COLOUR (TONALITY AND INTENSITY) IN WINE


DEFINITION AND SCOPE OF THE TEST
The chromatic features of a wine are defined by two analytical parameters: tonality and intensity Through
appropriate mathematical formulas, calculating the absorbance of the wine at 420nm, 520nm and 620nm, is
possible to determine intensity and tonality value in wine. These two elements, which represent the
chromatic profile, change with the evolution of wine and they show the quality and the health of wine.
TEST PRINCIPLE
Readings of absorbance at 420 nm, 520 nm and 620 nm are made on wine (sample as is) pre-vialed into the
cuvettes. The instrument automatically calculates the tonality (according to the formula Abs 420 nm/Abs 520)
and the intensity (according to the formula Abs 420 nm + Abs 520 nm + Abs 620 nm). The innovative CDR
method makes the standard procedure easier and faster. The special photometric technology together with
the provided cuvettes, enable to reach higher optical densities compared to normal spectrophotometers
obviating the problems of sample dilution.

SAMPLE TREATMENT MEASURING RANGE - ACCURACY - LINEARITY
Wine: use the wine as is.
Must: simplified extraction method at pH 1 and pH 3,2. Modified Glories method.
Extraction operating technique:
1. Chop some grape in a mixer for 30 seconds.
2. Take two samples both of 6 gr. and put them in the special plastic disposable glasses.
3. Add to the first sample 6mL of pH 1 solution A.
4. Add to the second sample 6mL of pH 3,2 solution B.
5. Cover the glasses with the lid and wait for 1 hour. Shake the glasses twice.
6. Filter or centrifuge the suspensions.

For wine with an higher absorbance, the instrument indicates out of range; dilute sample.
In this case is possible dilute wine 1:1 with the special diluents and start again the analysis session. The
results of the intensity and of each reading (420, 520 and 620) should be multiply by 2. Tonality value should
not be multiplied since it is a ratio of readings.
SYSTEM STANDARDIZATION
The instrument is supplied pre-calibrated and ready for use.
Results are expressed in accordance with the reference method.
It is also possible to standardize the system using samples with known titration.

OPERATING TECHNIQUE
1. Fill a semi-micro cuvette with sample.
Wine absorbance changes with temperature. It is advisable to carry out the analysis on a sample
with constant temperature.
2. From the main menu select Colour and press ENTER. INSERT SEMI-MICRO cell1 position A
displays.
3. Insert the semi-micro cuvette containing the sample in the reading cell 1 (the cell is indicated by a
green light) in the position A and press ENTER.
4. At the end of the reading INSERT SEMI-MICRO cell 2 position A displays.
5. Insert the semi-micro cuvette containing sample in the reading cell 2 (the cell is indicated by a
green light) in the position A and press ENTER..
For sample with high absorbance values, PULL SEMI-MICRO AND PRESS ENTER displays and
after this operation INSERT SEMI-MICRO in position B displays; insert again the semi-micro
cuvette with sample in the reading cell indicated by the green light in position B.
For sample with higher absorbance values the above described reading is not enough and INSERT
MICRO displays. Pour the sample in a micro cuvette, put it in the reading cell indicated by the
green light and press ENTER.
6. At the end of the readings, the results of intensity and tonality and the three readings at 420nm,
520nm 620nm, will be automatically printed as absorbance referred to optical path 1 cm.
When the instrument verify reading cells, it is necessary remove cuvettes from cells.

COMPARATIVE TEST
Comparative tests, carried out by Italian ISVEA laboratory, have shown an excellent correlation on the entire
measure range with the reference method.
