Four Hfr strains of E. coli were mated with an F- strain to determine the order of genes on the E. coli chromosome using interrupted conjugation. Each Hfr strain transmitted genes to the F- strain in a unique order. A circular map of the E. coli chromosome needs to be constructed showing the proper order of genes, noting the time in minutes between adjacent genes transferred from each strain.

25 Feb 2010 Interrupted Conjugation practice problem: -------------------------------------------------------------------------------------------------------------------- Using the technique of interrupted mating, four different Hfr strains of E. coli were mated to an F- strain to determine the origin and the sequence of genes on the bacterial chromosome. Each of the Hfr strains transmitted its genes to the F- strain in a unique sequence. Construct a circular map for E. coli placing each gene in its proper order on a circular map and note the time in minutes between adjacent genes. NOTE: The experimentally useful genes differ among the strains, but the gene order on the E. coli chromosome does not change.
