2. MATERIALS AND METHODS

1. INTRODUCTION

6-Aminopenicillanic acid is the main precursor for semi-synthetic beta-lactamic


antibiotics production (amoxycillin, ampicillin, etc.).
This precursor could be obtained by means of chemical synthesis and also by
Penicillium spp. fermentation without precursor, but the current method applied at
industrial scale for 6-Aminopenicillanic acid production consists on the deacylation of
natural Penicillins.
This work studied the efficiency of enzymatic hydrolysis of Penicillin G to 6Aminopenicillanic acid under substrate inhibition for a bioreactor with mobile bed of
immobilized penicillin amidase
pH [7, 9], t [20 oC, 40 oC]
: (1; 1,5: 2) mm.

The enzymatic hydrolysis of Penicillin G to 6-Aminopenicillanic


acid with penicillin amidase immobilized in Eupergit C particles indicated
a optimum evolution of the process at the 8 pH-value and temperature
of 30oC, regardless of particless size.
The inhibitory effect of substrate could be diminished by cells
immobilization, the substrate consumption rate increasing with the
increase of biocatalyst particle size.
