you are using.

5. Choose the right Magnification in the Magnification row and Type in the name you want to name your file. Finally Click OK.

6. Click the Calibrate button in the upper left corner. The full scale is 1000 um, so one unit is 10 um. Using the mouse to adjust the lines. In order to make more accurate, try to use the full scale under the microscopy. And then choose the measument unit and type in the number of units used duing calibration. Finally click OK.

7. The sentence Would you like to copy the calibration to all writeable drivers? will pop out. Click No since we save in the hard drive.

8. After each magnification calibration, need to double check. Close the image and then go to the back to last folder button. Double click the image to open it.

9. Click Draw Horizontal icon. Draw a line and check whether the measurement is correct.

10. Click the close in the upper right corner to close the image.
