2. What is the use of UV light ? Why that bacteria must be observed under UV light? The pGREEN plasmid contains a mutant version of GFP linked to another gene called beta-galactosidase. This combination of genes was chosen because the protein produced from this combination turns bacteria yellow-green, even in normal light. If we expose the colonies to a UV light, they also fluoresce. The plasmid also contains the antibiotic resistance gene to allow growth in the presence of ampicillin. http://www.apsnet.org/EDCENTER/K12/TEACHERSGUIDE/PLANTBIOTECHNOLOGY/Pages/Activity4.aspx-
3. What to do if water sample has some background color? How to make the dilution? If a water sample has some background color compare inoculated colilert sample to control blank of the same water sample. Use sterile water, not buffered water for making dilutions. Colilert is already buffered. Always add Colilert to the proper volume of diluted sample after making dilutions.
